[0001] CROSS-REFERENCE TO RELATED APPLICATIONS This application claims the benefit of U.S. Provisional Application No. 63 / 050,970, filed July 13, 2020, the entire contents of which are incorporated herein by reference.
[0002] The present disclosure relates generally to pipettors and pipette tips. The present disclosure also relates to methods and devices for installing, using, and removing pipette tips, and for storing pipette tips. [Background technology]
[0003] Pipettors are used in research laboratories and other environments to aspirate and dispense precise volumes of liquid, typically air-displacement pipettors. A wide variety of pipettors are available, each with a variety of liquid handling functions. A pipettor includes a syringe or other aspiration block coupled to a pipette tip. The syringe includes a piston that moves within a barrel. The pipette tip is coupled to the syringe so that a fluid path is established between the barrel and the pipette tip. The piston is driven, either manually or by a motor, to perform alternating forward and reverse strokes within the barrel. The forward stroke can be used to dispense liquid from the pipette tip, while the reverse stroke creates a suction force that can be used to aspirate liquid into the pipette tip. An automated pipetting device may include one or more pipettors, which may be part of a liquid handling apparatus that includes liquid containers with a variety of different liquids.
[0004] Disposable pipette tips have been used in pipettors for decades. Research laboratories may use thousands of pipette tips to aspirate and / or dispense samples and reagents in their various analytical procedures. When a laboratory is unable or unwilling to reuse pipette tips, the pipette tips are discarded after each use. The use of disposable pipette tips increases costs and labor for such laboratories because they must constantly purchase and replenish pipette tips and generate more waste from discarded pipette tips. As laboratories seek to increase the number of samples processed and analyzed, the costs and waste resulting from the disposal of pipette tips can become significant. Furthermore, there are many applications in which disposable pipette tips made of molded plastic do not have sufficient properties for the application. This increases the need to use more expensive, non-disposable tips made from metal, ceramic, etc. Non-disposable tips must be cleaned between handling different samples.
[0005] Pipette tips are available in many different sizes (e.g., 10 μL, 50 μL, 300 μL, 1 mL, 5 mL, etc.) and geometries, including both disposable and non-disposable types. Different procedures or protocols may require different pipette tips (size and / or type), and in some cases, the same procedure or protocol may require the use of different pipette tips. Some existing pipetting devices have the ability to accommodate different pipette tip styles but require the user to manually change pipette tips between automated procedures or protocols. Some existing pipetting devices have the ability to automatically change between disposable and non-disposable tips during a procedure or protocol, but the end of the pipette tip (which will be mated with a syringe) must have the same interface geometry. To accommodate disposable pipette tips sized to hold larger volumes (e.g., 1000 μL), the interface geometry can be designed around these larger tips. In this case, the connection to the pipettor used by smaller-volume disposable tips (e.g., 10 μL) is necessarily the same as that used by much larger tips, resulting in, for example, smaller-volume tips not being optimally designed for their small volume and use or application. For example, the connection to the smaller-volume tip may be much larger than desired. Furthermore, because the connection to the smaller-volume tip must be large (again, so that it can be interchangeable with a larger-volume tip coupled to the same pipettor), the smaller-volume tips cannot be placed next to each other in a grid with the relatively small spacing (e.g., 4.5 mm) required to access adjacent wells in a standard-sized multi-well (e.g., 384-well) plate. Furthermore, various procedures or protocols may require the use of multi-channel and / or single-channel pipetting devices. These two types of pipetting devices often have different mating surfaces, which may not allow the same size or type of pipette tip to be used in both multi-channel and single-channel pipetting devices.
[0006] In general, there has been a long-standing desire to increase the level of automation offered by liquid handling and pipetting devices to increase the throughput of such equipment and the "walk away" time available to users. In particular, there is a need to increase the level of compatibility between different types and sizes of pipette tips, as well as to automate the tasks of coupling pipette tips to syringes, uncoupling pipette tips from syringes, and swapping between different pipette tips in an automated manner. There is a need for an automated, highly reliable method for attaching, detaching, and storing pipette tips for reuse in the research process. Summary of the Invention

[0013] The present teachings are best understood from the following detailed description when read in conjunction with the accompanying drawing figures, the features of which are not necessarily drawn to scale.
[0014] Some embodiments of the present devices and methods provide for easy and quick attachment and detachment of reusable pipettes to pipettors. In some embodiments, among other advantages, pipette tips are held securely in place during use, the seal between the pipette adapter and the pipettor's aspiration block is substantially airtight and liquid-tight, and dead volume is minimized. Some embodiments of the pipetting system allow pipette tips to be removed from a pipette tip storage location and returned to the same location or a different location if desired. In some embodiments, the magnetic attachment mechanism employed in the present devices can accommodate possible errors in the location of pipette tips relative to the pipette tip storage device, including, for example, + / - 0.75 mm in the X and Y directions when the pipette tip is placed in the storage device.
[0015] In one aspect, the novel pipettor comprises an aspiration block and one or more magnets. The magnets are fixed to the aspiration block end of the aspiration block. In some embodiments, the magnets are recessed from the aspiration block face so that they do not physically contact the pipette tip when the pipette tip is magnetically attached to the pipettor. Any suitable magnets can be used, including small but powerful neodymium-iron-boron magnets, and can be positioned within the aspiration block face. The aspiration block may be comprised in whole or in part of stainless steel or other materials and may include tubing or other aspiration channels. The magnets should be strong enough to pick up and securely hold the pipette tip. In some embodiments, the magnets are strong enough to compress a compliant material positioned to form a seal between the aspiration block and the pipette tip.
[0016] In some embodiments, the pipettor is adapted to transfer fluids by pipetting, including, for example, aspirating or dispensing volumes of fluids from 10 μl to 500 μl. The pipettor can also be adapted to mix fluids in a mixing strip, change pipette tips, detect liquid levels, and deliver water or solvent to a slide processing module, humidifier pad, or mixing strip to adjust wettability. In some embodiments, the pipettor includes one or more features adapted to clean the inside of a pipette tip. The pipettor can be connected to one or more, or all, of a processing module, a washing station, a pipette tip storage station, a reagent vial storage, and a gantry.
[0017] The present methods and devices may include a pipettor having a pipette tip. In some embodiments, the pipette tip is a disposable or reusable pipette tip. Reusable pipette tips require cleaning between uses to avoid contamination. It is contemplated that the present methods and devices may optionally be used with disposable pipette tips, thereby reducing the cost and waste associated with replacing pipette tips after a single use.
[0018] FIG. 1A illustrates an exemplary embodiment of a pipettor. The pipettor 100 includes an aspiration block 102 and a piston 104 that controls the movement of fluid into and out of the aspiration block 102. The piston 104 can be actuated by any suitable mechanism. In FIG. 1A, it is actuated by a motor 106 that rotates a lead screw 108, which moves a stage 112, thereby moving the piston 104 up and down. The piston 104 moves up to aspirate fluid into the aspiration block and down to dispense fluid from the pipettor. The pipettor 100 also includes a valve pack 110 to control which fluid is delivered to the aspiration block 102. In some embodiments, the pipettor is configured to deliver at least two types of liquid to the pipette tip. For example, the pipettor can be configured to deliver one or more reagents or wash fluids. The aspiration block can deliver pressurized air or other gas to the interior of the tip. Pressurized gas can be delivered at two different flow rates (low and high) to perform different steps or functions.
[0019] FIG. 1B provides another view of the aspiration block 102. One or more liquids and / or one or more pressurized gases may enter the aspiration block 102 through conduits, valves, or other pathways. In FIG. 1B, a valve pack 110 controls the delivery of reagents, wash fluids, pressurized gases, and other fluids (e.g., solvents and buffers) to the aspiration block 102. As fluids are delivered through the aspiration block 102, they pass through its outlet 114 to an attached pipette tip. The outlet 114 of the aspiration block 102 may include a tapered cone 116 to facilitate pipette tip attachment.
[0020] In some embodiments, pipette tips are attached to the pipettor using one or more magnets 120, e.g., six magnets arranged in a circle, that are affixed to the aspiration block by adhesive or the like. The device may include a seal (e.g., an O-ring seal) between the pipette tip and the nipple of the aspiration block of the pipettor. The tapered cone 116 may have a recess for an O-ring or other seal. A pipette tip sensor 118 may be positioned to detect whether a pipette tip is present or attached to the aspiration block 102 and provide a signal to the controller to enable or stop fluid flow from the pipettor to the outlet 114.
[0021] In some embodiments, the pipettor can be automatically moved along one or more axes (X, Y, and / or Z) by operation of a gantry or other robotic device. Gantries and other devices can be positioned to move the pipettor horizontally and vertically to change pipette tips, such as by moving up and down to separate a first pipette tip and attach a second pipette tip.
[0022] 2 shows the interior of an exemplary embodiment of a pipetting device comprising a pipettor 200 and a reusable pipette tip 201. The pipettor 200 comprises an aspiration block 202 comprising an aspiration channel 222 and an aspiration block end 224, which has an opening (outlet 214) at the aspiration block end 224. The pipettor 200 also comprises one or more magnets 220 fixed within the aspiration block 202 at the aspiration block end 224.
[0023] The pipetting device also includes a reusable pipette tip 201 that includes a pipette adapter 203 and a pipette tube 215. The pipette adapter 203 includes an adapter proximal end 205, an adapter distal end 207, and an adapter side 209. The pipette adapter 203 also includes an adapter lumen 211 that has openings at the adapter proximal end 205 and the adapter distal end 207. The adapter side 209 of the pipette adapter 203 includes an adapter radial groove 213. The pipette adapter 203 includes, for example, is formed from, or contains an amount of, a magnetically attractive material. The pipette tube 215 of the reusable pipette tip 201 includes a pipette tube proximal end 217 and a pipette tube distal end 219. The pipette tube proximal end 215 is secured within the adapter lumen 211, which opens at the adapter distal end 207. The pipette tip 201 is magnetically attached to the pipettor 200 to provide a substantially fluid-tight flow path between the aspiration channel 222 and the pipette tip channel 221 .
[0024] In another aspect, a reusable pipette tip is provided having a pipette adapter including a magnetically attractable material. The pipette adapter further comprises an adapter proximal end, an adapter distal end, an adapter side, and an adapter lumen. In some embodiments, the adapter side comprises an adapter radial groove. The reusable pipette tip also comprises a pipette tube having a pipette tube proximal end and a pipette tube distal end. The pipette tube proximal end is secured within a central lumen opening in the adapter distal end.
[0025] FIG. 3 is a cross-sectional view of a portion of a pipette tip 301 magnetically attached to the aspiration block 302 of a pipettor. The reusable pipette tip 301 includes a pipette adapter 303 with an adapter proximal end 305, an adapter distal end 307, and an adapter side 309. The adapter radial groove 313 can facilitate storage of the pipette tip 301 by providing a feature that can engage with a storage device. A compliant material 323 can facilitate placement of the pipette tip 301 within the storage device, compensating for slight deviations from the expected position of the adapter radial groove 313 upon engagement within the storage device, enabling its versatility. The pipette adapter 303 also includes an adapter lumen 311 with openings at the adapter proximal end 305 and the adapter distal end 307. The adapter side 309 includes the adapter radial groove 313 with the compliant material 323 within the adapter auxiliary groove 325. Pipette adapter 303 includes a magnetically attractive material, and suction block 302 includes a magnet 320 recessed from suction block face 324 facing adapter proximal end 305 .
[0026] In some embodiments, the aspiration block 302 also includes an extension 326 through which the aspiration channel 322 extends, thereby providing an extended outlet. The extension 326 is adapted to fit within an opening 327 of the pipette tip 301, such that the opening 327 of the pipette tip 301 is in fluid communication with the aspiration channel 322. In some embodiments, the extension 326 includes a tapered cone 328 extending from the aspiration block face 324, with the aspiration channel 322 extending through the tapered cone 328.
[0027] In some embodiments, the adapter lumen 311 of the pipette adapter 303 includes an opening at the adapter distal end 307 configured to receive a pipette tube, into which the pipette tube can be secured. The pipette tip 301 is magnetically attached to the aspiration tube 302 to provide a substantially fluid-tight flow path between the aspiration channel 322 and the pipette tube within the adapter lumen 311.
[0028] In some embodiments, the pipette adapter 303 further comprises an adapter cup formed by an adapter proximal end 305 and an adapter cup wall 337 extending from the adapter proximal end 305. The adapter cup wall may surround part of or all of the adapter proximal end 305. The adapter cup wall 337 may also comprise a cup wall guide 339 opposite the adapter proximal end 305. For example, the cup wall guide 339 may be a sloped surface. The adapter cup wall 337 may further comprise an inner surface 341 and one or more radial rings 343 on the inner surface 341 facing toward the center.
[0029] FIG. 4 shows an internal view of another embodiment of a pipette tip 401 magnetically attached to the suction block 402 of a pipettor. The reusable pipette tip 401 includes a pipette adapter 403 having an adapter proximal end 405, an adapter distal end 407, and an adapter side 409. The pipette adapter 403 also includes an adapter lumen 411 with openings at the adapter proximal end 405 and the adapter distal end 407. The adapter side 409 includes an adapter radial groove 413. Unlike the pipette tip 301 of FIG. 3, the pipette adapter 403 does not include a compliant material within the radial groove 413. Such an embodiment would be useful if the storage device could compensate for slight displacement of the pipette tip from its expected position during storage. The pipette adapter 403 includes a magnetically attractable material, and the suction block 402 includes a magnet 420 recessed from the suction block face 424 facing the adapter proximal end 405.
[0030] The aspiration block 402 also includes an extension 426 that extends into an opening 427 in the pipette adapter 403. The extension 426 includes a tapered cone 428 extending from the aspiration block face 424. The opening 427 includes a tapered opening 429 configured to receive the tapered cone 428. The pipette adapter 403 further includes an opening groove 431 at the bottom of the tapered opening 429 and a compliant sealant 433, such as an O-ring, within the opening groove 431. The inclusion of the compliant sealant 433 provides a radial-type seal between the aspiration tube 402 and the pipette adapter 403. The radial seal should be substantially liquid-tight.
[0031] 5 is a cross-sectional view of yet another embodiment of a pipette tip 501 magnetically attached to an aspiration block 502 of a pipettor. The reusable pipette tip 501 includes a pipette adapter 503 having an adapter proximal end 505, an adapter distal end 507, and an adapter side 509. The pipette adapter 503 also includes an adapter lumen 511 having openings at the adapter proximal end 505 and the adapter distal end 507. The pipette tip 501 is magnetically attached to an aspiration tube 502 to provide a substantially fluid-tight flow path between the aspiration channel 522 and the pipette tube within the adapter lumen 511.
[0032] The adapter side 509 includes an adapter radial groove 513 with a compliant material 523 in an adapter auxiliary groove 525. The pipette adapter 503 includes a magnetically attractable material, and the aspiration block 502 includes a magnet 520 recessed from an aspiration block face 524 facing the adapter proximal end 505. The aspiration tube 502 and pipette adapter 503 of FIG. 5 differ from those shown in FIGS. 3 and 4 in that the aspiration tube 502 does not include a tapered cone and the pipette adapter 503 does not include a tapered opening. The end of the aspiration block 502, i.e., the aspiration block face 524, is substantially flat and faces the substantially flat adapter proximal end 505. The magnet 520 is slightly recessed from the aspiration block face 524 to prevent the magnet from physically contacting the pipette adapter 503.
[0033] The adapter proximal end 505 includes an adapter proximal end groove 535 and a compliant seal 533 (e.g., an O-ring) within the adapter proximal end groove 535, which provides a static face seal between the aspiration tube 502 and the pipette adapter 503. In some embodiments, the pipette adapter 503 further includes an adapter cup formed by the adapter proximal end 505 and an adapter cup wall 537 extending from the adapter proximal end 505. The adapter cup wall 537 also includes a cup wall guide 539 on the opposite side of the adapter proximal end 505. The adapter cup wall 537 in FIG. 5 includes two radial rings 543 on its inner surface 541 that engage the aspiration block 502. In this embodiment, tightly toleranced radial rings 543 are used at the top and bottom of the adapter cup to align the pipette adapter 503. Two radial rings 543, which may be spaced apart by, for example, about 15 mm, are advantageous for controlling concentricity and tilt.
[0034] 6 shows a cross-sectional view of yet another embodiment of a pipette tip 601 magnetically attached to an aspiration block 602 of a pipettor. The reusable pipette tip 601 includes a pipette adapter 603 including an adapter proximal end 605, an adapter distal end 607, and an adapter side 609. The adapter side 609 includes an adapter radial groove 613 having a compliant material 623. The pipette adapter 603 includes a magnetically attractive material, and the aspiration block 602 includes a magnet 620 recessed from an aspiration block face 624 facing the adapter proximal end 605. The adapter proximal end 605 includes a compliant seal 633 (e.g., an O-ring) in an adapter proximal end groove 635.
[0035] The pipette adapter 603 of Figure 6 further includes an adapter cup formed by an adapter proximal end 605 and an adapter cup wall 637 extending from the adapter proximal end 605. The pipette adapter 603 of Figure 6 differs from that shown in Figure 5 in that the adapter cup wall 637 includes only one radial ring 643 and the adapter cup wall 637 has a relatively low height. The adapter proximal end 605 also includes rounded or chamfered edges.
[0036] 7 is a cross-sectional view of an exemplary embodiment of a pipette tip 701 magnetically attached to an aspiration block 702 of a pipettor. The reusable pipette tip 701 includes a pipette adapter 703 including an adapter proximal end 705, an adapter distal end 707, and an adapter side 709. The pipette adapter 703 also includes an adapter lumen 711 having openings at the adapter proximal end 705 and the adapter distal end 707. The adapter side 709 includes an adapter radial groove 713, which may or may not include a compliant material and an adapter auxiliary groove. The pipette adapter 703 includes a magnetically attracting material, and the aspiration block 702 includes a magnet 720 recessed from an aspiration block face 724 facing the adapter proximal end 705.
[0037] In some embodiments, the aspiration block 702 also includes an extension 726 through which the aspiration channel 722 extends. The extension 726 is adapted to fit within an opening 727 of the pipette tip 701 such that the opening 727 of the pipette tip 701 is in fluid communication with the aspiration channel 722. In some embodiments, the extension 726 includes a tapered cone 728 extending from the aspiration block face 724, the opening 727 including a tapered opening 729, and the aspiration channel 722 extending through the tapered cone 728.
[0038] In some embodiments, the adapter lumen 711 of the pipette adapter 703 includes an opening at the adapter distal end 707 configured to receive a pipette tube 715, which can be secured therein. The pipette tip 701 is magnetically attached to the aspiration tube 702 to provide a substantially fluid-tight flow path between the aspiration channel 722 and the pipette tube 715 within the adapter lumen 711.
[0039] In some embodiments, the pipette adapter 703 further comprises an adapter cup formed by an adapter proximal end 705 and an adapter cup wall 737 extending from the adapter proximal end 705. The adapter cup wall 737 does not surround or enclose the suction block face 724. Instead, the adapter proximal end 705 has a size equal to or smaller than the outer periphery of the suction block face 724. For example, the adapter proximal end 705 and the suction block face 724 each have an outer periphery, e.g., a circumference, and the outer periphery of the adapter proximal end 705 is equal to or smaller than the outer periphery of the suction block face 724. The adapter cup wall 737 may also comprise a cup wall top 745 that faces the suction block face 724.
[0040] In another aspect, a method for using a reusable pipette tip with a pipettor is provided. The method may include providing a pipettor having an aspiration block with an aspiration channel and an aspiration block end. The aspiration channel has an opening at the aspiration block end. The aspiration block also includes one or more magnets secured within the aspiration block at the aspiration block end. The method may also include positioning the aspiration block end sufficiently close to the reusable pipette tip to form a magnetic attachment. The reusable pipette tip includes a pipette adapter having a magnetically attractable material, an adapter proximal end, an adapter distal end, and an adapter lumen. The reusable pipette tip also includes a pipette tube having a pipette tube proximal end and a pipette tube distal end. The pipette tube proximal end is secured within a central lumen opening at the adapter distal end. The pipette tip is magnetically attached to the pipettor to provide a substantially fluid-tight flow path between the aspiration channel and the pipette tip channel.
[0041] In some embodiments, where the pipette adapter further comprises adapter sides, the adapter sides comprising adapter radial grooves, the method may also include moving the pipettor and the magnetically attached pipette tip to a storage device adapted to receive the pipette adapter and transferring the pipette adapter to the storage device. In some embodiments, where the storage device comprises an empty storage fork having storage fork prongs separated by a storage fork mouth, the pipettor is moved such that the magnetically attached pipette tip is inserted into the storage fork mouth in a direction substantially parallel to the storage fork prongs. The method may also include moving the pipettor away from the engaged pipette in a direction substantially perpendicular to the storage fork prongs, thereby overcoming the magnetic attraction between the magnet in the aspiration body and the pipette adapter while the storage fork prongs prevent the inserted adapter from disengaging from the storage fork.
[0042] In some embodiments, the storage device further includes a second storage fork that holds a second pipette tip. The method further includes moving the pipettor so that the aspiration channel is positioned above the second pipette tip held by the storage fork, moving the pipettor toward the second pipette tip to magnetically attach the pipettor to the second pipette tip, and moving the pipettor in a direction substantially parallel to the storage fork blades, thereby removing the second pipette tip from the storage device. The method may also include detecting whether the second pipette tip is attached to the aspiration channel before removing the second pipette tip from the storage device.
[0043] The present methods and apparatus may further include, or be part of, a method or system for preparing a sample for analysis. Such a system may include other devices for performing other functions. For example, the pipettor may be mounted on a gantry for automating movement between various locations (e.g., between a pipette tip storage location and a location for pipette use (e.g., a sample processing location, a reagent aspiration location, etc.)). In some embodiments, the present methods and apparatus are included in a liquid handling apparatus that further includes one or more of a reagent source, a sample processing module, a pipette washing device, or other devices.
[0044] 8A and 8B show a pipetting system 800 including a pipette tip storage device 801 configured to hold one or more pipette tips. The storage device 800 includes multiple storage forks 802, 804 configured to engage and hold pipette adapters 803, 805, 807. In some embodiments, the storage forks 802, 804 are configured to receive the pipette adapters 803, 805 in their adapter radial grooves. The storage forks 802, 804 are mounted on a storage block 806. The pipette adapter 807 is magnetically attached at an aspiration block connection 808, which is held by an aspiration mounting block 810 on a base 812 of the system 800. The system 800 may also include a gantry 814 that can move the aspiration mounting block 810 to the pipette tip storage device 801 and / or to other devices or locations for pipetting use in the system 800 (e.g., sample processing locations or devices, reagent aspiration locations or devices, etc.).
[0045] 8B provides a close-up view of storage forks 802, 804 for holding one or more pipette tips. The storage forks 802, 804 are configured to engage and hold the adapter radial grooves of a pipette adapter for reusable pipette tips. Each storage fork includes storage fork tines separated by a storage fork mouth, and a storage fork may include two sets of storage forks and storage fork mouths. For example, storage fork 802 includes a pair of storage fork tines 820, 822 separated by storage fork mouth 821 and another pair of storage fork tines 830, 832 separated by storage fork mouth 831. The storage fork tines 830, 832 are configured to receive the adapter radial groove 813 into the storage fork mouth 831 in a direction substantially parallel to the storage fork tines 830, 832 and to prevent the inserted pipette adapter 803 from disengaging from the storage fork 802 in a direction substantially perpendicular to the storage fork tines 830, 832. The storage fork mouth 831 is wider at the adapter radial groove 813 than the pipette adapter and narrower than the adapter side 809 adjacent to the adapter radial groove 813.
[0046] The storage fork can be fixedly or pivotally mounted on the storage block. For example, the storage fork 802 is fixedly mounted on the storage block 806 by a fixture 836. The position of the pipette adapter 803 can be established and compensated for within the rigid storage fork mouth 831 by an O-ring in the adapter radial groove 813. The installation diameter of the pipette adapter is smaller than the outer diameter of the O-ring. As another example, the storage fork 804 is pivotally mounted on the storage block 806 by a single fixture 834 in the center of the storage fork 804. The pipette adapter 805 extends into the storage fork 804, but there is no O-ring and its installation diameter is a tight fit with the fork. The pivoting ability of the storage fork 804 allows the system to accommodate misalignment of the pipette tip in the X or Y direction. The storage fork 804 includes a magnet 837 for attracting the pipette adapter 805 into the storage fork 804. The storage fork 804 also includes a spring plunger 838 to resist the horizontal magnet 837 and hold the pipette adapter 804 in its nominal position. In some embodiments, the storage device includes one or more magnets aligned with opposite poles of the magnet in the storage fork.
