Abstract
Description
[Technical field]
[0001] The present invention relates to an external skin preparation and a kit for an external skin preparation. [Background technology]
[0002] There are external skin preparations called peeling agents that have the effect of peeling off the old stratum corneum on the surface of the skin, regenerating a new stratum corneum, and promoting the turnover (metabolism) of the epidermis. Some conventional peeling agents peel off the necessary stratum corneum and forcefully speed up the turnover cycle of the skin. In this case, the skin becomes thinner and the skin becomes more transparent temporarily, but the immature stratum corneum is pushed up to the surface of the skin, which weakens the skin's barrier function and can cause rough skin such as dryness, wrinkles, pimples, and acne.
[0003] Patent Document 1 discloses a peeling agent that contains 15-25% by mass of horsetail, 10-20% by mass of white poria, 0.2-1% by mass of dokudami, 15-25% by mass of han-ge-shou, 10-20% by mass of white poria, and 25-35% by mass of sulfur, as a peeling agent that effectively removes keratin without side effects. White poria refers to the dried silkworm larvae of the Bombyx mori family that are infected with white poria bacteria, become white, become rigid, and die. White poria is known as a Chinese herbal medicine that has the effects of releasing sputum, stopping spasms, and reducing phlegm, and is sometimes taken to suppress fever, headache, sore throat, spasms, and itching caused by rubella due to colds. [0004] [Patent Document 1] Korean Patent No. 10-0791089 Summary of the Invention [Problem to be solved by the invention]
[0005] As a result of the inventor's investigations, it was found that the peeling agent described in Patent Document 1 may, in rare cases, be unsuitable for the skin of some users, causing skin troubles such as rashes, redness, and itchiness.
[0006] Sulfur has a reducing effect and reacts with proteins (such as keratin) on the surface of the skin to soften the keratin, so it is sometimes used in peeling agents to remove damaged keratin. However, the use of high concentrations of sulfur can cause dermatitis as a side effect. As described above, in the peeling agent described in Patent Document 1, sulfur is used at a high concentration of 25 to 35 mass% of the total solid content. Therefore, it was initially thought that the high concentration of sulfur was the cause of the above skin troubles, but further intensive research by the present inventors revealed that the use of high concentrations of sulfur alone was not the cause of the above skin troubles.
[0007] To provide an external preparation for skin and a kit for an external preparation for skin that can promote skin turnover and naturally regenerate a new stratum corneum without causing skin troubles such as rash, redness, and itchiness. [Means for solving the problem]
[0008] A first aspect of the present invention provides an external skin preparation containing a plant-derived component, a keratin softener, and silk powder.
[0009] In this embodiment, the keratin softening agent may be sulfur. In this embodiment, the content of the keratin softening agent may be 0.05 to 5% by mass.
[0010] In this embodiment, the skin topical preparation may contain, as the plant-derived component, one or more plant-derived components selected from horsetail, Poria cocos, Houttuynia cordata, and Pinellia arvensis.
[0011] In addition, in this embodiment, the skin topical preparation may further contain a fullerene compound and / or a component derived from pomegranate stem cells. In this embodiment, the skin topical agent may be a peeling agent.
[0012] A second aspect of the present invention provides a kit for an external skin preparation comprising the above-mentioned external skin preparation, a first aqueous solution, and a second aqueous solution different from the first aqueous solution.
[0013] In this embodiment, the first aqueous solution is a dissolving agent for dissolving or dispersing the skin external preparation. The first aqueous solution may have a pH of 3 to 4 at 25° C., and preferably contains at least one selected from lactic acid, sodium lactate, citric acid, and sodium citrate, and more preferably contains at least lactic acid and / or sodium lactate.
[0014] In this embodiment, the first aqueous solution may further contain allantoin and / or dipotassium glycyrrhizinate.
[0015] In this embodiment, the second aqueous solution may be a percutaneous absorption enhancer for the topical skin preparation contained in the coating film formed on the skin. In this embodiment, the second aqueous solution may contain at least one selected from glycylglycine, a peptide, and vitamin P. Examples of peptides include human recombinant oligopeptide-1, synthetic human recombinant polypeptide-31, and human recombinant polypeptide-10 (hereinafter referred to as "three peptides"). It is preferable to contain at least one selected from these three peptides, and it is more preferable to contain all of these three peptides. Effect of the Invention
[0016] The present invention makes it possible to provide an external skin preparation and a kit for an external skin preparation that can promote skin turnover and naturally regenerate a new stratum corneum without causing skin troubles such as rash, redness, itchiness, etc. When skin turnover is promoted and a new stratum corneum can be naturally regenerated, the barrier function of the skin is improved, and the skin becomes more elastic, wrinkles and pores become less noticeable, dryness is reduced, inflammation is relieved, and other effects are observed. DETAILED DESCRIPTION OF THE PREFERRED EMBODIMENTS
[0017] An embodiment of the present invention will be described below. First Embodiment The skin external preparation according to the present embodiment comprises a plant-derived component, a keratin softener and silk powder. The above-mentioned white-leaf silkworm, which is generally taken as a Chinese herbal medicine, has the effect of promoting skin turnover by percutaneous penetration, but the inventor's intensive research has found that percutaneous penetration of white-leaf silkworm may cause symptoms such as rash, redness, and itching depending on the user. It has also been found that this symptom is not limited to white-leaf silkworm, but also occurs when dried powder of silkworm is percutaneously penetrated.
[0018] The skin external preparation according to this embodiment is characterized in that it uses silk powder instead of the dried powder of silkworm such as white silkworm. The inventor has confirmed that silk powder can promote skin turnover by percutaneous penetration, and does not cause skin troubles such as rash, redness, and itching. Furthermore, by incorporating silk powder into the skin external preparation according to this embodiment, it is possible to reduce the amount of keratin softener such as sulfur described below while maintaining its action and effect, and it is also possible to suppress side effects such as dermatitis caused by the use of high concentrations of sulfur. Each component contained in the topical skin preparation according to this embodiment will be described below.
[0019] (Silk Powder) Silk powder is a powder made from cocoons, and is made of a polymeric protein called silk fibroin, which is the main component of silk fibers. Examples of the manufacturing method include a method of crushing silk fibers to produce water-insoluble silk powder, a method of dehydrating a recrystallized silk aqueous solution to obtain silk powder, a method of freezing, thawing, separating, and drying a recrystallized silk aqueous solution to obtain silk powder, and a method of hydrolyzing silk fibers (see, for example, JP 2007-006762 A). As described above, silk powder has the effect of promoting skin turnover while suppressing the occurrence of skin troubles such as rashes, redness, and itching, so the skin topical agent according to this embodiment can be used safely by people with some kind of skin disease, such as acne or atopic dermatitis.
[0020] The blending ratio of silk powder can be appropriately set, for example, taking into consideration the effect of promoting skin turnover and the blending ratio of other components described later. In one embodiment, the silk powder is blended in an amount of usually 1% by mass or more, preferably 2% by mass or more, more preferably 3% by mass or more, based on the total mass of the skin external preparation. On the other hand, the silk powder is blended in an amount of usually 20% by mass or less, preferably 15% by mass or less, more preferably 10% by mass or less, particularly preferably 7% by mass or less, based on the total mass of the skin external preparation.
[0021] (keratin softener) The keratin softener is a component that reacts with proteins (such as keratin) on the surface of the skin to soften the keratin and remove damaged keratin. The keratin softening action of the keratin softener is also suitable for acne, which is clogged with keratin masses. The skin topical agent according to this embodiment contains silk powder and can be safely used by people with weak skin, such as those with skin diseases, so it is effective not only as a general peeling agent but also in treating acne. Specific examples of the keratin softener include sulfur, selenium disulfide, salicylic acid, betaines, urea, glycolic acid, lactic acid, trichloroacetic acid, and derivatives and salts thereof, and one or more selected from these can be used.
[0022] (sulfur) Among the specific examples of keratin softening agents described above, sulfur has an antibacterial / germicidal effect and an anti-inflammatory effect in addition to a keratin softening effect. Therefore, it is preferable to use at least sulfur as a keratin softening agent from the viewpoint of expected effects in treating atopic dermatitis and acne, and in treating inflammation. The sulfur used in this embodiment is usually a powdered solid obtained by pulverizing sulfur ore.
[0023] The blending ratio of the keratin softener can be set appropriately. In one embodiment, the keratin softener is blended in an amount of usually 0.05% by mass or more, preferably 0.1% by mass or more, more preferably 1% by mass or more, based on the total mass of the skin external preparation. In this range, the effects of keratin softening, atopy treatment, acne treatment, etc. are easily achieved. On the other hand, the keratin softener is blended in an amount of usually 5% by mass or less, preferably 3% by mass or less, more preferably 2% by mass or less, based on the total mass of the skin external preparation. As described above, as a result of blending silk powder in the skin external preparation according to this embodiment, even if the blending ratio of the keratin softener is reduced to the upper limit value, the effects, that is, the keratin softening effect, anti-inflammatory effect, atopy treatment, acne treatment, etc., can be achieved. This makes it possible to avoid the adverse effects caused by the excessive use of the keratin softener (especially sulfur), specifically the above-mentioned skin troubles caused by increased irritation to the skin.
[0024] (Plant-derived ingredients) The skin topical preparation according to the present embodiment contains one or more plant-derived ingredients. [0046] (How to use) Since the skin topical preparation according to the present embodiment is usually in the form of a solid (powder) at room temperature and humidity, when it is applied to the skin, it is preferable to dissolve or disperse it in an aqueous solution and use it in a liquid form or in a state having a certain viscosity (gel, paste, ointment, etc.) By applying the skin topical preparation according to the present embodiment to the skin in such a liquid form or in a state having a certain viscosity, the turnover of the skin is promoted and a new stratum corneum can be regenerated smoothly.
[0047] Here, the above-mentioned aqueous solution (hereinafter referred to as "first aqueous solution") used for dissolving or dispersing the skin topical preparation according to this embodiment is a solution containing water as a base solvent. The first aqueous solution may contain a solvent other than water, and preferably further contains a solvent that is miscible with water (e.g., glycerin, ethanol, propylene glycol, glycerin, butylene glycol, pentylene glycol, etc.). The water content in the first aqueous solution may be, for example, 70% by mass or more.
[0048] In one embodiment, the first aqueous solution preferably contains lactic acid. Since lactic acid has the effect of softening keratin, the incorporation of lactic acid can promote the penetration of the skin topical agent into the skin. Lactic acid also functions as a pH adjuster.
[0049] In one embodiment, the first aqueous solution preferably contains allantoin. By adding allantoin, moisturizing and anti-inflammatory effects can be improved. In addition, by adding allantoin, the effect of removing old keratin and promoting the generation of new epidermal tissue can be expected.
[0050] In one embodiment, the first aqueous solution preferably contains dipotassium glycyrrhizinate. Dipotassium glycyrrhizinate has excellent anti-inflammatory properties while being less irritating to the skin, so that the anti-inflammatory properties can be safely improved by incorporating dipotassium glycyrrhizinate.
[0051] The first aqueous solution may contain various optional components other than the above-mentioned components. Examples of other components that the first aqueous solution may contain include hydroxypropyl methylcellulose stearoxy ether, potassium hydroxide, PPG-6 decyl tetradeceth-20, lavender oil, sodium lactate, citric acid, sodium citrate, mannitol, water-soluble collagen, human recombinant oligopeptide-1, human recombinant polypeptide-10, etc. Among these, sodium lactate, citric acid, and sodium citrate also function as pH adjusters.
[0052] The blending ratio of each component contained in the first aqueous solution is not particularly limited and can be set appropriately.
[0053] The first aqueous solution is preferably adjusted to a pH value of 3 to 4 at 25° C. from the viewpoints of solubility and dispersibility in the topical skin preparation and permeability of the topical skin preparation into the skin.
[0054] The mixing ratio of the skin external preparation according to this embodiment and the first aqueous solution can be appropriately set. For example, the first aqueous solution is usually 1 mL or more, preferably 2 mL or more, more preferably 2.5 mL or more, and usually 5 mL or less, preferably 4 mL or less, more preferably 3.5 mL or less, per 1 g of the skin external preparation. By setting it in this range, the composition (hereinafter also referred to as "paste") generated by mixing the skin external preparation according to this embodiment and the first aqueous solution can be easily applied to the skin, and the penetration of the skin external preparation into the skin can be promoted.
[0055] The paste is applied to the skin to form a coating film, and time is allowed to penetrate the skin topical agent. During this time, it is preferable to firmly insert the skin topical agent into the coating film by pressing a finger against it. The coating film gradually dries, but in a dry state, the skin topical agent is not absorbed percutaneously and does not penetrate into the skin. Therefore, when the coating film dries, a second aqueous solution is sprayed onto the coating film as a percutaneous absorption promoter for the skin topical agent contained in the coating film, and the second aqueous solution is mixed into the paste. This state is left for a while to further promote the penetration of the skin topical agent into the skin.
[0056] The second aqueous solution is a solution containing water as a base solvent. The second aqueous solution may contain a solvent other than water, and preferably further contains a solvent miscible with water (e.g., butylene glycol, glycerin, pentylene glycol, etc.). The water content in the second aqueous solution may be, for example, 70% by mass or more.
[0057] In one embodiment, the second aqueous solution preferably contains glycylglycine. By adding glycylglycine to the second aqueous solution, it is expected that the effect of improving abnormal keratinization and tightening pores can be achieved.
[0058] In one embodiment, the second aqueous solution preferably contains a peptide. Examples of the peptide include human recombinant oligopeptide-1, synthetic human recombinant polypeptide-31, and human recombinant polypeptide-10 (three peptides). The second aqueous solution preferably contains one or more selected from these three peptides, and more preferably contains all of these three peptides.
[0059] In one embodiment, the second aqueous solution preferably contains vitamin P (glucosyl hesperidin). By incorporating vitamin P, it is expected that the effect of expanding capillaries in the skin and promoting blood flow will be achieved.
[0060] The second aqueous solution may contain various optional components other than the above-mentioned components. Examples of other components that the second aqueous solution may contain include glycosyl trehalose, phenoxyethanol, dihydroxypropyl arginine HCl, hydrogenated starch hydrolysate, PPG-6 decyltetradeceth-20, sodium, dipotassium glycyrrhizinate, citric acid, lavender oil, grapefruit peel oil, comfrey leaf extract, rosemary leaf oil, mannitol, and water-soluble collagen.
[0061] The blending ratio of each component contained in the second aqueous solution is not particularly limited and can be set appropriately.
[0062] After the paste has sufficiently penetrated the skin, it is wiped off. It is then preferable to moisturize the skin with lotion, gel, cream, etc. The treatment time can be set appropriately, but for example, the time from applying the paste to the skin until wiping it off can be about 20 minutes as a guideline.
[0063] <Second embodiment> The kit for an external skin preparation according to this embodiment includes the above-mentioned external skin preparation, the above-mentioned first aqueous solution, and the above-mentioned second aqueous solution that is different from the first aqueous solution. [0064] <Test Example 1> An evaluation test was conducted for skin trouble caused by each of the components shown in Table 1 alone. 0.3 g of the first aqueous solution was added to 0.1 g of each dry powder and mixed to obtain pastes each consisting of a single component. The first aqueous solution used contained lactic acid, allantoin, and dipotassium glycyrrhizinate as main active ingredients, and water, butylene glycol, glycerin, and pentylene glycol as solvents (water content: about 80% by mass). The subjects were 10 women and 3 men aged 19 to 53 who had no skin disease. Each paste was applied to the arm, and the skin troubles were evaluated after 15 minutes. The results are shown in Table 1.
[0065] [Table 1]
[0066] From the results shown in Table 1, it can be seen that, of the 13 subjects, 13 subjects treated with silkworm and 6 subjects treated with sulfur showed symptoms of skin trouble such as rash, redness, and itching. On the other hand, no subjects showed symptoms of skin trouble when using horsetail powder, Poria columbine powder, Houttuynia cordata powder, and Pinellia gracilis powder. From these results, it is speculated that the cause of skin trouble in rare cases in users who used the peeling agent disclosed in Patent Document 1 was the white silkworm and the sulfur blended at a high concentration. In addition, as with the above plant-derived ingredients, no subjects showed symptoms of skin trouble when using silk powder. From these results, it can be seen that silk powder, which has the effect of promoting skin turnover, can be used safely.
[0067] <Example 1> The components shown in Table 2 below were mixed in the ratios shown in the same table to prepare an external skin preparation according to this embodiment.
[0068] [Table 2]
[0069] The subjects were 15 women and 5 men aged 15 to 73 years old who had no skin diseases (different from the subjects in Test Example 1). The skin topical preparation prepared above was applied according to the procedure described below, and skin trouble evaluation and sensory evaluation were performed. The skin trouble evaluation was performed based on whether symptoms such as rash, redness, or itchiness were observed, as in the above-mentioned Test Example 1. The sensory evaluation was a sensory evaluation of the improvement in the skin's barrier function due to the promotion of skin turnover and the natural regeneration of a new stratum corneum, and specifically, was performed by selecting the applicable item from among "skin became more elastic," "wrinkles became less noticeable," "pores became less noticeable," "dryness became less likely," and "inflammation subsided."
[0070] (procedure) (1) 1 g of the skin topical preparation obtained above was placed in a glass bowl, and 3 mL of the first aqueous solution used in Test Example 1 above was added and mixed to obtain a paste-like composition. (2) The paste obtained in (1) is applied to the subject's face to form a coating. (3) Press your finger against the coating to allow the paste to penetrate into the skin. (4) When the coating becomes dry and difficult to slide with your fingers, spray the second aqueous solution described below onto the coating and spread it out while blending the second aqueous solution into the paste with your fingers. (5) Once the paste has fully penetrated the skin (approximately 20 minutes after application), lightly wipe it off with a wet sponge, then wipe it off with a wet, cool towel. (6) Apply lotions, gels and creams to keep skin moisturized. (7) Three days after the first treatment consisting of steps (1) to (6) above, the second treatment will be performed following steps (1) to (6) above.
[0071] The second aqueous solution used was an aqueous solution containing glycylglycine, three types of peptides, and vitamin P (glucosyl hesperidin) as the main active ingredients, and water, butylene glycol, glycerin, and pentylene glycol as the solvent (water content: approximately 80% by mass).
[0072] The skin trouble evaluation and sensory evaluation by the subjects three days after the second treatment are shown in Table 5 below.
[0073] [Table 3]
[0074] From the results shown in Table 5, it was found that the topical skin preparation of this embodiment can promote skin turnover and smoothly regenerate a new stratum corneum without causing skin problems such as rashes, redness, and itching.
[0075] The present invention is not limited to the above-mentioned embodiment, and can be modified in various ways without departing from the gist of the present invention. The embodiments may be combined as appropriate, and in that case, the combined effect can be obtained. Furthermore, the above-mentioned embodiment includes various inventions, and various inventions can be extracted by combinations selected from the multiple components disclosed. For example, if the problem can be solved and the effect can be obtained even if some components are deleted from all the components shown in the embodiment, the configuration from which the components are deleted can be extracted as an invention.
