Abstract
Description
Technical Field
[0001] The present invention belongs to the technical field of cosmetics, and relates to a Chinese herbal medicine composition, wherein the composition is made of or contains extracts of burdock fruit, gentian, stephania tetrandra and centella asiatica. The present invention also relates to a preparation method and use of the Chinese herbal medicine composition. The present invention further relates to a medicine or cosmetic containing the Chinese herbal medicine composition. Background Art
[0002] As the largest organ of the human body, the skin bears the important task of resisting harmful damage from the external environment and maintaining the health of the body. When the homeostasis of the skin barrier is damaged, the skin will appear in a sensitive metastable state, and in severe cases it may even lead to various skin diseases such as atopic dermatitis and psoriasis. As the number of people with sensitive skin continues to increase worldwide, people are paying more and more attention to the repair of sensitive skin. Therefore, repairing the skin barrier is currently a key development direction in the field of skin science and cosmetic application research.
[0003] The skin is mainly composed of four major barriers: physical barrier, chemical barrier, microbial barrier and immune barrier. Among them, the integrity of the physical barrier is jointly determined by the stratum corneum and the tight junction barrier, which is the basis for the other three barriers to play a normal physiological role. Therefore, repairing the physical barrier in the skin is crucial to improve sensitive skin and maintain skin health.
[0004] Keratinocytes are the main component of the skin barrier. Filaggrin encoded by FLG is an important marker protein at the end of keratinocyte differentiation, and its decomposition product, natural moisturizing factor (NMF), is a water-soluble hygroscopic component of the skin. Therefore, filaggrin plays an important role in maintaining normal pH, hydration capacity and mechanical integrity of the stratum corneum. Tight junction proteins are a type of structural protein that connects keratinocytes and prevents solute penetration in the epidermis. They have material and ion size selectivity, so they are also called tight junction barriers. The tight junction barrier mainly includes Occludins, Claudins and ZO molecular families, among which Claudin-1 and Zonula occludens-1 (ZO-1) are the main transmembrane and cytoplasmic proteins in tight junction proteins, respectively. ZO-1 can connect transmembrane proteins to the cytoskeleton to transmit various signals to cells. When the skin barrier is damaged, the stratum corneum and tight junctions are destroyed, resulting in reduced levels of Filaggrin, Claudin-1 and ZO-1, impaired skin mechanical integrity, hydration function, and epidermal permeability, making it easier for external stimuli to penetrate the skin, thereby triggering an inflammatory response, further promoting barrier damage, and ultimately leading to skin sensitivity. Therefore, for sensitive skin damaged barrier repair, it is crucial to improve the physical barrier (especially the tight junction barrier).
[0005] At present, there are various technologies for skin barrier repair. For example, CN202211017250 discloses a composition and its application with synergistic effect of repairing skin barrier, the composition includes inca inchi seed oil and ceramide, and the composition involved achieves synergistic improvement of gene expression level and transcutaneous resistance of skin barrier-related proteins by supplementing structural lipids, showing the effect of repairing skin barrier; CN202311161297 discloses a grease composition that can promote skin barrier function, the composition involved includes sea buckthorn seed oil and sunflower seed oil as active ingredients, which can increase the aggregation force of corneocytes and improve skin barrier function by enhancing one of the main proteins of tight junctions of keratinocytes-demosome protein. However, the above inventions all achieve the effect of repairing barrier function by compounding fat-soluble components, and there are problems such as poor stability of fat-soluble component formulas and inability to meet the application of some dosage forms.
[0006] Based on this, it is very important to develop a stable water-soluble plant extract composition that can repair the skin barrier by improving the damaged skin physical barrier. Summary of the invention

[0129] 1. Cytotoxicity evaluation of burdock extract and combined extracts of gentian, stephania tetrandra and centella asiatica
[0130] 1.1 Cell culture
[0131] Human immortalized keratinocytes (HaCaT) were cultured in 90% DMEM + 10% fetal bovine serum (FBS) + 1% penicillin-streptomycin double-antibody culture medium (hereinafter referred to as DMEM complete culture medium) in a 37°C incubator containing 95% air and 5% carbon dioxide. The culture medium was replaced every two days. After the cell confluence reached 70-80%, the cells were digested with trypsin-EDTA digestion solution (0.25%) for the next experiment.
[0132] 1.2 Cytotoxicity test
[0133] HaCaT cells were cultured at 1.5 × 10 5Cells / mL were inoculated in a 96-well plate, 100 μL of cell suspension was added to each well, and the cells were cultured in a 37°C incubator for 24 hours before attachment. Components a and b were dissolved in DMSO in the sample group and diluted to the concentration to be tested with DMEM complete medium before adding to the well plate. The control group was replaced with an equal amount of complete medium and the cells were incubated for 24 hours. 10 μL of CCK-8 solution was added to each 100 μL of culture medium and placed in a cell culture incubator for 2 hours. The cytotoxicity of each sample can be determined by comparing the absorbance of the sample group and the control group at a wavelength of 450 nm.
[0134] The calculation formula is as follows: cell viability (%) = sample group OD value / control group OD value*100%. It is generally believed that when the cell viability is greater than 80%, the sample has no obvious toxicity to the cells.
[0135] Experimental results: The effect of component a (burdock extract) on HaCaT cell viability is shown in Figure 1 The results of the effect of component b (combined extracts of Gentiana, Stephania tetrandra and Centella asiatica) on the viability of HaCaT cells are shown in Figure 2 .
[0136] The experimental results show that when the mass concentration of component a (burdock extract) is 500 μg / mL or less, the survival rate of HaCaT cells is 80% or more, and when the mass concentration of component b (combined extract of gentian, stephania tetrandra and Centella asiatica) is 250 μg / mL or less, the survival rate of HaCaT cells is close to 100%, indicating that components a and b have no toxic effects on HaCaT cells within the above mass concentration range and do not affect cell growth.
[0137] 2. Evaluation of repair efficacy
[0138] 2.1 Establishment of UVB-induced barrier damage cell model
[0139] HaCaT cells were cultured at 1.5 × 10 5 Cells were inoculated at a density of 100 cells / mL in a 12-well plate, and 1 mL of cell suspension was added to each well. After culturing in a 37°C incubator for 24 hours, the cells adhered to the wall, and the culture medium in the well was discarded and replaced with 200 μL PBS. A blank group, UVB model group, and sample group were set up, with 2 replicate wells in each group. After covering the blank group with aluminum foil, the well plate was placed in a UV irradiator (VILBER LOURMAT) and irradiated with UVB at a wavelength of 312 nm at 0.15 J / cm 2 After irradiation, each well was washed twice with PBS, 1 mL of DMEM complete medium was added to each well of the blank group and UVB model group, and the corresponding samples of the embodiment and the comparative example were added to the sample group, and the culture was continued in a 37°C incubator for 3 h.
[0140] 2.2 Quantitative analysis of gene expression levels
[0141] (1) An in vitro UVB-induced barrier damage cell model was constructed according to the method in 1.2, and samples were added and cultured for 3 h according to the two group ratios in the examples and comparative examples in Table 2;
[0142] (2) RNA was extracted using the SteadyPure RNA extraction kit (ACCURATE BIOLOGY), and RNA was quantified using a NanoDrope2000 micro-volume UV spectrophotometer (Thermo Fisher).
[0143] (3) After adjusting the RNA concentration, the RNA was reverse transcribed into cDNA using the Evo M-MLV reverse transcription kit (ACCURATE BIOLOGY) for fluorescence quantitative qPCR testing.
[0144] (4) RT-qPCR test was performed using SYBR Green Pro Taq HS premixed qPCR kit (ACCURATE BIOLOGY). GAPDH was used as the internal reference gene to calculate the relative expression levels of filaggrin (FLG), zonula occludens-1 (ZO-1), and tight junction protein-1 (CLDN1) mRNA. The expression levels of the target genes in Examples 1-4 were compared with those in the UVB model group and Comparative Examples 1-4. The target gene primer sequences are shown in Table 3.
[0145] Table 3: Target gene sequences
[0146]
[0147] 2.3 Experimental results:
[0148] Depend on Figure 3-Figure 5It can be seen that after UVB irradiated HaCaT cells, the expression of cell FLG, ZO-1 and CLDN1 mRNA decreased significantly, indicating that the UVB-induced barrier damage cell model was successfully constructed. Component a (Comparative Example 1) alone only has an improvement effect on ZO-1 mRNA expression; component b (Comparative Example 2) alone has an improvement effect on FLG mRNA, but no improvement effect on ZO-1 and CLDN1 mRNA expression, indicating that component b can be used alone to regulate epidermal filament protein, thereby repairing the skin physical barrier. Component a and component b are combined in different proportions in Table 2. When the ratio range is 30:1-10:1, the expression of the target gene mRNA in Comparative Examples 3 and 4 is not significantly improved compared with the use of two components (Comparative Examples 1 and 2) alone; and when the ratio range of the two components is 4:1, 1:1, 1:4 and 1:10, Examples 1-4 have significant improvements in the expression of FLG, ZO-1 and CLDN1 mRNA (p<0.05). Other embodiments also have similar effects. Therefore, this study demonstrated that the composition of the present invention can achieve a synergistic effect on enhancing the mRNA expression levels of FLG, ZO-1 and CLDN-1 proteins, and further demonstrated that when component a (burdock extract) and component b (combined extracts of gentian, stephania tetrandra and Centella asiatica) are in the range of a compound ratio of 6:1 to 1:12, especially 4:1-1:10, they have a synergistic effect on repairing the skin's physical barrier.
