Abstract
Description
[0001] This application is a divisional application. The application number of the original application is 201980047555.2, the application date is June 5, 2019, and the name of the invention is "Cosmetic composition for enhancing keratin exfoliation". Technical Field
[0002] This application claims the benefit of priority based on Korean Application No. 10-2018-0083069 filed on Jul. 17, 2018 and Korean Application No. 10-2019-0063704 filed on May 30, 2019, and all contents disclosed in the specifications and drawings of those applications are incorporated herein by reference.
[0003] The invention relates to the use of the composition in the manufacture of cosmetics and a method for enhancing keratin exfoliation. Background Art
[0004] The stratum corneum is the outermost layer present in the skin, mainly composed of keratinocytes and the lipid layer of the lamellar structure comprising ceramide. The stratum corneum, while showing the effect of protection from external substances, also plays the role of the defense membrane that can prevent internal moisture from detaching, etc. The normal stratum corneum has a thickness of about 15-20 layers, and the decomposition of desmosomes is carried out by the action of the serine protease activated from the stratum corneum exterior, and the process of finally being completely detached from the skin can be experienced. For normal cells, this process needs about 15-20 days. Even if it is not a hereditary disease such as ichthyosis (ichthyosis), the time of the above-mentioned keratin detachment process of the skin of aging, dryness and acne, etc. is also longer than the normal time, therefore demonstrating the keratin multilayer phenomenon of stratum corneum thickening.
[0005] It is known that keratin multilayering is generally caused by factors such as reduced skin moisturizing ability, reduced expression or activity of serine proteases, ultraviolet rays, reduced cell activity, etc. In order to eliminate the generated keratin multilayering, methods such as keratin peeling by physical scrub preparations, keratin peeling by chemical preparations, and increasing physiological cell activity are generally used. The purpose of this type of keratin peeling method is to artificially remove the outer keratinocytes and eliminate the keratin multilayering by replacing the outermost keratinocytes with new keratinocytes from the inside, thereby solving appearance problems such as rough or dull skin.
[0006] When the stratum corneum is eliminated by enhancing the exfoliation of the stratum corneum, additional effects such as removing fine lines, removing blemishes, and improving rough skin can be obtained. Therefore, it is applied in a variety of products, and in particular, alpha hydroxyacid (AHA), beta hydroxy acid (BHA) or poly hydroxy acid (PHA) are typically used. However, the mode of action of these AHA, BHA, and PHA is to insert into the stratum corneum and release hydrogen ions to weaken the binding of desmosomes, thereby achieving stratum corneum exfoliation. Since the insertion into the stratum corneum is smooth and the degree of hydrogen ion transfer is high when applied at low pH, low pH is inevitably required. Therefore, since low pH can cause side effects such as skin burning, itching, and erythema, research is being conducted to overcome these side effects. However, in order to alleviate the side effects caused by such skin irritation, when adjusted to a weakly acidic to neutral pH, its activity is reduced by more than 80%, and when chemically deformed, its efficacy is offset, so there is a problem that it is substantially difficult to provide the benefits of keratin exfoliation. Therefore, a new keratin exfoliant is needed that can show the same keratin exfoliation efficacy as AHA under acidic conditions even under weakly acidic or neutral conditions.
[0007] Therefore, the inventors of the present invention have confirmed in the applied Korean Patent No. 10-2017-0165268 in the prior study that a composition containing serine and carnitine shows efficacy higher than that of AHA at the same concentration under neutral or weakly acidic conditions, and also confirmed that the irritation is relatively small or non-irritating, thus providing an excellent new exfoliating material. Summary of the invention
[0008] Technical issues
[0009] Therefore, the problem to be solved by the present invention is to provide a cosmetic whose composition is selected to maintain the accessibility of the keratin exfoliating agent to the keratin-binding protein, thereby fully exerting the additional keratin exfoliation enhancing effect without cosmetic irritation.
[0010] Another problem to be solved by the present invention is to solve the problem that the exfoliation effect is reduced when serine or carnitine, which has a superior effect than existing exfoliation materials at a weakly acidic to neutral pH, is actually used in a cosmetic formulation.

[0034] In order to achieve the above object, the present invention provides a cosmetic composition for enhancing skin keratin exfoliation, which comprises a keratin exfoliant and does not contain a polyol moisturizer or contains a polyol moisturizer in an amount of 10 wt % or less relative to the total weight of the composition. The present invention is described in detail with reference to the following drawings.
[0035] The inventors of the present invention attempted to produce commercially available cosmetics imparting a keratin exfoliation enhancing function by adding a keratin exfoliating agent to a cosmetic composition containing various additives such as a surfactant, a fragrance, a preservative, a moisturizer, a thickener, an antioxidant, etc., but confirmed that there was a problem in which the efficiency was significantly lower than the keratin exfoliation enhancing ability expected from the concentration of the keratin exfoliating agent. After conducting studies on this, the inventors found that the reason for the reduced keratin exfoliation function was that the moisturizing agent components in various cosmetic additives, especially the moisturizing agent having a polyol group, reduced the accessibility of the keratin exfoliating material to the keratin-binding protein, and as a result, it was unable to affect the keratin-binding protein, thereby reducing the keratin exfoliation ability. The inventors of the present invention proposed a solution to this problem, thereby completing the present invention.
[0036] The present invention provides a cosmetic composition having a composition that allows a keratin exfoliating agent, particularly serine or carnitine, to maintain its effect. More specifically, the present invention aims to provide a cosmetic containing serine or carnitine that imparts a keratin exfoliating effect superior to that of existing functional raw materials without irritation at a weakly acidic to neutral pH.
[0037] However, it has been confirmed that the keratin peeling effect of serine or carnitine is reduced when it is applied to a dosage form. As has been confirmed in the present invention, if a moisturizer having a polyol group such as glycerin, dipropylene glycol (DPG) and 1,3-butylene glycol (BG) is contained in serine or carnitine at a specific concentration or more, the keratin peeling effect is sharply reduced. Therefore, the present invention provides a polyol moisturizing agent component in serine or carnitine that does not substantially contain a polyol moisturizing agent component or provides a polyol moisturizing agent component within a range that does not significantly reduce its effect.
[0038] In one embodiment, the present invention provides a cosmetic composition for enhancing skin keratin exfoliation, comprising a keratin exfoliant and substantially free of a polyol moisturizer.
[0039] The term "substantially free of" as used in this specification means containing 2 wt% or less, preferably 1 wt% or less, more preferably 0.5 wt% or less, more preferably 0.3 wt% or less, further more preferably 0.1 wt% or less, and most preferably completely free of, relative to the total weight of the composition.
[0040] On the other hand, a humectant having a polyol group is a raw material widely used in cosmetic compositions like purified water. In particular, in the case of keratin exfoliating products, moisturizing is more required than usual. From this point of view, it is a problem to provide a cosmetic composition for enhancing keratin exfoliation containing a polyol humectant.
[0041] Therefore, in another embodiment, the present invention provides a cosmetic composition for enhancing skin keratin exfoliation, which comprises a polyhydric alcohol moisturizer and a keratin exfoliant. As described above, when the composition of the present invention comprises a polyhydric alcohol moisturizer, its content exceeds 0 and is less than 10% by weight, preferably exceeds 0 and is less than 2% by weight, and more preferably exceeds 0 and is less than 0.5% by weight, and when the content of the polyhydric alcohol moisturizer exceeds the above numerical range, the keratin exfoliation effect is significantly reduced, and the above content range has a critical significance.
[0042] The term "keratin peeling" used in this specification means that keratin deposited on the stratum corneum of the skin falls off or is removed from the stratum corneum of the skin. This concept includes not only the case of removal by applying physical force, but also the case of being able to be separated from the stratum corneum of the skin by only treating with the cosmetic composition without applying additional force.
[0043] According to one aspect of the present invention, the keratin exfoliating agent of the keratin exfoliation enhancing cosmetic composition of the present invention may be at least one of serine and carnitine.
[0044] According to one aspect of the present invention, the keratin peeling enhancing cosmetic composition of the present invention may contain 0.001 to 20% by weight, preferably 0.01 to 17% by weight, and more preferably 0.05 to 15% by weight, relative to the total weight of the composition. If the content is less than 0.001% by weight, it may be difficult to obtain the desired effect, and if it exceeds 20% by weight, the effect of the increase in content may be insufficient.
[0045] According to one aspect of the present invention, the polyol moisturizer of the keratin exfoliation enhancing cosmetic composition of the present invention may be one or more selected from the group consisting of glycerol (Glycerine), dipropylene glycol (Dipropylene glycol, DPG), butylene glycol (Butylene Glycol, BG), glyceryl polyether -26 (Glycereth-26), methyl glucose polyether -20 (methyl gluceth-20) and pentylene glycol (Pentylene glycol), specifically, it may be one or more selected from the group consisting of glycerol, dipropylene glycol, 1,3-butylene glycol.
[0046] According to one aspect of the present invention, the polyol polymer of the keratin exfoliation enhancing cosmetic composition of the present invention is not contained or may be contained at 10 wt% or less, preferably 5 wt% or less, more preferably 2 wt% or less, more preferably 1 wt% or less, more preferably 0.5 wt% or less, more preferably 0.3 wt% or less, and further more preferably 0.1 wt% or less relative to the total weight of the composition. If the content is greater than 10 wt%, it may be difficult to obtain a sufficient keratin exfoliation effect of the desired keratin exfoliating agent.
[0047] According to one aspect of the present invention, the pH of the keratin exfoliation enhancing cosmetic composition of the present invention may be 5 to 8, preferably 5 to 7.5, and more preferably 5.5 to 7.
[0048] The keratin peeling enhancing cosmetic composition of the present invention may be prepared by arbitrarily selecting components generally contained in cosmetics in addition to the above-mentioned components according to the dosage form or purpose of use of the cosmetic. In addition, the composition of each dosage form may contain various appropriate bases and additives required for the formulation of the dosage form, and may be manufactured by containing known compounds such as preservatives, thickeners, surfactants, silicone polymers, oxidation stabilizers, organic solvents, antioxidants, emollients, pigments, and fragrances within a range that does not reduce the effect.
[0049] Through the screening method of the pig skin keratin exfoliation evaluation method and the human body experiment method of the DHA staining method, it was confirmed that the cosmetic composition according to the present invention can also show an excellent keratin removal effect in a dosage form containing a moisturizing agent containing a polyhydric alcohol.
[0050] Specifically, through the pig skin keratin exfoliation evaluation method, it was confirmed that in the neutral or weakly acidic pH range of 5-7, the activity of serine or carnitine was equal to or higher than the effect of gluconolactone (PHA) or lactic acid (AHA) at pH 4, and thus the effect was confirmed by the combination of each material and concentration of the polyol moisturizing component.
[0051] When the keratin exfoliation effect of a cosmetic composition containing serine or carnitine and the polyol moisturizing ingredients glycerin, propylene glycol and 1,3-butylene glycol was evaluated by the pig skin keratin exfoliation evaluation method, the keratin exfoliation effect dropped sharply when the content of the polyol moisturizing agent was above 0.1% by weight, and if it exceeded 10% by weight, the ability of water as a negative control group became 0%, i.e., 0%, leaving no keratin exfoliation effect, which means that the keratin exfoliation ability could not be meaningfully expressed in human experiments.
[0052] In addition, when evaluating the keratin exfoliation effect of a cosmetic composition containing serine or carnitine and polyol moisturizing ingredients glycerin, propylene glycol and 1,3-butylene glycol through a human body experiment of DHA staining, when the content of the polyol moisturizing agent was 10% and 50%, compared with the existing ability, the keratin turnover days (TT100) did not show a significant difference compared with 20 days when not applied, but the keratin turnover (Turnover) days of the keratin exfoliation enhancing cosmetic composition of the present invention were confirmed to be reduced to 17 days or less, preferably 14 days, compared with 20 days when not applied.
[0053] Therefore, the activity of the keratin exfoliating agent serine or carnitine is reduced to an insignificant level by the polyhydric alcohol moisturizing agents glycerin, dipropylene glycol, and 1,3-butylene glycol, and therefore the present invention does not substantially contain a polyhydric alcohol moisturizing agent component or provides a polyhydric alcohol moisturizing agent component within a range that does not significantly reduce its effect. That is, the keratin exfoliating composition used in the present invention refers to a case where it contains serine or carnitine and does not substantially contain one or more of the polyhydric alcohol moisturizing agents glycerin, dipropylene glycol, and 1,3-butylene glycol, or selects them within a range that does not significantly reduce their effects.
[0054] Hereinafter, the present invention will be described in more detail by way of examples. However, these are only for describing the present invention in more detail, but the scope of the rights of the present invention is not limited thereto.
[0055] 1. Combination composition
[0056] In an experiment to confirm the effect of the present invention, a more specific example of the keratin exfoliation effect of a combination of serine or carnitine and a moisturizing agent component having a polyol group is shown in the following (A) to (H) configurations.
[0057] [Table 1]
[0058]
[0059] Example 1: Evaluation of pig skin keratin exfoliation
[0060] According to the existing clinical evaluation method, it takes a long time and is difficult to conduct all experiments on various concentrations and pH conditions of various raw materials. In addition, it is difficult to conduct experiments on all situations of compositions composed of various raw materials. Therefore, the need for non-clinical keratin peeling evaluation methods has emerged.
[0061] Therefore, the present invention uses the keratin peeling evaluation method mentioned in Journal of the European Academy of Dermatology and Venereology, 2014, 28, 415-423. In detail, pig skin is biopsied and the test material is added. Thereafter, the amount of detached keratin is measured with a cell counter, and the sample in which only water is placed is displayed as a negative control group (0%), and 10% gluconolactone pH 4 is displayed as a positive control group (100%).
[0062] (1) The experimental results of various combinations (combinations A and B) of glycerol in a 5% serine solution as shown in the table below are shown. Figure 1 middle.
[0063] [Table 2]
[0064]
[0065] The experimental results showed that the efficacy of serine decreased sharply due to the addition of glycerol, and reached a range of 0% or less, where no efficacy was shown at 10% or more. Thus, it was confirmed that the efficacy of serine decreased as the concentration of glycerol increased, and that in order to exert the efficacy of the keratin peeling agent, it was necessary to use 10% or less glycerol ( Figure 1 ).
[0066] (2) The results of the keratin exfoliation test on pig skin in various combinations (combinations E and F) as shown in the table below in a 5% carnitine solution are shown in Figure 2 middle.
[0067] [Table 3]
[0068]
[0069] The experimental results showed that the keratin removal effect decreased sharply when the glycerin content exceeded 5%, and when it exceeded 10% by weight, the keratin removal effect decreased to below that of water as a negative control group, thus confirming that there was no keratin removal effect ( Figure 2 ).
[0070] (3) The results are shown in Table 4 for the case where a composition containing 5% serine and 10% and 12% glycerol, dipropylene glycol, and 1,3-butylene glycol, respectively (combinations A, B, C, and D) was formed. Figure 3 In addition, the experimental results of the combination (combination D) in which DPG was added to a 5% serine solution at a more varied concentration are shown in Figure 4 middle.
[0071] [Table 4]
[0072]
[0073] The experimental results showed that when dipropylene glycol and 1,3-butylene glycol were included, their keratin exfoliation effect was also reduced, just like glycerin. Therefore, it was confirmed that when dipropylene glycol and 1,3-butylene glycol were combined at more than 10%, the keratin exfoliation activity was reduced to below water as the negative control group, and no significant difference was shown compared with the uncoated group. Therefore, it was confirmed again that in the combination with serine, one or more selected from the moisturizers glycerin, dipropylene glycol, and 1,3-butylene glycol having a polyol group in the range of less than 10% showed keratin exfoliation effect ( Figure 3 and Figure 4 ).
[0074] (4) The results of the composition in which glycerol, dipropylene glycol and 1,3-butylene glycol were 10% and 12% respectively in 5% carnitine (combinations E, F, G and H) are shown in FIG. Figure 5 middle.
[0075] [Table 5]
[0076]
[0077] The experimental results also confirmed that when carnitine was included, all keratin peeling activities were reduced to insignificant levels in human experiments ( Figure 5 ).
[0078] (5) In addition, the results of the combination (combination I) in which a humectant mixture (glycerol 1: DPG 1: 1,3-BG 1) was added to a 5% serine solution are shown in Figure 6 middle.
[0079] Thus, it was confirmed that the keratin peeling effect of the keratin peeling agent can also be reduced by the mixed use of glycerin, dipropylene glycol, and 1,3-butylene glycol, similar to the case of using them separately. In short, when the mixture contains a moisturizing agent having a polyol group, it is also confirmed that the keratin peeling effect is shown within the inclusion range of 10% or less ( Figure 6 ).
[0080] Therefore, in combination with serine or carnitine, when the content of one or more selected from the moisturizing agents having a polyol group, such as glycerol, dipropylene glycol, and 1,3-butylene glycol, is within a range of 10% by weight or less, a keratin exfoliation effect is exhibited. Therefore, a significant keratin exfoliation effect can only be exhibited when the moisturizing agent component as described above is substantially not contained or is used at a specific concentration or less, preferably 10% by weight or less.
[0081] (6) In addition, by changing the concentration of serine, the case of using serine alone in the 0.5% serine test group and the 5% serine test group was converted to the positive control group (100%), and the values of the reduction in keratin peeling effect caused by the concentration of glycerol were compared. The results are shown in FIG. Figure 7 middle.
[0082] The experimental results confirmed that with the increase of glycerol concentration, the 0.5% serine test group ( Figure 7 blue line) and 5% serine test group ( Figure 7 The keratin peeling efficacy levels of the compositions (orange line) decreased in a similar trend, thus confirming that the concentration of the moisturizing component having a polyol group in the composition plays a major role in reducing the keratin peeling efficacy, regardless of the concentration of serine or carnitine. In summary, this means that serine or carnitine should not substantially contain a moisturizing component having a polyol group or use a moisturizing component having a polyol group at a specific concentration, preferably 10% or less.
[0083] When summarizing the above results, the keratin exfoliation ability of serine and carnitine is affected by the concentration of humectants having a polyol group, such as glycerin, dipropylene glycol, and 1,3-butylene glycol. Therefore, it was confirmed that when the humectant component having a polyol group is not substantially used or is used at 10% or less, an effect that can be felt by consumers can be exhibited.
[0084] (7) In the combination of carnitine (5%) and glycerol, the concentration of glycerol was further subdivided to conduct an experiment to measure the relative comparative value of keratin exfoliation ability. The results are shown in Tables 6 and Figure 8 middle.
[0085] [Table 6]
[0086]
[0087] The experimental results confirmed that when the glycerol content was increased from 1 wt% to 2 wt%, the keratin exfoliation ability remained almost the same, but when the glycerol content was increased to 3 wt%, the keratin exfoliation ability was significantly reduced. In addition, when the glycerol content was increased from 0.1 wt% to 0.5 wt%, the keratin exfoliation ability increased, but when the glycerol content was increased to 1 wt%, the keratin exfoliation ability was significantly reduced, thus confirming the critical significance of the content value range.
[0088] (8) In the combination of serine (5%) and glycerol, the concentration of glycerol was further subdivided to conduct an experiment to measure the relative comparative value of keratin peeling ability. The results are shown in Tables 7 and Fig. 9 middle.
[0089] [Table 7]
[0090]
[0091] The experimental results confirmed that when the glycerol content was increased from 1 wt% to 2 wt%, the keratin exfoliation ability remained almost the same, but when the glycerol content was increased to 3 wt%, the keratin exfoliation ability was significantly reduced.
[0092] (9) In the case of carnitine (0.5%), in order to confirm how the keratin exfoliation ability was inhibited by the combination of polyols compared with the conventional 5% concentration, the concentration of glycerin was subdivided to measure the relative comparative value of the keratin exfoliation ability. The results are shown in Tables 8 and 9. Fig.10 middle.
[0093] [Table 8]
[0094]
[0095] The experimental results show that when the glycerol content is 0.5% by weight, the keratin exfoliation ability is comparable to that of the case without glycerol. However, when the glycerol content is increased to 1% by weight, the keratin exfoliation ability is significantly reduced, thus confirming the critical significance of the content value range.
[0096] (10) In the case of serine (0.5%), in order to confirm how the keratin exfoliation ability was inhibited by the polyol combination compared with the conventional 5% concentration, the relative comparative value of the keratin exfoliation ability was measured by subdividing the concentration of glycerol. The results are shown in Tables 9 and Fig.11 middle.
[0097] [Table 9]
[0098]
[0099] The experimental results show that when the glycerol content is 0.5% by weight, the keratin exfoliation ability is comparable to that of the case without glycerol. However, when the glycerol content is increased to 1% by weight, the keratin exfoliation ability is significantly reduced, thus confirming the critical significance of the content value range.
[0100] Example 2: Keratin turnover enhancement evaluation method based on DHA staining method
[0101] The DHA hair dyeing method used to evaluate the rate of keratin replacement is implemented using the method mentioned in Dermatology 1993, 186, 133-137, with 10 healthy men aged 20 to 40 as the subjects. Before applying the sample, the color of the lower arm and the inner side of the upper arm of the subject's arm was measured with a colorimeter (Chromameter), and then about 0.4 ml of 10% concentration of dihydroxyacetone (Dihydroxy acetone: DHA) was applied to the inner side of the upper and lower arms for 8 hours on a 1.5X1.5cm gauze. After 48 hours, the color of the part colored brown by DHA was measured to compare the color difference with that before the application of the sample. Thereafter, the sample was applied twice a day, and the degree of fading was measured with a colorimeter to measure the time required to restore the original skin color. TT (Replacement Time) 100% refers to the time required for 100% replacement of the existing stratum corneum with a new one. In this experiment, the time required for 100% recovery of the skin pigmented by DHA was shown by relative comparative analysis by regression analysis. The results of the negative control group were calculated as 20 days and the relative values were compared.
[0102] When the results of the untreated group were calculated as 20 days, the visual changes that occurred 10 days after application are shown in Fig.12It was confirmed by visual inspection that when the glycerol concentration was 10% or 50%, there was no significant difference from the uncoated group, and when serine was used alone, the effect was shown at both 0.5% and 5% concentrations.
[0103] If it is converted into a quantitative value by a colorimeter, it is Fig.13 When 5% serine was used, TT100% was confirmed to decrease from 20 days in the uncoated group to 14 days, a decrease of about 6 days, but when the concentration of glycerol was 10% or 50%, no significant change was found compared with the uncoated group.
[0104] All combination groups did not cause side effects such as erythema, itching, burning sensation, etc. during and after the experiment. Thus, the hindrance of keratin exfoliation ability within the concentration range selected by the pig skin keratin exfoliation evaluation method was also confirmed in the human experiment. In summary, it was confirmed that for the keratin exfoliation effect of serine or carnitine, by substantially not using or using glycerin, dipropylene glycol, 1,3-butylene glycol, a moisturizer having a polyol group, at a specific concentration of 10% or less, it is possible to show a keratin exfoliation enhancement effect that consumers can feel.
[0105] The applicant has described a preferred embodiment of the present invention above, but the embodiment described above is only one embodiment for realizing the technical idea of the present invention, and it should be understood that as long as the technical idea of the present invention is realized, any variation or modification also belongs to the scope of the present invention.
