Abstract
Description
Technical Field
[0001] The invention relates to the technical field of traditional Chinese veterinary medicines, and in particular to a traditional Chinese medicine composition for effectively preventing and treating livestock coccidiosis, and a preparation method and application thereof. Background Art
[0002] Coccidiosis is one of the important parasitic diseases that endanger the cattle and sheep breeding industry. It has a high incidence rate and mainly affects calves and lambs. The clinical manifestations of the disease are mainly characterized by persistent diarrhea, anemia and emaciation, and it is easy to lead to susceptibility to other diseases. Unlike the typical clinical symptoms of poultry after coccidia infection (such as bloody stools and high mortality rate), cattle and sheep usually only have bloody stools in 1%-5% of cases after coccidia infection, and about 95% of cases show varying degrees of non-hemorrhagic diarrhea. However, under hot and humid environmental conditions, when acute coccidiosis outbreaks occur in large-scale cattle farms, the incidence of bloody stools can be as high as 40% without group intervention. Both bloody stools and chronic diarrhea damage the intestines, thereby affecting nutrient absorption, intestinal function, feed conversion rate and animal growth. The economic losses caused by subclinical symptoms of cattle and sheep after coccidia infection far exceed the losses caused by clinical disease. For severely infected cattle and sheep, although they survive, their growth is stunted and it is difficult to generate economic benefits. In addition, subclinical coccidiosis is difficult to diagnose, which often leads to misdiagnosis or delayed treatment, and even directly causes the death of cattle and sheep. Studies have shown that the economic losses of dairy cows and buffaloes caused by coccidiosis are as high as US$731 million each year. Therefore, the potential threat of coccidiosis to the cattle and sheep breeding industry cannot be ignored.
[0003] At present, there is no drug in China that can prevent and control diarrhea caused by coccidiosis in cattle and sheep. In addition to purifying and preventing coccidiosis, good anticoccidial drugs should not affect the body's immunity to coccidiosis. Studies have shown that anticoccidial drugs that allow a small amount of oocysts to be produced are more desirable than anticoccidial drugs that completely inhibit oocyst production, and multiple immunizations with a small amount of coccidia can produce a "trickle-down effect." A large amount of clinical efficacy evaluation data confirms that traditional anticoccidial drugs still have many problems in controlling coccidiosis in cattle and sheep: (1) Toltrazuril can block different stages of anthelmintic development and kill coccidia completely, but cannot stimulate the body to produce immunity and cannot effectively relieve diarrhea caused by coccidiosis; (2) Sulfonamides only block the early development of coccidia and control secondary infections of coccidiosis, and have strong nephrotoxicity; ionophore anticoccidial drugs such as monensin need to be added for a long time and have a small safety range. The side effects caused by long-term addition have become an important factor in the digestive health problems of calves since the ban on antibiotics. In summary, traditional antibiotics or chemical drugs are more suitable for the treatment of coccidiosis rather than prevention. It not only affects the production of the body's natural immunity, but also has disadvantages such as toxic side effects, drug residues, and drug resistance in coccidia, which seriously threatens human and animal health and food public health safety. Therefore, in order to control the negative impact of coccidiosis in cattle and sheep, whether it is acute death from clinical coccidiosis or decreased production performance from subclinical coccidiosis, it is urgent to develop and innovate new "preventable and curable" health care drugs and alternatives to provide more options for optimizing the prevention and control of coccidiosis in cattle and sheep. [0027] The Chinese medicine composition of the present invention is obtained by screening according to the traditional Chinese veterinary medicine theory and targeting the symptom characteristics of coccidiosis. The analysis process is as follows:
[0028] 1. Discussion on the etiology, pathogenesis, treatment principles and methods of bovine coccidiosis in veterinary medicine
[0029] Cause: Traditional Chinese medicine explains the cause of parasitic diseases as "dampness and heat breed worms" mainly from the following aspects:
[0030] Dampness and heat breed worms: The traditional Chinese medicine theory of "dampness and heat breed worms" emphasizes that environmental dampness and heat are external factors that make worm eggs infectious. It also emphasizes that spleen and stomach dampness and heat are internal factors for the occurrence and development of parasitic diseases. As dampness traps the spleen and turns into heat, a hotbed of gastrointestinal dampness and heat is formed that is suitable for the proliferation of parasites. It also emphasizes that only when the three causes and conditions are complete can parasitic diseases become prevalent.
[0031] Unclean diet: Traditional Chinese medicine believes that unclean diet is one of the main causes of parasites. Because the oocysts excreted from the body with feces are not infectious, even if they are eaten by mistake, they will not multiply in the intestines and cause disease. They are only infectious when they complete sporulation in a hot and humid environment. At this time, eating infected water and soil contaminated by sporulated eggs may cause parasite infection.
[0032] Damp-heat in the spleen and stomach: Parasitic diseases often occur when the environment is damp and hot, which means that when the body is exposed to damp heat, the spleen likes dryness and hates dampness, and excessive dampness will trap the spleen. Damage to the spleen yang will inevitably disrupt the spleen's function of transporting water and dampness, resulting in excessive dampness, which will remain in the gastrointestinal tract and become heat after a long period of depression. Dampness will turn into damp heat when it meets heat. The spleen is also the source of phlegm. If the spleen yang is damaged for too long, too much phlegm will accumulate in the body. If the phlegm and dampness stay for too long, they will also become hot and gastrointestinal dampness and heat will combine. After the combination of dampness and heat, a breeding ground for parasites will be formed. Parasites proliferate abnormally in the gastrointestinal tract, disrupting the balance of the gastrointestinal microbiome and the balance of the gastrointestinal system in raising the clear and lowering the turbid, which will lead to the main clinical manifestation of "damp-heat diarrhea".
[0033] Pathogenesis: According to TCM theory, damp-heat bloody diarrhea, worm accumulation and other symptoms are caused by damp-heat in the internal and external environment and exogenous worm evil. On the one hand, the worm evil injures the yin, consumes blood and hurts the body fluid, and on the other hand, the damp-heat is accumulated internally, trapping the spleen yang. In the Eight-Principle Syndrome Differentiation, it is firstly "real syndrome" and then "deficient syndrome".
[0034] Damp-heat syndrome of large intestine: Exogenous damp-heat, intestinal qi blockage, spleen and stomach dysfunction, then worm accumulation occurs, leading to damp-heat intestinal diarrhea.
[0035] Spleen deficiency and damp-heat syndrome: Long-term accumulation of worms, spleen deficiency, long-term diarrhea and dysentery, and inability to lift.
[0036] (3) Syndrome of spleen and kidney deficiency: Pests damage Yin, consume blood and body fluid, damage both Qi and Yin, Yin is exhausted and Yang is lost, suddenly becoming dangerous.
[0037] In summary, the clinical manifestations caused by coccidiosis in cattle are mainly non-hemorrhagic diarrhea or bloody stools, which can be diagnosed as damp-heat. The affected animals mainly show the following symptoms of damp-heat diarrhea: loose or soft or watery stools, often mixed with bubbles, yellow-green in color, occasionally mixed with occult blood or mucus or pus and blood, with a strong or fishy or foul smell; red and yellow tongue, thick and greasy fur; thirsty and willing to drink but not in a hurry; short yellow urine, deep and rapid pulse. Damp-heat diarrhea is the most common syndrome of coccidiosis, and it is the best time to prevent disease. The treatment method is mainly to clear heat and dampness, cool blood and stop bleeding, and kill insects; while those with long-term worm accumulation, spleen deficiency and damp-heat should use the method of attacking and supplementing. The treatment method is mainly to suppress and kill coccidiosis and regulate the spleen and stomach. After deworming, Shenling Baizhu San is used to regulate the spleen and stomach. Severely ill cattle are damaged by worm evil, which consumes blood and body fluid. Because of supplementation first and elimination later, Bazhen Decoction is used to replenish qi and blood first, and then deworming.
[0038] 2. Cubic basis analysis
[0039] Based on the above causes of the disease, it is almost impossible to prevent and control coccidiosis by regulating the climate, regulating the humidity and heat of the pens, and disinfecting the site. If the causes of the disease are changed to dampness and heat in the spleen and stomach, and phlegm and fluid, then even if food with insect eggs is accidentally eaten, no parasites will occur. Therefore, it is necessary to use cold medicines to remove the heat, dry agents to remove the dampness, and expectorant medicines to remove the phlegm, and use sore-healing medicines in combination, all at the same time, to achieve the effects of clearing away heat, drying dampness, stopping diarrhea, and inhibiting (killing) insects.
[0040] 3. Formula composition, efficacy and explanation
[0041] Chinese medicine formula: Artemisia annua, Sophora flavescens, Coptis chinensis, Pulsatilla scabra, stir-fried Sophora japonica flowers, Sanguisorba officinalis, Platycladus orientalis charcoal and Amomum villosum.
[0042] Efficacy: It is mainly used to treat coccidiosis-induced damp-heat diarrhea in cattle and sheep. Symptoms include loose, soft or watery stools, often mixed with bubbles, yellow-green in color, occasionally mixed with occult blood, mucus or pus and blood, with a strong, fishy or foul smell; red and yellow tongue with thick and greasy coating; thirsty and willing to drink but not in a hurry; short yellow urine, deep and rapid pulse.
[0043] Explanation of the prescription: Artemisia annua in the prescription can stop diarrhea, clear away heat and detoxify, inhibit and kill intestinal protozoa, and stop malaria. Sophora flavescens can clear heat and dry dampness, kill insects and promote diuresis, and they are all the main drugs; Coptis chinensis and Pulsatilla striata can clear heat and toxicity, expel insects and stop diarrhea, and remove the cause of the disease. With the help of Artemisia annua, the effect of inhibiting insects and stopping dysentery is better, and they are all the assistant drugs; and Sanguisorba officinalis can cool blood and astringe, reduce swelling and heal sores, and cooperate with fried Sophora japonica flower and Platycladus orientalis charcoal to treat bloody stool and bloody dysentery in the lower part of the body. Amomum villosum is pungent, dispersing, warming and unblocking, invigorating the spleen and warming the stomach, warming the middle and removing turbidity, digesting food and removing accumulation, drying dampness and removing phlegm, unblocking the middle part of the body, removing dampness and heat, and then eliminating worm accumulation and food accumulation, and restraining each other with the bitter drugs in the prescription to prevent the drugs from being too cold and damaging the stomach, and they are the guiding drugs; the drugs are combined to clear heat and dry dampness, remove dampness and stop dysentery, eliminate worm accumulation, and take into account both the symptoms and the root causes, and play the role of drying dampness and stopping dysentery, inhibiting (killing) insects.
[0044] The Chinese medicine composition of the present invention has obvious therapeutic effect on natural coccidiosis of cattle, and the prescription is used to treat natural coccidiosis of sheep, and also achieves ideal effect. However, it is used to treat artificial coccidiosis of chicken, but the effect is very limited, which may be related to the infection mode, poultry physiology and pathology and prescription dosage form. Therefore, the prescription powder is suitable for the treatment of ruminant livestock (coccidiosis) infection.
[0045] The therapeutic effects of the Chinese medicine composition of the present invention are described in detail below with specific examples.
[0046] Example

[0050] Verification Example 1
[0051] From June 1, 2022 to June 30, 2022, dairy cows under 7 months old with clinical symptoms such as diarrhea were selected from four large-scale dairy farms in Ningxia (numbered A, B, C and D), and no drug treatment was used. Fresh feces were collected by rectal sampling, and the sample cow number, age and fecal characteristics were recorded in detail. They were stored at 4°C and sent to the laboratory for testing. After testing, 21 cows in dairy farm A, 40 cows in dairy farm B, 67 cows in dairy farm C, and 596 cows in dairy farm D were positive for coccidiosis and were all used as experimental animals in this study. Venous blood samples were collected from the experimental cows in the three dairy farms A, B, and C before and after the test, and serum was drawn after centrifugation for detection of immune and antioxidant indicators.
[0052] 1. Test methods
[0053] 1. Inclusion and Exclusion Criteria
[0054] Inclusion criteria: According to the diagnostic criteria for bovine coccidiosis in Ningxia (DB64 / T 1723-2020), cattle diagnosed as coccidia-positive; only 1%-5% of the test group showed typical clinical symptoms of bloody stools, and about 95% showed subclinical symptoms of varying degrees of non-hemorrhagic diarrhea.
[0055] Exclusion criteria: cows with poor body condition and other diseases; cows not treated according to the medication regimen; cows eliminated by the farm during the treatment process.
[0056] 2. Scoring criteria and observation content
[0057] The sick calves were diagnosed with coccidiosis diarrhea and met the inclusion criteria, and the basic information of the calves was registered, including age, rumen filling (feed intake), coat color, mental state, eye secretions, respiratory symptoms, fecal characteristics, etc. The fecal condition scoring criteria are: normal feces with good shape is 1 point; soft feces are 2 points; thin feces (with blood, bubbles or no bubbles) is 3 points; watery feces (no blood, no bubbles, or with blood and bubbles) is 4 points. The rumen filling scoring criteria are: full rumen is 1 point; rumen pits are 2 points; rumen collapse is 3 points. The mental state scoring criteria are: active is 1 point; like to lie down is 2 points; fatigue is 3 points. The coat color scoring criteria are: smooth and shiny is 1 point; smooth hair with stains is 2 points; dry hair is 3 points. The scoring criteria for eye expression and eye secretions are: 1 point for sensitive eyes without secretions, 2 points for sensitive eyes with secretions, and 3 points for dull eyes with secretions. The scoring criteria for respiratory symptoms are: 0 points for no symptoms, 1 point for runny nose, 2 points for cough, and 3 points for cough and wheezing.
[0058] 3. Trial grouping, drug administration and clinical observation
[0059] 21 calves aged 3 to 5 months that were positive for coccidiosis in dairy farm A, 40 calves in dairy farm B, and 67 calves in dairy farm C were administered 80 g / head / time of the Chinese medicine composition (prepared in Example 1) mixed with TMR pellets once a day for a course of treatment of 3 consecutive days. Samples were collected before and on the 6th day of medication, and the fecal condition, rumen fullness (feed intake), mental state, coat color, eye expression and eye secretions, and respiratory symptoms were recorded in detail.
[0060] A total of 596 dairy calves positive for coccidiosis in Dairy Farm D were randomly divided into three groups according to their monthly ages: 188 calves in the traditional Chinese medicine composition group, 180 calves in the 5% toltrazuril suspension group, and 228 calves in the control group. The medication methods were as follows: In the traditional Chinese medicine composition group, for calves aged 1 to 2 months (43 calves), the dosage was 10 - 20 g / head / time, orally administered by adding it to the milk; for calves aged 2.5 to 3 months (43 calves), the dosage was 40 - 50 g / head / time, administered by mixing it with the granular feed; for calves aged 3 to 4 months (41 calves), the dosage was 60 - 80 g / head / time, administered by mixing it with the TMR granular feed; for calves aged 6 to 7 months (61 calves), the dosage was 120 - 140 g / head / time, administered by mixing it with the TMR diet. The medication was given once a day, and a 3 - day course of treatment was used. In the 5% toltrazuril suspension group, for calves aged 1 to 2 months (40 calves), 2.5 to 3 months (40 calves), 3 to 4 months (40 calves), and 6 to 7 months (60 calves), the dosage was 0.3 mL / kg by gavage, once a day, and the medication was given once. In the control group, for calves aged 1 to 2 months (40 calves), 2.5 to 3 months (40 calves), 3 to 4 months (40 calves), and 6 to 7 months (108 calves), no medication treatment was given. Samples were taken before medication and on the 6th day of medication, and the fecal condition, rumen fullness (feed intake), mental state, hair color condition, eye expression and eye secretions, and respiratory symptoms were recorded in detail.
[0061] 4. Efficacy determination
[0062] After treatment, if the coccidia detection in the diseased calves was negative, the mental state and diet returned to normal, the feces were of moderate hardness, the nasal mirror was moist, the diarrhea symptoms disappeared continuously for 24 hours, and the fecal score reached 1 point, it was determined as cured; if the coccidia infection rate in the diseased calves decreased, the infection intensity decreased, the clinical symptoms improved, the diarrhea symptom score decreased to 2 points, and the diet and other symptoms improved, it was determined as effective; if all symptoms of the diseased calves did not improve or worsened, and even the treatment was ineffective and the calves died, it was determined as ineffective.
[0063] 5. Collection and analysis of experimental data
[0064] The Excel software was used to statistically analyze the detection data; the paired t - test in the SPSS 27.0 (SPSS Inc, Chicago, USA) software was used to analyze the differences in the immune and antioxidant index data before and after treatment, presented as means ± standard deviation (means ± SD). If 0.01 < P < 0.05, the difference in the infection rate between groups was significant; if P < 0.01, the difference in the infection rate between groups was considered extremely significant.
[0065] II. Experimental results
[0066] 1. Inclusion and exclusion of cases
[0067] According to the inclusion criteria, 21 calves from dairy farm A, 40 calves from dairy farm B, 67 calves from dairy farm C, 188 calves from dairy farm D in the calf-trezuril group, 180 calves from the toltrazuril group, and 228 calves from the control group were included in this study. Two calves from dairy farm A died after inclusion, so they were excluded from this study. Finally, 19 calves from dairy farm A, 40 calves from dairy farm B, 67 calves from dairy farm C, and 596 calves from dairy farm D were included in the study.
[0068] 2. Changes in coccidia infection rate, infection intensity and worm species
[0069] 2.1 Changes in infection rate and intensity of coccidia in cattle before and after treatment with the Chinese medicine composition

[0081] After treatment, the diarrhea, feed intake, coat color, mental state, eye secretions, and respiratory symptoms of the experimental calves in each dairy farm were greatly improved, with significant differences compared with before treatment ( Figure 2 ). The overall effective rate of the Chinese medicine combination for diarrhea reached 90.13% (283 / 314), which was significantly higher than the effective rate of the control drug toltrazuril treatment group of 58.33% (105 / 180) (P<0.001), among which the cure rate of the Chinese medicine combination for diarrhea was 80.89% (254 / 314), which was significantly higher than the cure rate of the control drug toltrazuril group of 31.67% (57 / 180). Among them, the cure or effective rate of diarrhea in the four dairy farms A, B, C, and D reached 89.47% (17 / 19), 95.00% (38 / 40), 92.54% (62 / 67), and 88.30% (166 / 188), respectively. The proportion of cattle with full rumen (greater than 2 months old) increased from 17.58% (48 / 273) before treatment to 85.24% (231 / 271), with a very significant difference (P<0.01). The proportion of cattle with smooth and shiny coat increased from 29.75% (94 / 316) to 64.33% (202 / 314), with a very significant difference (P<0.01). The proportion of cattle with sharp eyes and no secretions increased from 38.61% (122 / 316) to 86.94% (273 / 314), with a very significant difference (P<0.01). The proportion of cattle with eliminated respiratory symptoms increased from 31.33% (99 / 316) to 67.20% (211 / 314), with a very significant difference (P<0.01).
[0082] 3.2 Improvement of clinical symptoms in calves of different months in the experimental group and the control group
[0083] In order to explore the difference in clinical effects of Chinese medicine combination on calves of different months of age, 596 calves with positive coccidiosis in D dairy farm were randomly divided into a Chinese medicine combination group of 188 calves, a 5% toltrazuril suspension group of 180 calves and a control group of 228 calves according to their age. They were divided into four age groups: 1 to 2 months, 2.5 to 3 months, 3 to 4 months, and 6 to 7 months. The clinical symptoms of each age group after the experiment are as follows ( Figure 3 AF):
[0084] The diarrhea of calves aged 1 to 2 months was cured or improved, and the spirits were lively. The difference between the Chinese medicine group and the toltrazuril group and the control group was extremely significant (P<0.01); the eyes were sharp and there was no eye secretion. The difference between the Chinese medicine group and the control group was significant (P<0.05), but the difference between the Chinese medicine group and the toltrazuril group was not significant (P>0.05); the coat color was smooth and shiny, and the respiratory symptoms were eliminated. There was no significant difference between the Chinese medicine group and the toltrazuril group and the control group (P>0.05).
[0085] At 2.5 to 3 months of age, the rumen was full, the spirit was lively, the eyes were sharp and there was no eye secretion. The difference between the Chinese medicine group and the toltrazuril group and the control group was extremely significant (P<0.01). In terms of smooth and shiny coat color and elimination of respiratory symptoms, there was no significant difference between the Chinese medicine group and the toltrazuril group and the control group (P>0.05).
[0086] The diarrhea of 3 to 4 month old calves was cured or improved, the rumen was full, the spirit was lively, the eyes were sharp and without eye secretions, the coat was smooth and shiny, and the respiratory symptoms were eliminated. The differences in various indicators between the Chinese medicine group and the toltrazuril group and the control group were extremely significant (P<0.01).
[0087] The rumen of 6- to 7-month-old dairy cows was full, the spirit was lively, the eyes were sharp and there was no eye secretion. The difference between the Chinese medicine group and the toltrazuril group and the control group was extremely significant (P<0.01); the respiratory symptoms were eliminated, and the difference between the Chinese medicine group and the toltrazuril group and the control group was significant (P<0.05); the difference between the Chinese medicine group and the toltrazuril group in terms of diarrhea cure or improvement was extremely significant (P<0.01), and there was no significant difference between the Chinese medicine group and the control group (P>0.05); the coat color was smooth and shiny, and the difference between the Chinese medicine group and the toltrazuril group was significant (P<0.05), and there was no significant difference between the Chinese medicine group and the control group (P>0.05).
[0088] 4. Changes in immune and antioxidant indicators
[0089] Blood samples of the experimental cattle from the three dairy farms A, B, and C were collected before and after treatment with the Chinese medicine composition to detect immune and antioxidant indicators: immunoglobulin A (IgA), immunoglobulin G (IgG), immunoglobulin M (IgM), total antioxidant capacity (T-AOC), superoxide dismutase (SOD), malondialdehyde (MDA), and catalase (CAT). Through analysis, it was found that the Chinese medicine composition can improve the content and antioxidant properties of immunoglobulins in calves (Table 4). Among them, the difference between IgG in dairy farm A before and after treatment was extremely significant (P<0.01), and the difference between IgA in dairy farm B before and after treatment was extremely significant (P<0.01). The effect was more significant in dairy farm C. After treatment, the increase in the content of IgG, IgM, SOD, and CAT was extremely significant compared with that before treatment (P<0.01), and the content of MDA was significantly reduced compared with that before treatment (P<0.01).
[0090] Table 4 Changes in immune and antioxidant indicators of experimental cows in each dairy farm before and after treatment with the Chinese medicine composition
[0091]
[0092] From the above test results, it can be seen that the Chinese medicine composition of the present invention can effectively improve clinical symptoms such as diarrhea, reduce the infection rate and infection intensity of coccidia, and effectively remove highly pathogenic worm species in cattle, discharge low pathogenic worm species in small quantities (trickle effect), and has obvious therapeutic effects on respiratory symptoms caused by coccidiosis. According to the theory of the Five Elements of Traditional Chinese Medicine, the lung is an internal organ and the large intestine is an internal organ, and the lung and the large intestine are exterior and interior, so the spleen and stomach are healthy and the lungs are peaceful. It can also improve the immunity and antioxidant properties of animal bodies, and provide alternative drugs for the clinical treatment of bovine coccidiosis.
[0093] Verification Example 2
[0094] Sheep coccidiosis is a protozoan disease caused by Eimeria parasitizing the sheep intestine. All breeds of sheep and goats are susceptible to coccidiosis. Infected sheep show symptoms such as diarrhea, weight loss, anemia, poor growth, and even death. However, sheep farms are currently unaware of the existence of sheep coccidiosis and have not specifically carried out epidemic prevention against coccidiosis. In order to confirm that sheep coccidiosis infection is mostly latent infection, and the Chinese medicine composition of the present invention has a preventive and therapeutic effect on yangqiuchong, the following is an investigation of the coccidiosis infection in the sheep farm and the Chinese medicine composition of the present invention is used for prevention and treatment, and its effect on the excretion of coccidia oocysts in sheep feces and diarrhea is evaluated.
[0095] 1. Coccidia infection in sheep farms
[0096] In October 2023, 96 samples of fresh feces just excreted on the ground were collected at the Xinzhongshenghu Sheep Farm in Shaanxi Province, and placed separately in clean ziplock bags. The sample information (age in months, feces status) was recorded, and then the samples were sent to the laboratory of Ningxia Academy of Agricultural and Forestry Sciences. The fecal samples were placed in centrifuge tubes, added with 2.5% potassium dichromate solution and stored in a 4°C refrigerator. Coccidian oocysts were counted by the McMaster method. Take 2g of fecal sample and place it in a clean 100mL beaker, add 8mL of saturated saline solution, smash and mix the fecal sample with a glass rod, and then add 50mL of saturated salt solution. After mixing, filter with a 60-mesh sieve immediately, draw the filtrate to fill 2 counting chambers, let it stand on the microscope stage for 3-5min, and count under a 10x objective. There are 100 squares (1cm×1cm×0.15cm) in each counting chamber. After checking the number of oocysts n1 and n2 in 100 squares of the two counting chambers respectively, calculate OPG according to the following formula.
[0097] OPG=[(n1+n2) / (2×0.15)]×60 / 2.
[0098] The infection rate and infection intensity of coccidia in Hu sheep of different ages are shown in Table 5.
[0099] Table 5 Coccidia infection rate and infection intensity of Hu sheep at different ages
[0100]
[0101] The above data show that the infection rate of coccidia in Hu sheep of different ages in Shaanxi Xinzhongsheng Hu sheep farm was 48.96%, and the average infection intensity (OPG) was 699.5, among which the average infection intensity of Hu sheep aged 30-90 days was the highest, which was 1263.
[0102] 2. Experimental group design and treatment
[0103] In January 2024, a group of 3-month-old Hu sheep with severe diarrhea were randomly selected from Shaanxi Xinzhongsheng Hu sheep farm and fed with the Chinese medicine combination for 3 consecutive days. Five samples of fresh feces excreted around the anus or on the ground were collected on the 0th day (before medication), the 3rd day (after the end of medication), and the 13th day. They were placed separately in clean ziplock bags, and the sample information (age in months, feces status) was recorded. The samples were then sent to the laboratory of Ningxia Academy of Agricultural and Forestry Sciences.
[0104] 3. Test methods
[0105] (1) Coccidia oocyst count and OPG value
[0106] The feces samples were placed in centrifuge tubes, added with 2.5% potassium dichromate solution and stored in a refrigerator at 4°C. The coccidia oocysts in the feces were counted and the OPG values were calculated. The coccidia OPG values before and after medication are shown in Table 6.
[0107] Table 6 Coccidia OPG values before and after medication
[0108]
[0109] Before the test, the fecal samples were tested, and the positive rate of coccidia oocysts was 40%, and the average OPG value was 80; 3 days after the drug was used, the positive rate of coccidia oocysts was 60%, and the average OPG value was 540. On the 10th day after the end of the drug use, the positive rate of coccidia oocysts was 20%, and the average OPG value was 20, which was lower than the positive rate before the drug use, and the intensity was significantly reduced.
[0110] (2) Stool scoring
[0111] The collected feces were subjectively scored according to the Huyang feces scoring standard. The scoring standard and the feces scoring results before and after medication are shown in Tables 7 and 8, respectively.
[0112] Table 7 Scoring criteria for Hu sheep feces
[0113]
[0114] Table 8 Fecal scores before and after medication
[0115]
[0116] From the data in Table 8, it can be seen that among the 5 stools collected before the test, 1 had moderate diarrhea, 4 had severe diarrhea, and the diarrhea rate was 100%; 3 days after medication, 1 had mild diarrhea, 4 were normal, and the diarrhea rate was 20%; on the 10th day after the end of medication, all 5 stools were normal, and the diarrhea rate was 0.
[0117] From the above test results, it can be seen that the Chinese medicine composition of the present invention shows significant anticoccidial effect and the effect of treating diarrhea. After medication, as diarrhea is relieved and eliminated, the positive rate of coccidia and the infection intensity are significantly reduced. This is because the infection time of each sheep in the natural infection case is inconsistent, and the excretion of oocysts has no direct correlation with the degree of coccidiosis. However, it fully proves the effectiveness of the prescription of the invention. The Chinese medicine composition of the present invention provides an effective prevention and treatment method for sheep health management, which is beneficial to improve the production performance and health level of sheep.
[0118] Comparative test example
[0119] Since there are significant differences in the intestinal structure and physiological characteristics of poultry and livestock, this affects their susceptibility to coccidiosis, clinical manifestations and treatment methods. The Chinese medicine composition of the present invention is based on the pathogenesis, etiology and pathology of coccidiosis in livestock cattle, and is used for dialectical medication. In order to verify whether it has the same effect on poultry, the following test is conducted.
[0120] 1. Methods
[0121] 1. Experimental animals and experimental drugs
[0122] The experimental animals were one-day-old broilers; the experimental drug was the Chinese medicine composition of Example 1 of the present application, which was prepared into granules for the convenience of chickens to eat; the sensitive drug was toltrazuril.
[0123] 2. Experimental design grouping
[0124] The experimental chicks were raised to 14 days old, and after weighing each chick, the thin or overweight chicks were eliminated. Healthy chickens with individual weight differences within 10g were selected and randomly divided into 6 groups, with 20 in each group. Groups 1-3 were drug-treated insect-fighting groups, group 5 was a sensitive drug control group, and there were also non-drug-fighting insect-fighting test group No. 4 and non-drug-fighting insect-fighting test group No. 6. Each chicken in each drug test group and non-drug-fighting insect-fighting test group was orally inoculated with 1.2×10 5 The 1-3 groups were administered with feed; the 5th group was administered with drinking water, and the animals were fed and drank freely until the end of the experiment on the 7th day, as shown in Table 9.
[0125] Table 9 Experimental design grouping
[0126]
[0127] 3. Evaluation criteria for anticoccidial index
[0128] Scoring of bloody stools: On the fifth day after infection, observe the proportion of bloody stools, take photos and record them, and score bloody stools: For Eimeria tenella infection, within a given time range of 12-24 hours, 0 points means 100% of the stools are bloodless, +1 point means 25% of the stools are bloody, +2 points means 50% of the stools are bloody, +3 means 75% of the stools are bloody, and +4 points means 100% of the stools are bloody.
[0129] Relative weight gain rate: Weigh the chickens at the beginning and end of the experiment, and calculate the average weight gain and relative weight gain rate. Relative weight gain rate = (weight gain rate of the experimental group / weight gain rate of the experimental group without medication or insect attack) × 100%.
[0130] Survival rate: record the number of dead chickens in each group, determine the cause of death by autopsy, and calculate the survival rate. Survival rate = (number of surviving chickens at the end of the experiment / number of chickens in the experimental group) × 100%.
[0131] Oocyst value: Use the McMaster counting method to count fecal oocysts, find the number of fecal oocysts per gram group (OPG), calculate the number of oocysts, and convert the oocyst value according to Table 10.
[0132] Table 10 Conversion relationship between oocyst number and oocyst value
[0133]
[0134] Anticoccidial index (ACI): It is calculated according to the following formula, i.e.:
[0135] ACI = (relative weight gain rate + survival rate) × 100 - (lesion value + oocyst value).
[0136] Pharmacodynamic judgment criteria: ACI > 180 belongs to highly effective anticoccidial drugs; 160 < ACI < 180 belongs to moderately effective anticoccidial drugs; 120 < ACI < 160 belongs to low - effective anticoccidial drugs; ACI < 120 is ineffective against coccidia.
[0137] II. Results
[0138] 1. Clinical symptoms
[0139] In the non - medicated coccidia - challenged control group, the experimental chickens gradually showed reduced feed intake, poor spirit, etc. after being infected with sporulated oocysts. On the 4th day after infection, the experimental group and the non - medicated coccidia - challenged control group excreted bloody stools to varying degrees, with reduced water intake, which was more severe on the 5th and 6th days, and some groups (No. 2 and No. 4) had dead chickens. There was no bloody feces in the medicated control group and the non - medicated non - coccidia - challenged control group, and the feed intake and water intake were normal.
[0140] 2. Results of bloody stool scoring
[0141] From the observation results of bloody stools on the 5th day after coccidia - challenge, the bloody stool conditions of groups 1 - 3 and control group 4 all showed +4 points, and the bloody stool conditions of sensitive medicated control group 5 and blank control group 6 were 0 points.
[0142] 3. Results of drug sensitivity test
[0143] The test results showed that groups 1 - 3 of drugs were all ineffective; the anticoccidial index of medicated control group 5 was higher than 180, showing high efficiency. The detailed results are shown in Table 11.
[0144] Table 11 Results of drug sensitivity test
[0145] Group Relative weight gain rate / % Survival rate / % Lesion value Oocyst value ACI value standard No. 1 78.64 90.00 27.50 40.00 111.14 invalid No. 2 85.28 85.00 30.00 40.00 100.28 invalid No.3 61.52 90.00 24.00 40.00 97.52 invalid No. 4 71.35 85.00 29.00 40.00 87.35 Insect attack comparison No. 5 93.31 100.00 0.00 0.00 193.31 Drug control No. 6 100.00 100.00 0.00 0.00 200.00 Blank control
[0146] From the above results, it can be seen that the traditional Chinese medicine composition of the present invention is ineffective against chicken coccidiosis at different recommended concentrations.
[0147] The above test examples prove that the traditional Chinese medicine composition prepared in Example 1 of the present invention has obvious therapeutic effects on livestock coccidiosis, but for poultry coccidiosis, it has no obvious anticoccidial effect. This may be related to the infection mode, the intestinal structure of livestock and poultry, and their own biological characteristics. In order to explore the optimal drug ratio of the traditional Chinese medicine composition for treating livestock coccidiosis, adjustments such as Scheme 1 - 3 were made on the basis of the drug components in Example 1, and at the same time, the drug components were adjusted as in Scheme 4 - 6.
