Abstract
Description
[Technical field]
[0001] The present invention relates to a reactor used in a chemical solution synthesizer for synthesizing a chemical solution. [Background technology]
[0002] In a chemical synthesis apparatus for chemically synthesizing proteins, peptides, polymers, nucleic acids, etc., multiple chemical solutions (reagents) are supplied to a reaction vessel (hereinafter referred to as a reactor) to perform chemical synthesis. In this reactor, for example, when synthesizing nucleic acids, multiple carriers (porous beads) are provided in the reactor, and while chemical solutions are sequentially supplied into the reactor, processes such as detritylation, coupling, oxidation, and capping are repeated to successively extend the bases attached to the carrier.
[0003] The reactor used in such a chemical solution synthesis apparatus is, for example, a cylindrical container made of glass or the like. Specifically, as shown in Fig. 6, the reactor 900 has a cylindrical reactor body 910 extending in one direction for chemically synthesizing the supplied chemical solution, an end cap part 920 for connecting a pipe for supplying the chemical solution to the reactor body 910, and a case part 930 into which the reactor body 910 is inserted, and the reactor 900 is formed by fastening these parts with a fastening part 940. That is, as shown in Fig. 7, the end cap part 920 has an insertion part 921 into which the axial end of the reactor body 910 is inserted, and in a state in which the axial end of the reactor body 910 inserted into the case part 930 is inserted into the insertion part 921, the end cap part 920 and the case part 930 are fastened by the fastening part 940, so that the reactor body 910 is held by the end cap part 920 and the case part 930 (for example, Patent Document 1 below).
[0004] In the reactor 900, the gap between the inner peripheral surface of the insertion part 921 and the outer peripheral surface of the reactor main body part 910 is sealed by the O-ring 950. Specifically, as shown in FIG. 7, the insertion part 921 has a notch part 922 at its opening, the diameter of which decreases toward the direction in which the axial end part of the reactor main body part 910 is inserted, and the O-ring 950 is accommodated in the gap formed by the notch part 922, the axial end face 931 of the case part 930, and the outer peripheral surface of the reactor main body part 910. The O-ring 950 is pressed in the axial direction to seal the gap between the inner peripheral surface of the insertion part 921 and the outer peripheral surface of the reactor main body part 910. As a result, the chemical solution is supplied to the reactor main body part 910 without leaking out to the outside, and chemical synthesis is performed in the reactor main body part 910.
[0005] In such a reactor 900, particularly when used in a chemical synthesis device, the purity of the chemical is important, and in order to prevent mixing of a chemical used previously and a chemical used next in chemical synthesis in the reactor 900, a cleaning liquid is sent to the reactor 900 to clean the inside of the reactor 900. [0006] [Patent Document 1] JP 2019-163216 A Summary of the Invention [Problem to be solved by the invention]
[0007] However, in the reactor 900, there were cases where the inside of the reactor 900 could not be sufficiently cleaned. That is, in the reactor 900, as shown in FIG. 7, the gap between the inner circumferential surface of the inserting portion 921 of the end cap portion 920 and the outer circumferential surface of the reactor 900 was sealed by the O-ring 950 at a position away from the axial end surface 911 of the reactor main body portion 910, so that the portion between the O-ring 950 and the axial end surface 911 of the reactor main body portion 910 in the gap became a dead space, and the chemical liquid flowed into the dead space. Once the chemical liquid flows into the dead space, it is difficult to exchange the chemical liquid that has flowed into the dead space with the cleaning liquid, and it is difficult to exert the cleaning effect of the cleaning liquid on the dead space, so that the chemical liquid used previously tends to remain in the dead space. Therefore, the chemical liquid used previously may mix with the chemical liquid to be used next, and the purity of the chemical liquid to be used may decrease.
[0008] The present invention has been made in consideration of the above problems, and has an object to provide a reactor whose inside can be easily cleaned without impairing the sealing performance. [Means for solving the problem]
[0009] In order to solve the above problems, the reactor of the present invention is a reactor comprising a cylindrical reactor main body extending in one direction for reacting a supplied chemical solution, and end cap portions provided at both axial ends of the reactor main body and having insertion portions into which the axial ends of the reactor main body are inserted, and further comprises a sealing portion for sealing a gap between the insertion portion and the end face at the axial end of the reactor main body when the axial end of the reactor main body is inserted into the insertion portion.
[0010] According to the above reactor, the seal portion can seal the gap between the end cap portion and the reactor body portion at the axial end of the reactor body portion, thereby preventing the chemical solution from flowing into the dead space formed between the inner peripheral surface of the end cap portion and the outer peripheral surface of the reactor body portion. This makes it possible to thoroughly clean the inside of the reactor with the cleaning solution. Therefore, cleaning of the inside of the reactor can be facilitated without impairing the sealing property.
[0011] As a specific aspect, it is preferable that the sealing portion is formed as a part of the insertion portion, and that by inserting the axial end portion of the reactor main body portion into the insertion portion, the sealing portion is pressed against the end surface at the axial end portion of the reactor main body portion and tightly adheres to the end surface.
[0012] In addition, as a specific aspect, it is preferable that the sealing portion is arranged between the insertion portion and the end face at the axial end of the reactor main body, and by inserting the axial end of the reactor main body into the insertion portion, the sealing portion is pressed against the insertion portion and the end face at the axial end of the reactor main body, thereby being in close contact with the insertion portion.
[0013] In addition, the sealing portion may have a tapered shape that narrows in the direction in which the axial end of the reactor main body is inserted into the insertion portion, and the sealing portion may be configured to be pressed against the outer edge of the end face at the axial end of the reactor main body.
[0014] According to this configuration, the seal part having a tapered shape that is inclined so as to narrow toward the direction in which the axial end of the reactor main body is inserted is pressed against the outer edge of the end face at the axial end of the reactor main body, so that the seal part can be deformed to conform to the outer circumferential surface of the reactor main body and the end face at the axial end of the reactor main body, thereby improving the adhesion between the seal part and the reactor main body, thereby improving the sealing performance of the seal part.
[0015] The reactor may further include a reactor holding portion for holding the axial end portion of the reactor body in a state inserted into the insertion portion, and the seal portion may be pressed by the reactor holding portion.
[0016] According to this configuration, the reactor holding portion can maintain a state in which the seal portion is in intimate contact with the end surface at the axial end of the reactor main body.
[0017] As a specific aspect, it is preferable that the reactor holding portion is configured to include a case portion having an insertion portion through which the reactor main body portion is inserted, and a fastening portion that fastens the end cap portion and the case portion together in a state in which the reactor main body portion is inserted into the insertion portion and the axial end portion of the reactor main body portion is inserted into the insertion portion.
[0018] As a specific aspect, it is preferable that the reactor holding portion has a screw portion for screwing the insertion portion onto the outer circumferential surface of the reactor main body.
[0019] In addition, a filter to be inserted into the reactor main body may be provided on both axial ends of the reactor main body, and the end cap may be formed with a filter holding portion that holds the filter within the reactor main body by abutting against the filter when the axial end of the reactor main body is inserted.
[0020] According to this configuration, the filter is held within the reactor main body by the filter holding portion, so that the filter can be used as a lid for the reactor main body, and the compound can be prevented from spilling out of the reactor main body even if the reactor main body is tilted.
[0021] Further, a filter protection portion for protecting the filter may be provided between the filter and the filter holding portion, and the filter holding portion may be configured to abut against the filter via the filter protection portion.
[0022] According to this configuration, since the filter protection portion is provided between the filter and the filter holding portion, the filter is less susceptible to wear than when the filter holding portion is in direct contact with the filter. Effect of the Invention
[0023] According to the reactor of the present invention, the inside can be easily cleaned without impairing the sealing performance. [Brief description of the drawings]

[0025] DETAILED DESCRIPTION OF THE PREFERRED EMBODIMENTS An embodiment of a reactor according to the present invention will be described with reference to the drawings.
[0026] Fig. 1 is a diagram that shows a schematic diagram of a chemical liquid synthesis apparatus 100 including a reactor 2 of the present invention. Fig. 2 and Fig. 3 are diagrams for explaining the reactor 2 in one embodiment of the present invention, Fig. 2 is a cross-sectional view of the reactor 2, and Fig. 3 is an enlarged view of an end of the reactor 2. Note that, in this embodiment, an example in which a chemical liquid (reagent) is used as the fluid will be described, but the present invention is not limited to a chemical liquid, and can also be applied to cases in which a liquid other than a chemical liquid is chemically synthesized, mixed, or the like.
[0027] As shown in Fig. 1, the chemical synthesis apparatus 100 includes a container 11 in which a chemical is stored, a reactor 2 in which a carrier (porous beads) (not shown) is stored, and a waste tank 12 in which the chemical discharged from the reactor 2 is stored, all of which are connected by a pipe 3. Then, by sequentially supplying the predetermined chemical required for the reaction from the container 11 to the reactor 2, the bases attached to the carrier are successively extended by repeating processes such as detritylation, coupling, oxidation, and capping. This produces synthetic products such as peptides and nucleic acids. Then, the chemical after the reaction is completed is discharged from the reactor 2 to the waste tank 12.
[0028] The storage vessel 11 is for storing a chemical solution used in chemical synthesis. In the example of Fig. 1, a plurality of storage vessels 11 are provided, and each storage vessel 11 is connected to the reactor 2 by a pipe 31. The chemical solution stored in the storage vessel 11 is sent to the reactor 2 through the pipe 31. The pipe 31 is formed of a hollow tubular member, and the pipes 32, 33, ... that appear in the following description have the same configuration. The pipes 31, 32, ... will be simply referred to as pipe 3 unless there is a need to distinguish them from one another.
[0029] In addition, a pressurizing means (gas tank) 13 is connected to the storage container 11 through a pipe 32. When the pressurizing means 13 supplies gas to the reactor 2 through the pipe 32, the storage container 11 is pressurized and the chemical liquid is sent to the reactor 2 through the pipe 31. A valve 14 is provided on the pipe 31 and the pipe 32, and by switching the open / close state of the valve 14, only the chemical liquid selected from the multiple storage containers 11 can be sent to the reactor 2. The gas supplied to the storage container 11 by the pressurizing means 13 is a gas that does not react with the chemical liquid stored in the storage container 11 (for example, an inert gas such as argon gas).
[0030] Moreover, one of the storage containers 11 contains a cleaning liquid. This cleaning liquid is for cleaning the reactor 2. That is, when a specific chemical liquid is sent to the reactor 2 and the specific chemical liquid cannot be mixed with the previously sent chemical liquid, the previous chemical liquid is sent to the reactor 2, and then the cleaning liquid is sent before the next chemical liquid is sent to the reactor 2, thereby cleaning the reactor 2. Moreover, the cleaning liquid is sent in a configuration similar to that of the aforementioned chemical liquid.
[0031] Further, downstream of the reactor 2, a waste liquid tank 12 is provided for storing the chemical liquid and the like discharged from the reactor 2. The waste liquid tank 12 is connected to the reactor 2 by a pipe 33, and the chemical liquid discharged from the reactor 2 is sent to the waste liquid tank 12 through the pipe 33. A valve 14 is provided on the pipe 33, and the discharge of the chemical liquid from the reactor 2 to the waste liquid tank 12 and the discharge are stopped by switching the open / close state of the valve 14. Further, the waste liquid tank 12 is formed to have a larger capacity than the reactor 2, and is formed to have a capacity capable of storing the chemical liquid and the like even when the chemical liquid is discharged from the reactor 2 multiple times.
[0032] The reactor 2 provides a reaction field for chemically synthesizing the supplied chemical solution. As shown in FIG. 2, the reactor 2 includes a cylindrical reactor body 21 extending in one direction for reacting the supplied chemical solution, and an end cap portion 4 having an insertion portion 41 into which the axial end portion of the reactor body 21 is inserted. The reactor body 21 in this embodiment is a cylindrical glass tube extending in one direction, and contains a large number of carriers inside. When the chemical solution is supplied into the reactor body 21, the chemical solution and the carrier are chemically synthesized, and the base attached to the carrier is elongated. As a result, a synthetic product is produced in the reactor body 21.
[0033] The end cap parts 4 are for connecting the reactor body part 21 and the piping 3, and are provided at both axial ends of the reactor body part 21 as shown in Fig. 2. That is, the piping 31 is connected to one end cap part 4, and the piping 33 is connected to the other end cap part 4. By inserting the axial ends of the reactor body part 21 into these end cap parts 4, the reactor body part 21 and the storage container 11, and the reactor body part 21 and the waste liquid tank 12 are connected. As a result, the chemical liquid sent from the storage container 11 is supplied to the reactor body part 21 through the piping 31 and the end cap part 4, and the chemical liquid is discharged from the reactor body part 21 to the waste liquid tank 12 through the end cap part 4 and the piping 33.
[0034] 2 and 3, the reactor 2 further includes a seal portion 5 for preventing leakage of the chemical solution from the reactor 2. The seal portion 5 in this embodiment is formed as a part of the insertion portion 41, and is formed at a position where the seal portion 5 is pressed against the end face 22 at the axial end of the reactor main body 21 when the axial end of the reactor main body 21 is inserted into the insertion portion 41. The seal portion 5 is formed of a material having chemical resistance and elasticity, such as PTFE (polytetrafluoroethylene) or PEEK (polyether ether ketone).
[0035] That is, when the axial end of the reactor main body 21 is inserted into the insertion portion 41, the seal portion 5 is pressed against the end face 22 at the axial end of the reactor main body 21, and the pressed seal portion 5 is deformed along the end face 22 at the axial end of the reactor main body 21, so that the seal portion 5 is in close contact with the reactor main body 21. As a result, the seal portion 5 can seal the gap between the end cap portion 4 and the reactor main body 21 at the end face 22 at the axial end of the reactor main body 21, so that the chemical solution can be prevented from flowing into the dead space formed between the inner peripheral surface of the end cap portion 4 and the outer peripheral surface of the reactor main body 21.
[0036] The seal portion 5 has a tapered shape that is inclined so as to narrow toward the direction in which the axial end of the reactor main body 21 is inserted into the insertion portion 41, and the seal portion 5 is pressed against the outer edge of the end face 22 at the axial end of the reactor main body 21. That is, when the axial end of the reactor main body 21 is inserted into the insertion portion 41, the seal portion 5 is pressed against the outer edge of the end face 22 at the axial end of the reactor main body 21, and the pressed seal portion 5 deforms to fit along the outer peripheral surface of the reactor main body 21 and the end face 22 at the axial end of the reactor main body 21, thereby increasing the adhesion between the seal portion 5 and the reactor main body 21. Therefore, the sealing performance by the seal portion 5 can be improved.
[0037] 2 and 3, the reactor 2 further includes a reactor holding part 6 that holds the axial end of the reactor body 21 inserted into the insertion part 41, and the reactor holding part 6 presses the seal part 5. In other words, the reactor holding part 6 maintains the seal part 5 in intimate contact with the end face 22 at the axial end of the reactor body 21.
[0038] 2 and 3, the reactor holding part 6 in this embodiment has a case part 61 having an insertion part 62 through which the reactor body part 21 is inserted, and a fastening part 63 that fastens the end cap 4 to the case part 61. That is, in a state in which the reactor body part 21 is inserted into the insertion part 41 by inserting the reactor body part 21 into the insertion part 62 and the axial end part of the reactor body part 21 is inserted into the insertion part 41, the end cap part 4 and the case part 61 are fastened by the fastening part 63, thereby holding the reactor body part 21 in a state in which the axial end part is inserted into the insertion part 41.
[0039] The case part 61 covers the reactor body part 21 except for the axial end part, and has an insertion part 62 that covers the outer circumferential surface of the reactor body part 21 and accommodates the reactor body part 21 therein. The fastening part 63 is for fastening the end cap part 4 to the case part 61, and has a restricting part 64 that restricts the end cap part 4 from moving in the axial direction, and the restricting part 64 is configured to push the end cap part 4 toward the axial end part of the reactor body part 21. That is, a male screw part 65 is formed on the outer circumferential surface of the case part 61, and a female screw part 66 is formed on the inner circumferential surface of the fastening part 63, and the end cap part 4 is fastened to the case part 61 by screwing the male screw part 65 and the female screw part 66 together. Therefore, when the male screw portion 65 and the female screw portion 66 are screwed together, the regulating portion 64 pushes the end cap portion 4 toward the axial end of the reactor main body portion 21, and the sealing portion 5 is pressed against the end face 22 at the axial end of the reactor main body portion 21, thereby tightly adhering the sealing portion 5 and maintaining that state.
[0040] 2 and 3, a filter 7 is provided at both axial ends of the reactor body 21 to be inserted into the reactor body 21. The filter 7 is intended to prevent impurities from entering the reactor body 21. The filter 7 has a tapered shape in which the diameter decreases toward the direction of insertion into the reactor body 21, and is inserted into the reactor body 21 while being compressed. When the filter 7 is inserted into the reactor body 21, a part of the filter 7 remains outside the reactor body 21. Specifically, the end of the filter 7 in the direction opposite to the direction in which the filter 7 is inserted into the reactor body 21 is formed to have a diameter larger than the inner diameter of the reactor body 21, and this part remains outside the reactor body 21 when the filter 7 is inserted. This makes it possible to more easily access the filter 7 and remove it from the reactor body 21 when taking out the compound from the reactor body 21 than when the entire filter 7 is inserted into the reactor body 21.
[0041] 3, the end cap portion 4 is formed with a filter holding portion 42 for holding the filter 7 in the reactor body portion 21. The filter holding portion 42 is formed to abut against the filter 7 when the axial end of the reactor body portion 21 is inserted into the insertion portion 41. That is, the inner diameter of a part of the end cap portion 4 is formed smaller than the diameter of the filter 7, thereby forming the filter holding portion 42 for abutting against the filter 7. As a result, the filter 7 can be held in the reactor body portion 21, and the filter 7 can be used as a lid for the reactor body portion 21, and the compound in the reactor body portion 21 can be prevented from spilling out even if the reactor body portion 21 is tilted.
[0042] 3, the reactor 2 is provided with a filter protection part 8 for protecting the filter 7. The filter protection part 8 is for preventing wear of the filter 7 and is made of a material softer than the filter holding part 42, for example, POM (polyacetal) or nylon. By disposing the filter protection part 8 between the filter 7 and the filter holding part 42 and abutting the filter holding part 42 against the filter 7 via the filter protection part 8, the filter 7 is less likely to wear than when the filter holding part 42 is directly abutted against the filter 7. When the axial end of the reactor main body 21 is inserted into the insertion part 41, the filter protection part 8 is pressed by the filter holding part 42 and deformed. This deformation of the filter protection part 8 makes it possible to correct the position of the seal part 5 so that the seal part 5 is pressed at the position where it should be pressed.
[0043] In addition, as shown in FIG. 2 and FIG. 3, the end cap section 4 connected to the storage container 11 by the piping 31 is formed with a buffer section 43 for temporarily storing the chemical solution and an exhaust section 44 for exhausting gas from the end cap section 4. The buffer section 43 is a space formed between the part where the end cap section 4 and the piping 31 are connected and the filter 7, and is capable of storing the chemical solution sent to the end cap section 4 through the piping 31 until it is supplied to the reactor body section 21 through the filter 7. The exhaust section 44 is an exhaust port for exhausting gas from the end cap section 4, and is connected to the piping 34. The end cap section 4 can be depressurized by exhausting gas from the exhaust section 44 through the piping 34. This makes it possible to prevent the pressure in the end cap section 4 from becoming excessively large.
[0044] As described above, according to the reactor 2 in the above embodiment, the seal portion 5 seals the gap between the end cap portion 4 and the reactor body portion 21 at the end surface 22 at the axial end of the reactor body portion 21, so that it is possible to prevent the chemical solution from flowing into the dead space formed between the inner peripheral surface of the end cap portion 4 and the outer peripheral surface of the reactor body portion 21. This makes it possible to thoroughly clean the inside of the reactor 2 with the cleaning solution. Therefore, it is possible to easily clean the inside of the reactor 2 without impairing the sealing property.
[0045] In the above embodiment, the seal portion 5 is formed as a part of the insert portion 41, but the seal portion 5 and the insert portion 41 may be configured to be separate. Specifically, the seal portion 5 is a packing, and is disposed between the insert portion 41 and the end surface 22 at the axial end of the reactor main body 21, as shown in FIG. 4. By inserting the axial end of the reactor main body 21 into the insert portion 41, the seal portion 5 is pressed and crushed by the insert portion 41 and the end surface 22 at the axial end of the reactor main body 21, and the seal portion 5 is in close contact with the reactor main body 21. As a result, the seal portion 5 can seal the gap between the end cap portion 4 and the reactor main body 21 at the end surface 22 at the axial end of the reactor main body 21.
[0046] Furthermore, when the seal part 5 and the insertion part 41 are configured to be separate, the seal part 5 may be used as the filter protection part 8. That is, the seal part 5 is formed of a material softer than the filter holding part 42, and is arranged so as to be disposed not only between the insertion part 41 and the end face 22 at the axial end of the reactor main body 21, but also between the filter 7 and the filter holding part 42, so that the filter holding part 42 abuts against the filter 7 via the seal part 5. This makes the filter 7 less susceptible to wear than when the filter holding part 42 abuts directly against the filter 7.
[0047] In the above embodiment, an example has been described in which the reactor holding part 6 has the case part 61 and the fastening part 63, but the reactor holding part 6 may have a screw part for screwing the insertion part 41 into the outer circumferential surface of the reactor main body part 21. Specifically, as shown in Fig. 5, the reactor holding part 6 has a female screw part 45 formed on the inner circumferential surface of the insertion part 41 and a male screw part 23 formed on the outer circumferential surface of the reactor main body part 21. Then, by screwing the female screw part 45 into the male screw part 23, the axial end part of the reactor main body part 21 can be inserted into the end cap part 4 and held in that state.
[0048] The above describes the embodiments of the present invention in detail with reference to the drawings. However, the configurations and combinations thereof in each embodiment are merely examples, and additions, omissions, substitutions, and other modifications of the configurations are possible without departing from the spirit of the present invention.
