Description

[0001] Cross-reference to related applications

[0002] This application claims priority to U.S. Provisional Application No. 63 / 571,154, filed March 28, 2024, and European Application No. 24174251.9, filed May 6, 2024, the contents of which are incorporated herein by reference as if their entire contents were set forth herein. Technical Field

[0003] This disclosure relates to the field of flavorings or perfumes. More specifically, this disclosure relates to a composition comprising a cooling compound and at least one oil containing carvone, and optionally a solvent. This disclosure also relates to the use of said composition in consumer products such as flavored products and body care products. Background Technology

[0004] From simple metazoans to the most complex vertebrates, the entire animal kingdom possesses taste and sensory systems that provide sensory information about the chemical composition of the external world. This sensory information can encompass a wide range of taste sensations, including but not limited to a cooling sensation.

[0005] In the field of fragrances and flavorings, ingredients that impart a cold or cooling sensation are highly sought after. These cooling ingredients provide a refreshing and clean feeling, making them extremely popular in many flavored or aromatic consumer products. Over the past few decades, considerable effort has been devoted to finding and acquiring compounds with this cooling effect.

[0006] WO2012 / 061698 and WO2017 / 058594 report several highly potent cooling substances. These compounds are modulators of the transient receptor potential channel M8 (TRPM8), which is closely related to the generation of a cooling sensation. These compounds produce significant cooling effects even at extremely low concentrations; only trace amounts are needed to achieve the desired cooling sensation. However, these compounds are typically in the form of fine powders under ambient conditions (i.e., room temperature and normal pressure). They have low solubility in solvents commonly used in the flavoring and fragrance industries, and their potency makes them unsuitable for direct use (i.e., undiluted use), which poses difficulties for their processing and incorporation into consumer products.

[0007] Therefore, the industry continues to require suitable solvents to dissolve the aforementioned types of cooling compounds, so that these compounds can be provided in a more convenient diluted form. This disclosure provides a solution to the above problem by using an oil containing carvone. Summary of the Invention

[0031] In a first embodiment, this disclosure relates to a liquid composition comprising:

[0032] a) at least one compound of formula (I),

[0033]

[0034] The compound is in any of its stereoisomers; where R 1 Represents a phenyl group, which may optionally have one or two C atoms substituted. 1-3 alkyl or alkoxy or one or two halogen groups; R 2 and R 3 Each of these groups independently represents a heterocyclic group containing one to three heteroatoms selected from nitrogen, oxygen, and sulfur, which may optionally be substituted with one or two C atoms. 1-3 alkyl;

[0035] b) Oils containing carvone; and

[0036] c) Optional solvents.

[0037] For clarity, the phrase "any stereoisomer thereof" or similar expressions refer to the common meaning understood by those skilled in the art, namely, that a compound of formula (I) can be any diastereomer, enantiomer, or mixture thereof. Furthermore, a compound of formula (I) can also have at least one stereocenter, which can have different stereochemical configurations (i.e., when a stereocenter is present, compound (I) can have an (R) or (S) configuration). Each stereocenter can have a relative configuration of R or S, or a mixture thereof. The compound of formula (I) can be in the form of a pure enantiomer or diastereomer, or a mixture of stereoisomers.

[0038] According to any of the above implementation schemes, R 1Represents a phenyl group, which may optionally have one or two C atoms substituted. 1-3 Alkyl or alkoxy or one or two halogen groups. In one embodiment, R 1 Represents a phenyl group, which has one or two C atoms as substitutions. 1-3 Alkyl or alkoxy or one or two halogen atoms. In another embodiment, R 1 Represents a phenyl group, which has one or two C atoms as substitutions. 1-2 Alkyl or alkoxy or one or two fluorine atoms. In yet another embodiment, R 1 This represents a phenyl group, which is substituted with one or two methyl, ethyl, methoxy, or fluorine atoms. The substituents on the phenyl group can be located relative to the oxygen atom and R. 1 The ortho, meta, and / or para positions of the bonds between groups. In one embodiment, R 1 It can be selected from the group consisting of p-tolyl, 4-ethylphenyl, 4-methoxyphenyl, 4-fluorophenyl, 3,4-dimethylphenyl, and m-tolyl. In another embodiment, R 1 It can be p-tolyl or 4-ethylphenyl. In yet another embodiment, R 1 It can be p-tolyl.

[0039] As used herein, the terms “ortho,” “meta,” and “para” are used in the usual sense in the art, namely, ortho refers to two substituents on a phenyl group occupying adjacent positions; meta refers to two substituents on a phenyl group occupying positions 1 and 3; and para refers to two substituents on a phenyl group occupying positions 1 and 4.

[0040] According to any of the above implementation schemes, R 2 and R 3 Each of these groups independently represents a heterocyclic group containing one to three heteroatoms selected from nitrogen, oxygen, and sulfur, which may optionally be substituted with one or two C atoms. 1-3 Alkyl group. In one embodiment, R 2 and R 3 Each of these groups independently represents a heterocyclic group containing one to three heteroatoms selected from nitrogen, oxygen, and sulfur. In another embodiment, R 2 and R 3 Each of these groups independently represents a heterocyclic group containing one to three heteroatoms selected from nitrogen and sulfur.

[0041] According to any of the above implementation schemes, R 2 This represents a heterocyclic group containing one or two nitrogen atoms. In one embodiment, R 2 It represents the pyrazole group.

[0042] According to any of the above implementation schemes, R 3This represents a heterocyclic group containing one or two sulfur atoms. In one embodiment, R 3 Represents thiophene group.

[0043] According to any of the above embodiments, the compound of formula (I) can be selected from the group consisting of: N-(1H-pyrazol-3-yl)-N-(thiophen-2-ylmethyl)-2-(p-tolyloxy)acetamide, 2-(4-fluorophenoxy)-N-(1H-pyrazol-3-yl)-N-(thiophen-2-ylmethyl)acetamide, 2-(4-ethylphenoxy)-N-(1H-pyrazol-3-yl)-N- (thiophene-2-ylmethyl)acetamide, 2-(4-methoxyphenoxy)-N-(1H-pyrazole-3-yl)-N-(thiophene-2-ylmethyl)acetamide, N-(1H-pyrazole-3-yl)-N-(thiophene-2-ylmethyl)-2-(m-tolyloxy)acetamide, and 2-(3,4-dimethylphenoxy)-N-(1H-pyrazole-3-yl)-N-(thiophene-2-ylmethyl)acetamide. The compounds all possess a pyrazole group. This group is known to exist in two tautomer forms, namely 1H-pyrazole-3-yl and 1H-pyrazole-5-yl. These two isomers readily reach equilibrium. In other words, when a composition contains N-(1H-pyrazol-3-yl)-N-(thiophen-2-ylmethyl)-2-(p-toluoxy)acetamide, the composition necessarily also contains N-(1H-pyrazol-5-yl)-N-(thiophen-2-ylmethyl)-2-(p-toluoxy)acetamide. The above conclusion also applies to the other compounds described herein. For example, 2-(4-fluorophenoxy)-N-(1H-pyrazol-3-yl)-N-(thiophen-2-ylmethyl)acetamide is in equilibrium with 2-(4-fluorophenoxy)-N-(1H-pyrazol-5-yl)-N-(thiophen-2-ylmethyl)acetamide; 2-(4-ethylphenoxy)-N-(1H-pyrazol-3-yl)-N-(thiophen-2-ylmethyl)acetamide is in equilibrium with 2-(4-ethylphenoxy)-N-(1H-pyrazol-5-yl)-N-(thiophen-2-ylmethyl)acetamide; 2-(4-methoxyphenoxy)-N-(1H-pyrazol-3-yl)-N-(thiophen-2-ylmethyl)acetamide is in equilibrium with 2-( 4-Methoxyphenoxy)-N-(1H-pyrazol-5-yl)-N-(thiophen-2-ylmethyl)acetamide is in equilibrium; N-(1H-pyrazol-3-yl)-N-(thiophen-2-ylmethyl)-2-(m-tolyloxy)acetamide is in equilibrium with N-(1H-pyrazol-5-yl)-N-(thiophen-2-ylmethyl)-2-(m-tolyloxy)acetamide; 2-(3,4-dimethylphenoxy)-N-(1H-pyrazol-3-yl)-N-(thiophen-2-ylmethyl)acetamide is in equilibrium with 2-(3,4-dimethylphenoxy)-N-(1H-pyrazol-5-yl)-N-(thiophen-2-ylmethyl)acetamide. In one embodiment, the compound of formula (I) is N-(1H-pyrazol-3-yl)-N-(thiophen-2-ylmethyl)-2-(p-tolyloxy)acetamide.

[0044] Surprisingly, it was discovered that the compound of formula (I) can be dissolved at a useful concentration using an oil containing carvone (and optionally a solvent), thereby obtaining a clear and homogeneous solution. The term "clear" means that the solution has 100% transmittance at a 1 cm optical path length in the visible light range (500–800 nm) with deionized water as a reference, without the addition of colorants or fluorescent agents. Therefore, in one embodiment, the liquid composition is a clear and homogeneous solution.

[0045] In one embodiment, the transparent and homogeneous solution is also stable. The term "stable" means that the liquid composition does not undergo phase separation and remains transparent without detectable turbidity, clouding, or precipitation at room temperature or within a certain temperature range (e.g., room temperature ± 10 to 20°C, typically between 10°C and 45°C, more typically between 3°C and 45°C), and remains stable over a relatively long period of time (typically at least a few days, more typically at least a few weeks, or even longer).

[0046] Oils containing carvone can be from any carvone source known to those skilled in the art. Oils containing carvone are generally naturally occurring oils known to those skilled in the art. Carvone (2-methyl-5-(prop-1-en-2-yl)cyclohex-2-en-1-one) is a terpene compound that can be obtained from natural sources by chemical synthesis or extraction using known methods. Carvone has two enantiomers: R-(-)-carvone (also known as levorotatory (L)-carvone) and S-(+)-carvone (also known as dextrorotatory (D)-carvone). This disclosure contemplates racemic mixtures of carvone and its enantiomers and their resolved forms. Racemic mixtures of carvone and its enantiomers and their resolved forms can be pure, i.e., unmixed with other substances, or can be part of a mixture (e.g., an extract). In one embodiment, the oil containing carvone can be pure carvone, spearmint oil, caraway seed oil, or dill oil. The sweetener may be a low-calorie, zero-calorie, or high-intensity sweetener. The term "high-intensity sweetener" refers to a sweetener that is sweeter than sucrose, for example, at least 10 times, or at least 20 times, or at least 50 times, or at least 100 times, or at least 250 times, or at least 500 times, or at least 750 times, or at least 1000 times, or at least 2500 times, or at least 5000 times, or at least 7500 times, or at least 10000 times sweeter than sucrose. In this document, saying that a sweetener is x times sweeter than sucrose means that a concentration of the sweetener in an aqueous solution relative to 1 / x the weight of sucrose produces a sweetness equivalent to that of sucrose.

[0062] Examples of suitable sweeteners are described in more detail below. In some embodiments, the sweetener is sucralose, which refers to 1,6-dichloro-1,6-dideoxy-β-D-fructofuranosyl-4-chloro-4-deoxy-α-D-galactopyranoside. In some embodiments, the sweetener is sodium saccharin. Sodium saccharin may be present at any suitable concentration. As used herein, the term "GSG" may be used interchangeably with "glucosylated natural steviol glycoside".

[0064] In embodiments where a GSG is present, any suitable GSG can be used. Therefore, the GSG can have any suitable degree of additional glucosylation, and any suitable type of glucosylation. In some embodiments, the GSG used has a glucosylation degree of at least 1.0, meaning that at least one mole of glucose has been enzymatically added to the natural steviol glycoside for every mole of natural steviol glycoside. In other embodiments, the GSG used has a glucosylation degree of at least 1.1, or at least 1.2, or at least 1.3, or at least 1.4, or at least 1.5, or at least 1.6, or at least 1.7, or at least 1.8, or at least 1.9, or at least 2.0, or at least 2.1, or at least 2.2, or at least 2.3, or at least 2.4, or at least 2.5. The glucosylation can be of any suitable type. For example, in some embodiments, the glucose enzymatically added to the natural steviol glycoside is α-glucose, β-glucose, or a combination thereof. In some embodiments, the glucose added enzymatically to the natural steviol glycoside is α-glucose. In some embodiments, the glucose added enzymatically to the natural steviol glycoside is β-glucose. In some embodiments, the glucose added enzymatically to the natural steviol glycoside is a combination of α-glucose and β-glucose. The glucose added via enzymatic glucosylation can be incorporated through any suitable linkage. Typically, the added glucose unit is linked to other glucose units already present on the natural steviol glycoside. In some embodiments, the enzymatically added glucose unit is added via a 1,6 bond, meaning the glucose unit is added via the 1-position of the added glucose unit and the 6-position of a glucose unit already present on the natural steviol glycoside. In some other embodiments, the enzymatically added glucose unit is added via a 1,4 bond. In some other embodiments, the enzymatically added glucose unit is added via a 1,2 bond.

[0156] The compositions and consumer products based on this disclosure are further illustrated by the following non-limiting examples.

[0157] Example

[0158] Solubility measurement:

[0159] Compound N-(1H-pyrazol-3-yl)-N-(thiophen-2-ylmethyl)-2-(p-tolyloxy)acetamide of formula (I) was mixed with a liquid solvent or a mixture of solvents and transferred to an 8 mL vial equipped with a top-mounted stirrer. The sample was placed in a Crystalline PV instrument (Technobis, Netherlands). Heating and cooling steps were performed with mechanical stirring, and the transparency of the sample was monitored by an LED beam passing through the vial. Complete (100%) transparency was considered achieved when N-(1H-pyrazol-3-yl)-N-(thiophen-2-ylmethyl)-2-(p-tolyloxy)acetamide was completely dissolved in the solvent. Conversely, when the measured transparency value was less than 100%, the dissolution process was considered incomplete.

[0160] Determination of dissolution temperature:

[0161] First, set the temperature to 0℃ and allow the sample to stand for 5 minutes to reach equilibrium without stirring. Then, start stirring and heat at a slow rate (+0.1℃ / min) to dissolve the sample under quasi-isothermal conditions. Monitor the change in sample transparency over time during the heating process. The dissolution process is complete when the transparency reaches 100%, and this temperature is the dissolution temperature.

[0162] The solubility of N-(1H-pyrazol-3-yl)-N-(thiophen-2-ylmethyl)-2-(p-tolyloxy)acetamide in various solvents at 22 °C was determined using pre-established solubility curves (fitted from the solubility temperatures of at least four different concentrations of N-(1H-pyrazol-3-yl)-N-(thiophen-2-ylmethyl)-2-(p-tolyloxy)acetamide samples in different solvents). These solubility curves were fitted to an exponential function. The solubility at 22 °C was obtained by extrapolation or interpolation of this exponential function.

[0163] The percentages (%) shown in the examples are the weight percentages of N-(1H-pyrazol-3-yl)-N-(thiophen-2-ylmethyl)-2-(p-tolyloxy)acetamide in the total liquid composition.

[0164] Example 1. Solvent Comparison

[0165] The compound N-(1H-pyrazol-3-yl)-N-(thiophen-2-ylmethyl)-2-(p-toluoxy)acetamide of formula (I) was dissolved in various solvents and solvent blends. The mass percentage of N-(1H-pyrazol-3-yl)-N-(thiophen-2-ylmethyl)-2-(p-toluoxy)acetamide dissolved at 22 °C and the dissolution temperature of a 10% solution of N-(1H-pyrazol-3-yl)-N-(thiophen-2-ylmethyl)-2-(p-toluoxy)acetamide were determined. Table 1 summarizes the dissolution results of N-(1H-pyrazol-3-yl)-N-(thiophen-2-ylmethyl)-2-(p-tolyloxy)acetamide in classical solvents, and Table 2 summarizes the dissolution results of N-(1H-pyrazol-3-yl)-N-(thiophen-2-ylmethyl)-2-(p-tolyloxy)acetamide in carvone alone and in blends of carvone and cosolvents.
