Description

Composition for skin soothing or anti-inflammatory comprising a metal-phenol complex and melanin or a precursor thereof

[0001] It relates to a composition for soothing or anti-inflammatory skin.

[0002] When external stimuli occur on the skin, the amount of reactive oxygen species in the skin increases. Natural products with antioxidant properties initially lower the concentration of reactive oxygen species, thereby alleviating external stimuli. Even if antioxidants successfully lower the concentration of reactive oxygen species, continued stimulation can lead to damage to the skin barrier. When the stratum corneum is damaged, abnormalities in the filaggrin gene reduce the production of natural moisturizing factors and alter the lipid composition of the stratum corneum. This reduces the moisture content of the stratum corneum, and increases moisture loss, leading to dryness and itching, which induce an inflammatory response. When inflammation occurs on the skin, it can lead to skin diseases such as papules, telangiectasia, erythema, and atopy.

[0003] Therefore, in order to solve the above problems, it is necessary to develop raw materials that have anti-inflammatory effects and affect skin soothing.

[0004] One aspect is to provide a composition comprising a metal-phenolic complex (MPC) and melanin or a precursor thereof as active ingredients.

[0005] Another aspect provides the use of a composition comprising a metal-phenolic complex (MPC) for preparing a medicament for preventing, improving or treating inflammation; and melanin or a precursor thereof as an active ingredient.

[0006] Another aspect provides a method of preventing, ameliorating or treating inflammation in a subject comprising the step of treating or administering to a subject in need thereof an effective amount of the composition.

[0007] One aspect provides a composition comprising a metal-phenolic complex (MPC) and melanin or a precursor thereof as active ingredients.

[0008] In one embodiment, a metal-phenol complex, melanin, or its precursor was demonstrated to be non-cytotoxic to human keratinocytes and exhibited anti-inflammatory effects by reducing the expression of inflammatory cytokines. Therefore, the composition may be used for the prevention, improvement, or treatment of inflammation, and may be used as a soothing agent to alleviate the symptoms of skin inflammation.

[0009] The term "metal-phenolic complex (MPC)" used herein refers to a complex formed by the interaction of a metal ion and a phenol compound under specific conditions, and stabilized by electronic interactions between the positive charge of the metal ion and the negative charge of the phenol. The metal may include a transition metal or an alkali metal. The transition metal may be, for example, iron (Fe), copper (Cu), silver (Ag), zinc (Zn), platinum (Pt), gold (Au), etc., and the alkali metal may be, for example, lithium (Li), sodium (Na), potassium (K), rubidium (Rb), cesium (Cs), barium (Ba), calcium (Ca), nickel (Ni), cobalt (Co), manganese (Mn), magnesium (Mg), etc. In addition, the phenol compound may be at least one selected from the group consisting of tannic acid, gallic acid, flavones, anthocyanins, polyphenols, isoflavones, catechins, methyl gallate, ethyl gallate, propyl gallate, octyl gallate, lauryl gallate, epicatechin gallate, epigallocatechin gallate, gallocatechin gallate, and theaflavin-3-gallate, or a derivative thereof.

[0010] In one specific example, the metal-phenol complex may be a mixture of iron(II) D-gluconate hydrate and tannic acid and gallic acid.

[0011] In one specific example, the metal-phenol complex may be formed by mixing iron(II) D-gluconate hydrate, gallic acid, and tannic acid in a weight ratio of 1:0.01 to 10:0.01 to 5. The iron(II) D-gluconate hydrate, gallic acid, and tannic acid may be mixed in a concentration ratio of, for example, 1:0.01 to 10:0.01 to 5, 1:0.01 to 10:0.01 to 3, 1:0.01 to 5:0.01 to 0.5, 1:0.1 to 5:0.05 to 3, or 1:0.1 to 2.5:0.05 to 1.5. At this time, if the mixing ratio of the D-gluconate iron (II) hydrate and gallic acid and tannic acid is below the above range, there is a problem that the metal-phenol complex is not sufficiently formed, and thus the synergistic effect of the cosmetic composition cannot be sufficiently exerted. If it exceeds the above range, there is a problem that the particle size of the metal-phenol complex increases and the particles precipitate.

[0012] As used herein, the term "melanin" refers to a general term for black or brown pigments present in tissues such as skin, hair, and eyes, and "melanin precursor" plays a role in the biosynthesis of melanin, and its amount determines skin color, body hair color including hair, eye color, etc. Accordingly, the composition can be used not only to prevent or improve graying by promoting melanin production, but also to dye hair, eyelashes, or skin.

[0013] In one specific example, the melanin precursor may be poly-dihydroxyindole. Poly-dihydroxyindole is an indole-based compound found in plant and animal tissues, and primarily plays a role in the biosynthesis of melanin, a skin pigment. In particular, poly-dihydroxyindole has an antioxidant effect, which can help protect the skin from oxidation and environmental damage. Therefore, the composition according to one aspect can exhibit a natural dyeing effect on the skin or hair without irritating the skin, and thus can be used as a dye for dyeing hair, eyebrows, eyelashes, skin, nails, etc. In addition, it can strengthen hair roots and prevent, improve, or treat hair loss, and thus can be used for strengthening hair roots and preventing, improving, or treating hair loss.

[0014] The composition may further comprise a plant extract having skin soothing or anti-inflammatory activity. The plant extract may be, for example, pomegranate extract, green tea extract, blueberry extract, cacao extract, apricot extract, persimmon extract, grape extract, avocado extract, apple extract, beet extract, or mulberry extract.

[0015] In one specific example, the plant extract may be a pomegranate extract.

[0016] The above pomegranate extract is obtained by extracting a pomegranate tree with water, a lower alcohol having C1 to C4 content, or a mixture thereof, and may be obtained by extracting flowers, fruits, seeds, leaves, branches, stems, or a mixture thereof of the pomegranate tree. When extracting, the dried pomegranate tree may be cut into pieces, and then water, alcohol, or a mixture thereof may be added in an amount of 2 to 20 times the weight of the pomegranate tree to perform the extraction. The extraction may be performed at a temperature of 20 to 100°C for 1 to 10 hours. Any of cold steeping, ultrasonic extraction, or reflux cooling extraction methods may be used as the extraction method, and the extraction may be repeated 1 to 5 times.

[0017] In another specific example, the pomegranate extract may be a butylene glycol fraction obtained by extracting a pomegranate tree with water, a lower alcohol having C1 to C4, or a mixture thereof, and then refining the fraction with butylene glycol. The lower alcohol used for extracting the pomegranate tree may be ethanol or methanol. During extraction, the dried pomegranate tree may be cut into pieces, and then water, alcohol, or a mixture thereof may be added in an amount of 2 to 20 times the weight of the pomegranate tree to perform the extraction. The extraction temperature may be 30°C to 100°C. The extraction time may be 1 to 10 hours. The extraction method may utilize a cold steeping, ultrasonic extraction, or reflux cooling extraction method. The number of extractions may be 1 to 5 repeated extractions.

[0018] In another specific example, the pomegranate extract may contain an active compound. The active compound may be ellagic acid or a derivative thereof. The ellagic acid derivative may be a sodium salt, calcium salt, or amine salt derivative of ellagic acid, an ellagic acid derivative having a saturated or unsaturated acyl group having 2 to 22 carbon atoms bonded to it, or a form in which glucose is mono- or poly-ether bonded to one or more hydroxyl groups of ellagic acid. In addition, the ellagic acid or a derivative thereof may be contained in an amount of 400 ppm or more per 100 g of the pomegranate extract.

[0019] The composition may be a mixture of a metal-phenol complex, melanin or a precursor thereof, and a plant extract in a concentration ratio of 1:0.001 to 5:0.001 to 5. The metal-phenol complex, melanin or a precursor thereof, and a plant extract may be mixed in a concentration ratio of, for example, 1:0.001 to 5:0.001 to 5, 1:0.001 to 4.5:0.001 to 2.5, 1:0.01 to 4:0.005 to 2, 1:0.1 to 3:0.05 to 1.5, or 1:0.5 to 1:0.25 to 0.5. At this time, if the mixing ratio of the metal-phenol complex, melanin or its precursor and plant extract is below the above range, there is a problem that the synergistic effect of skin soothing or anti-inflammation cannot be sufficiently exerted, and if it exceeds the above range, there is a problem that the stability of the formulation is reduced because a precipitation phenomenon occurs.

[0020] The composition is comprised of 0.001 wt% to 80 wt%, for example, 0.01 wt% to 60 wt%, 0.01 wt% to 40 wt%, 0.01 wt% to 30 wt%, 0.01 wt% to 20 wt%, 0.01 wt% to 10 wt%, 0.01 wt% to 5 wt%, 0.05 wt% to 60 wt%, 0.05 wt% to 40 wt%, 0.05 wt% to 30 wt%, 0.05 wt% to 20 wt%, 0.05 wt% to 10 wt%, 0.05 wt% to 5 wt%, 0.1 wt% to 60 wt%, 0.1 wt% to 40 wt%, 0.1 wt% to 30 wt%, 0.1 wt% to 20 It may include a metal-phenol complex and melanin or a precursor thereof in an amount of 0.1 wt% to 10 wt%, or 0.1 wt% to 5 wt%. In this case, if the content of the metal-phenol complex and melanin or a precursor thereof is less than the above range, there is a problem in that the skin soothing or anti-inflammatory effect is not sufficiently exerted.

[0021] In this specification, the term "including as an active ingredient" means that the metal-phenol complex of this specification and melanin or a precursor thereof are added to an extent that the above-mentioned effect can be exhibited, and includes the formulation in various forms by adding various components as auxiliary components for drug delivery and stabilization, etc.

[0055]

[0056] [Example]

[0057] Examples 1-4. Preparation of compositions

[0058] A composition comprising a metal-phenolic complex (MPC), poly-dihydroxyindole, and / or pomegranate extract was prepared. First, iron(II) D-gluconate dehydrate (699014-53-4), gallic acid, and tannic acid were mixed in a ratio of 1:2:1, and then freeze-dried to prepare a metal-phenolic complex.

[0059] Next, a pomegranate extract with increased ellagic acid content was prepared. Specifically, 150 g of crushed raw powder of the whole pomegranate tree containing more than 90 wt% of pomegranate flowers was extracted with a 70% ethanol aqueous solution at 60℃ for 5 hours. Afterwards, the powder was filtered using a 250 mesh, 5 ㎛ filter and concentrated until the amount of raw powder became 20 wt% of the initial amount. The pomegranate ethanol extract was prepared by diluting the ellagic acid content to 400 ppm or more using 1,3-butylene glycol.

[0060] Thereafter, a composition was prepared by mixing the metal-phenol complex, poly-dihydroxyindole (PHI) (CM Tech Co., Ltd.) and pomegranate extract to include the concentration ratio (ppm) of Table 1 below.

[0074] Cytotoxicity confirmation

[0075] In order to confirm the cytotoxicity of the composition according to the aspect, the cell viability of the main components constituting the composition was confirmed. Specifically, human keratinocyte HaCaT cells were cultured in a 6-well plate containing Dulbecco's modified Eagle's medium (DMEM) medium supplemented with 10% fetal bovine serum (FBS) and 1% penicillin. Then, the cells were seeded in a 96-well plate at 1 Υ 10 4 After dividing into cells / well and culturing for an additional 24 hours, the cells were washed with PBS (Phosphate Buffered Saline) and cultured for 24 hours in a starvation state in a medium not containing FBS (Fetal Bovine Serum). After removing the medium, the cells were cultured for 24 hours in a new medium not containing FBS, and then further cultured by adding Comparative Examples 1 to 3 at 0.01, 0.1, 1, and 10 ppm, respectively. Afterwards, 10 μL of 0.5% MTT (3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl-tetrazolium bromide) was added to the cultured cells and cultured for 4 hours. The culture medium was removed, and 100 μL of DMSO (Dimethyl Sulfoxide) solution was added to dissolve the produced MTT formazan. Absorbance was measured at 540 nm using a microplate spectrophotometer (BioTek, USA).

[0076] Figure 1 shows the results of confirming the cytotoxicity of the composition according to the daily aspect.

[0077] As a result, as shown in Fig. 1, it was confirmed that Comparative Examples 1 and 2 had higher cell viability than the untreated control group regardless of concentration. On the other hand, it was confirmed that Comparative Example 3 had a concentration-dependent decrease in cell viability.

[0078] Therefore, it can be seen that the composition according to one aspect is non-cytotoxic and harmless to the human body by including a metal-phenol complex, poly-dihydroxyindole and a pomegranate extract at a specific concentration.

[0079]

[0080] Anti-inflammatory effect confirmed

[0081] The anti-inflammatory activity of the composition according to the aspect was confirmed. Specifically, human keratinocyte HaCaT cells were cultured in a 6-well plate containing Dulbecco's modified Eagle's medium (DMEM) medium supplemented with 10% fetal bovine serum (FBS) and 1% penicillin. 10 μg / ml of Poly I:C, an immunostimulant, and 10 ng / ml of IL-4 protein were added to the culture medium, and dexamethasone, which has an anti-inflammatory effect, was used at a concentration of 1 μM as a positive control. Examples 1 to 4 and Comparative Examples 1 to 3 (0.01, 0.1, 1, and 10 ppm) were added to the cell culture medium, respectively. The culture medium containing the samples was cultured for 4 hours in an incubator maintained at 37°C, 5% CO2 concentration, and 95% humidity. Afterwards, the cultured cells were harvested, washed with chilled phosphate buffered saline (PBS), and total RNA was extracted using the PureLink® RNA Mini Kit (Thermo Fisher Scientific). The specific RNA extraction was performed according to the manual provided with the product. Gene expression changes were measured using quantitative real-time PCR (qRT-PCR), which conjugates a fluorescent material to the DNA product amplified in the polymerase chain reaction (PCR) and continuously detects the fluorescent material. To quantify the expression of the inflammatory response factors IL-1β, IL-6, and TSLP genes, the fluorescence value emitted by the PCR products was measured using SYBR Green (Thermo Fisher Scientific). The reaction solution was prepared by mixing 0.2 μM primers, 50 mM KCl, 20 mM Tris / HCl pH 8.4, 0.8 mM dNTP, 0.5 U Extaq DNA polymerase, 3 mM MgCl2, and 1XSYBR Green in a PCR tube. After primary denaturation at 94°C for 3 minutes using a PCR machine, denaturation, annealing, and polymerization were performed for a total of 40 cycles: 94°C for 30 seconds, 58°C for 30 seconds, and 72°C for 30 seconds. The fluorescence intensity was measured after each cycle. The fluorescence intensity of the IL-6, IL-1β, and TSLP genes was standardized to the actin value, converted to 100% of the control group, and the differences were compared to measure the expression levels of each gene, and the results are shown in Figures 2 to 4.

[0082] Figure 2 shows the effect of a composition according to one specific example on the expression of IL-6.

[0083] Figure 3 shows the effect of a composition according to one specific example on the expression of IL-1β.

[0084] Figure 4 shows the effect of a composition according to one specific example on the expression of TSLP.

[0085] As a result, as shown in FIGS. 2 to 4, it was confirmed that Examples 1 to 4 significantly reduced the expression of IL-6, IL-1β, and TSLP in human keratinocytes compared to Comparative Examples 1 to 3.

[0086] Therefore, the composition according to the aspect has a synergistic effect in reducing the expression of inflammatory cytokines, and is effective in soothing the skin or preventing, improving or treating skin inflammation.

[0087]
