Abstract
Description
Technical Field
[0001] The present invention belongs to the technical field of pharmaceutical chemistry, and in particular relates to a crystal form of a compound used as a kinase inhibitor, a preparation method and an application thereof. Background Art
[0002] The epidermal growth factor receptor belongs to the receptor tyrosine kinase (RTK) family, which includes EGFR / ERBB1, HER2 / ERBB2 / NEU, HER3 / ERBB3 and HER4 / ERBB4. The epidermal growth factor receptor activates its tyrosine kinase activity through homodimerization or heterodimerization, and then phosphorylates its substrate, thereby activating multiple downstream pathways related to it in the cell, such as the PI3K-AKT-mTOR pathway involved in cell survival and the RAS-RAF-MEK-ERK pathway involved in cell proliferation. Mutations or amplifications of the epidermal growth factor receptor will lead to the activation of the epidermal growth factor receptor kinase, leading to the occurrence of various human diseases, such as malignant tumors. For example, in patients with non-small cell lung cancer, about 10% of American patients have EGFR mutations, while the proportion of patients with EGFR mutations in Asian patients can reach nearly 50%. At the same time, the incidence of HER2 mutations in patients with non-small cell lung cancer is about 2-4%.
[0003] EGFR mutations mainly include deletions, insertions, and point mutations, among which exon 19 deletions and exon 21 L858R point mutations account for nearly 90% of EGFR mutations. For patients with tumors with these EGFR mutations, the EGFR-TKIs currently on the market include first-generation Iressa, Tarceva, and Conmena, second-generation afatinib and dacomitinib, and third-generation osimertinib. The other 10% of EGFR mutations mainly involve exons 18 and 20 of EGFR, and the insertion mutation of EGFR exon 20 accounts for about 9% of the entire EGFR mutation. For patients with tumors with HER2 mutations, the most common HER2 mutation is the insertion mutation of HER2 exon 20.
[0004] TAK-788 has a therapeutic effect on exon 20 insertion mutations of EGFR and HER2. The compound has been marketed in the United States. According to the reported clinical trial results, the objective response rate is 43%. Recently, it has been reported that DZD9008 is effective in treating advanced non-small cell lung cancer with EGFR or HER2 mutations. The objective response rate of EGFR exon 20ins is about 40%, which is not satisfactory.
[0005] Therefore, the development of new compounds with EGFR / HER2 kinase inhibitory activity and better pharmacodynamics and pharmacokinetic properties has become an important research project for the development of new anti-tumor drugs, and will ultimately be used in the treatment of human tumors and other diseases.
[0006] Patent application number (PCT / CN2023 / 081557) discloses a series of compounds used as EGFR / HER2 kinase inhibitors, and the series of compounds disclosed in this patent have very good EGFR / HER2 inhibitory activity. In this patent application, a compound with a structural formula as shown in formula (A) is disclosed.
[0007]
[0008] At present, there is no report on the patent of the crystal form of the acid salt of the compound. Screening and developing the crystal form of the compound represented by formula (A) to find a stable and reliable crystal form to ensure the quality stability of the compound represented by formula (A) so that it can be better used in pharmacy and drug preparations and future clinical use, so as to achieve stable and controllable drug quality, better dissolution and higher bioavailability, thereby avoiding safety issues such as side effects caused by toxic impurities generated by unstable drugs, which will be of great significance for later advancement. [0196] After long-term and in-depth research, the inventors have prepared a series of crystal forms of the acid salt compound shown in Formula A with excellent stability, especially the citrate crystal form I. The crystal form of the present invention has excellent stability and better pharmacokinetics than the free base crystal form, and has a higher in vivo exposure. On this basis, the inventors have completed the present invention.
[0197] Crystal form
[0198] Crystal form is the solid state of a drug. The study of drug crystal form is the study of the basic state of the drug. Only when we have a relatively full and comprehensive understanding of the crystal state of chemical drugs can we find more suitable drug crystal solid substances for treating diseases. The crystal form of a drug can affect the physical and chemical properties of the drug and directly affect the basis for the drug to play a role in treating diseases in clinical practice. Different crystal forms of the same drug may have significant differences in appearance, solubility, melting point, dissolution, bioavailability, etc., thereby affecting the stability, bioavailability and efficacy of the drug. Therefore, it is of great significance to study the stable crystal form of the compound.
[0199] The active ingredients of the same drug generally exist in two or more crystal forms, which are called drug polymorphs. Different crystal forms have different solubility and dissolution rates, which affect the clinical therapeutic effect of the drug by causing changes in bioavailability in the body. The difference in drug crystal forms may affect its dissolution and absorption in the body, and then affect the bioavailability, clinical efficacy and safety of the drug. At the same time, the stability of the drug crystal form is also very important. In order to improve the bioavailability of the drug, reduce toxicity, and enhance the therapeutic effect, more attention should be paid to the stability of the drug crystal form. Crystal forms with good stability can ensure the physicochemical stability of the drug dosage form during preparation and storage, maintain good solubility and bioavailability of the drug dosage form, and ensure the equivalence between each batch of drugs. The same drug often has multiple crystal forms. At present, the crystal form with better therapeutic effect and the most suitable clinically is called the dominant drug crystal form.
[0200] The present invention screens the crystal forms of the acid salt of the compound represented by formula (A) to find out as many different crystal forms as possible. The screened crystal forms are identified by powder X-ray diffraction analysis (XRPD), differential scanning calorimetry (DSC), thermogravimetric analysis (TGA), hydrogen spectrum liquid nuclear magnetic resonance (1H NMR) and high performance liquid chromatography (HPLC), and further by thermodynamic stability and hygroscopicity research, the superior drug citrate crystal form I is determined, which provides a reference for the selection of crystal forms in subsequent drug metabolism dynamics experiments and animal experiments.
[0201] After further pharmacokinetic research, the citrate crystal form I shown in formula (A) provided by the present invention is easily absorbed in vivo and has good stability. It has a good blood drug concentration distribution in vivo and high bioavailability. At the same time, due to its good physical stability, the shelf life will be longer, which is also very beneficial to the drug quality control in the later stage of formulation development, so that the drug can achieve better therapeutic effects.

After reacting at room temperature for about 4 hours, add 6 mL of methyl tert-butyl ether. After continuing the reaction for 1 day, filter the suspension, dry it under vacuum at 50 °C, and characterize it.
[0328] The DSC and TGA results of mesylate form II are as follows: Fig.17 As shown, the weight loss is 0.9% at 23-80°C, and there is an endothermic peak at 76°C corresponding to DSC. NMR shows that only 0.18% methyl tert-butyl ether remains. After the mesylate crystal form II was heated to 120°C by DSC, XRPD detection was immediately performed. The crystal form did not change. The weight loss should be caused by surface adsorption of water. Based on the above, it is speculated that the mesylate crystal form II should be anhydrous crystal form. The mesylate crystal form II has a group of crystal transformation peaks at 168-189°C. After the mesylate crystal form II is heated to 210°C, it is transformed into the mesylate crystal form I. The endothermic peak of the mesylate crystal form II at 257°C should be the melting point after heating and crystal transformation to the crystal form I.
[0329] The powder X-ray diffraction pattern of mesylate salt form II is as follows: Fig.16 The data are shown in Table 11:

[0366] 4. Evaluation of the crystal forms of the acid salt and free base of the compound represented by formula (A)
[0367] In this salt form screening study, 7 crystal forms were obtained: citrate crystal form I, maleate crystal form I, maleate crystal form II, oxalate crystal form I, oxalate crystal form II, methanesulfonate crystal form I, and methanesulfonate crystal form II.
[0368] Based on the solid-state characterization data of the salt and the free-state crystal screening results, citrate crystal form I, maleate crystal form II and methanesulfonate crystal form I were selected for solubility, stability and drug metabolism studies.
[0369] 4.1 Solubility study
[0370] The solubility of citrate salt Form I, maleate salt Form II, mesylate salt Form I and free base Form I was tested in biological media and water at 37° C. All results are shown in Table 19.
[0371] Table 19 Solubility test results in biological solvents and water
[0372]
[0373]
[0374] The solubility of the three salt forms, citrate form I, maleate form II and methanesulfonate form I, in water and biological solvents (FaSSIF and FeSSIF) is better than that of the free base form I, and their solubility in SGF is comparable.
[0375] 4.2 Solid-state stability studies
[0376] Appropriate amounts of citrate crystal form I, maleate crystal form II, and methanesulfonate crystal form I were spread flat in sample bottles and placed open at 80°C (3d), 60°C, 25°C / 60%RH, 40°C / 75%RH, and 25°C / 90±5%RH for 7 days. Samples were taken at 3 days and 7 days for HPLC and XRPD detection.
[0377] Stability studies have shown that: the crystal form and chemical purity of the citrate salt form I did not change significantly under the above five conditions; the maleate salt form II transformed into an unstable hydrated maleate in 7 days under 25°C / 90±5%RH, and no significant changes were observed under other conditions; the methanesulfonate salt form I did not change its crystal form under the above conditions. The results are summarized in Table 20.

[0379]
[0380] (a) The above purity values are the averages of 3 sets of parallel experiments
[0381] 4.3 Drug metabolism studies
[0382] The citrate salt form I, the methanesulfonate salt form I and the free base form I were subjected to drug metabolism experiments in rats.
[0383] SD rats were given a single oral administration of citrate crystal form I, methanesulfonate crystal form I and free base crystal form I, respectively. Blood samples were collected at different time points to determine the drug concentration in rat plasma after administration of the test substances and calculate the relevant pharmacokinetic parameters.
[0384] 4.3.1. Preparation of test sample solution
[0385] The preparation method is as follows:
[0386] Accurately weigh citrate crystal form I, mesylate crystal form I and free base crystal form I, add 5% DMSO, 10% solutol HS-15 and 85% saline, vortex mix and set aside.
[0387] 4.3.2. Analysis of test solution
[0388] The prepared test solution was tested and analyzed by the analytical department of this laboratory using LC-MS / MS.
[0389] 4.3.3 Animal reception and adaptation
[0390] The animals were fasted (no less than 10 hours) but not watered from the day before the test. On the day of the test, they were weighed and their tails were marked. Blank blood was collected before administration. Blood was collected from the tail vein.
[0391] 4.3.4 Sample collection and processing
[0392] The blood sampling time points are:
[0393] Oral administration: 0.5, 1, 2, 4, 6, 8, 12 and 24h after administration.
[0394] Collect 0.1 ml of whole blood in an EDTA-Na2 anticoagulant tube, mix by inverting 3-4 times, centrifuge at 10000g for 5 min at 4°C to separate plasma, and store at -80°C for testing. Blood was collected from the tail vein.
[0395] Plasma samples were analyzed by LC-MS / MS at the laboratory analytical department.
[0396] 4.3.5 Pharmacokinetic Analysis
[0397] According to the blood drug concentration data, the pharmacokinetic parameters AUC of the test products were calculated using the pharmacokinetic calculation software WinNonlin non-compartmental model. 0-last , C max 、Tmax and T 1 / 2 and its mean and standard deviation.
[0398] 4.3.6 Experimental Results
[0399] The main pharmacokinetic parameters in plasma after a single oral administration of citrate form I, mesylate form I and free base form I are shown in Table 21.
[0400] Table 21 Pharmacokinetic parameters in rats
[0401]
[0402] It can be seen from the above pharmacokinetic results that the in vivo exposure of citrate form I and mesylate form I in rats is higher than that of free base form I.
[0403] In summary, citrate crystal form I has good solubility, high exposure in animals, and excellent solid-state stability, making it suitable for further research and development.
