Abstract
Description
Technical Field
[0001] The invention relates to the technical field of veterinary medicines, and in particular to a Tibetan veterinary medicine composition for treating spleen deficiency type diarrhea in weaned yaks. Background Art
[0002] Yaks are the main source of income in high-altitude and cold regions, and yak breeding is extremely important for agricultural and pastoral areas. Diarrhea is one of the main diseases that plague the normal growth of yaks. Breeders should attach great importance to it, do a good job in the prevention and treatment of diarrhea, and ensure the healthy growth of yaks. After birth, yaks are prone to low immunity and digestive system disorders, which makes yaks vulnerable to external factors and cause diseases. Young yaks need to obtain the nutrients needed for growth from breast milk to ensure the growth of the body and the improvement of immunity. If the number of immunoglobulins in milk is low, the yak's own immunity will not be improved. Once infected, diarrhea will occur. After weaning, due to the immature development of various digestive organs and the lack of a complete digestive system, its digestive ability is weak. Even if excessive milk is consumed, it will increase the burden on the intestines. The milk that cannot be absorbed in time in the intestines will continue to ferment and cause diarrhea.
[0003] Diarrhea due to deficiency of spleen and stomach in weaned yaks is a common and frequently occurring disease, which is mainly manifested by persistent diarrhea, dehydration, slow production and high mortality in young weaned yaks, seriously affecting the healthy and sustainable development of yak breeding. At present, the prevention and treatment of diarrhea due to deficiency of spleen and stomach in weaned yaks mainly rely on the intervention of drugs, especially chemical drugs and herbal drugs, but both have certain shortcomings and deficiencies. For example, although the single use of herbal drugs such as traditional Chinese medicine has good safety and effectiveness, the preparation process is complicated, the ingredients are unstable, the administration method is inconvenient, the acceptance is low, the efficacy is slow, and it is difficult to meet emergency needs; although chemical drugs have faster efficacy and higher acceptance, they have large side effects and are prone to drug resistance, affecting spleen and stomach function and increasing drug residues, endangering the health and safety of weaned yaks while affecting food safety.
[0004] Based on the above analysis, the art urgently needs a new method for preventing and treating yak spleen and stomach deficiency and coldness, and provides a safe, effective, convenient and economical prevention and treatment preparation to solve a series of problems caused by spleen and stomach deficiency and coldness diarrhea in weaned yaks. Finally, the liquid is divided into 100mL polypropylene plastic bottles and sterilized at 100℃ for 15min for standby use.
[0049] 2.2 Acute toxicity test of oral solution of combined composition—modified Koch method
[0050] First, a preliminary test was conducted on the experimental animals to explore the experimental dose range and find out the estimated lethal dose that causes 0-100%. Then, a formal test was conducted: 50 mice weighing 20±2g, half male and half female, were randomly divided into 10 groups, 5 mice / group. Kunming mice were randomly divided into groups, 5 mice in each group, and after fasting for 16 hours, they were gavaged with 500-5000mg·Kg-1 of the test drug in the dose gradient of 500mg·Kg-1. The control group was given an equal amount of physiological saline, and the reaction of the mice within 24 hours was observed and recorded to make a preliminary evaluation of acute toxicity. Statistical calculations were performed according to the modified Coulthard method based on the survival of the animals.
[0051] 2.3 Evaluation of the taste of oral liquid composition - Determination of preference index
[0052] The preference index, also known as the preference degree (TGI, Targeted Group Index), is mainly used to measure the difference between the attention degree of the target group under a specific feature and the overall level. This study used a two-bottle preference test method to evaluate the mice's preference for experimental oral solution and water: before the experiment, the mice were trained to drink water to adapt to the environment and become familiar with drinking water. In the formal experimental stage, the mice were placed in the experimental device and given oral solution of experimental drugs and pure water at the same time. During the experiment, the drinking behavior data of the mice were recorded in real time and the oral solution preference index (oral solution consumption / total water intake × 100%, calculated as a percentage) was calculated. In the experiment, there were 20 Kunming mice in each oral solution group and pure water group, each group was divided into 5 cages, and each had 4 replicates.
[0053] 2.4 Verification of the Rhubarb + Senna Spleen Deficiency Diarrhea Model in Mice - Spleen Strengthening Effect
[0054] 42 BAB / c mice (18±2g) were randomly divided into 6 groups. After acclimation for 3 days, except for the negative control group which was given normal saline, the other groups were gavaged with rhubarb + senna leaf decoction (0.5g+0.5g / 10g) for 8 consecutive days to replicate the spleen deficiency diarrhea model. The weight of mice in each group was recorded after the last gavage. The mice were fasted for 16 hours before the experiment. The high, medium and low drug dose groups were given 720, 600 and 500 mg·Kg respectively. -1 (Calculated as crude drug), the model group and negative control group were given normal saline, and the positive control group was given 60 mg·Kg -1 Guipi Pills were administered once a day for 7 consecutive days in each group. The weight of mice was recorded again before the last administration. 1 hour after the last administration, each mouse was given 0.2 mL of ink, and 30 minutes later, the mouse was killed by dislocation. The abdominal cavity was quickly opened and the small intestine, thymus and spleen were taken to calculate the intestinal propulsion rate, thymus index and spleen index. During the experiment, the diarrhea of mice in each group was recorded at the same time, including the loose stool rate, loose stool grade and diarrhea index.
[0055] 2.5 Evaluation of clinical effects of anti-diarrhea
[0056] 2.5.1 Clinical treatment trial of weaned yaks
[0057] 49 weaned yaks with diarrhea were selected and randomly divided into a test group (25 yaks) and a control group (24 yaks). The combination oral solution and the control drug were used for treatment respectively. Dosage regimen: combination oral solution (120 mg·Kg -1 , calculated according to the dosage of crude drug for oral solution preparation), 2 times / d, for 5 consecutive days; Guipi Pills (60mg·Kg -1 ), 2 times / d, for 5 consecutive days.
[0058] 2.5.2 Clinical prevention trial in weaned yaks
[0059] 47 weaned yaks with diarrhea were selected and randomly divided into a test group (24 yaks) and a control group (23 yaks). The oral solution of the composition and normal saline were used respectively. Dosage regimen: oral solution of the composition (120 mg·Kg -1 ,, calculated according to the dosage of crude drug for oral solution preparation), 2 times / d, for 5 consecutive days; the control group was given normal saline. Observe whether diarrhea occurs during medication and within 3 days after stopping medication, and calculate the protection rate.
[0060] 2.5.3 Growth promotion test
[0061] 54 healthy weaned yaks were selected and randomly divided into an experimental group (27 yaks) and a blank control group (27 yaks). The dosing regimen of the experimental group was the same as 2.5.1, and it was used for 21 days; no drug was added to the blank control group. The incidence of diarrhea in weaned yaks during the medication period was observed, and the feed intake and body weight on the weaning day, 7th day, 14th day and 21st day after weaning were recorded.
[0062] Experimental weaning day, weaning
[0063] 3. Test results
[0064] 3.1 Optimal ratio of the composition and the principle of compatibility of monarch, minister, assistant and envoy
[0065] According to the results of the implementation case, the optimal formula ratio was determined to be: 25:20:15:25:10:15:15:40, namely: 25 parts of single-flowered delphinium, 20 parts of fennel, 15 parts of spatholobus, 25 parts of wild onion, 10 parts of Polygonum capitatum, 15 parts of small drooping chrysanthemum, 15 parts of Artemisia suspensa (processed), and 40 parts of sludge. In the formula, single-flowered delphinium, fennel, and spatholobus are the main drugs, wild onion and Artemisia suspensa (processed) are the ministerial drugs, Polygonum capitatum and small drooping chrysanthemum are the adjuvant drugs, and sludge is the guiding drug. The single-flowered Delphinium flower in the Tibetan veterinary medicine composition and oral liquid warms the stomach and intestines and stops diarrhea; the Tibetan fennel warms the middle and dispels cold, strengthens the spleen and stomach; the spoon-leaved Nardostachys chinensis warms the stomach and strengthens the spleen; the Artemisia selengensis (processed) dispels cold and dampness, dissipates blood stasis and relieves pain; the wild onion strengthens the spleen and stimulates appetite, aids digestion, etc.; the Polygonum capitatum is used for cold accumulation and chronic diarrhea, nausea and vomiting; the small drooping chrysanthemum calms the wind and relieves heat, promotes diuresis and reduces swelling; the sludge element is astringent and stops diarrhea, and is used for enteritis and dysentery, etc.
[0066] 3.2 Preparation of oral solution
[0067] According to the implementation case 1 in the preparation process of oral solution 2.1, the composition is added to the deionized water in a ratio of 1:10, soaked for 1 hour, and then decocted in a ceramic Chinese medicine pot with high heat until boiling, and then decocted for 30 minutes with low heat. After filtering the liquid, deionized water is added in a ratio of 1:5 and the above steps are repeated for 1 time. After filtering, the filtrate is concentrated to a certain volume in a rotary evaporator, and anhydrous ethanol is added to the concentrated liquid by volume until the final alcohol content is 75%. After standing overnight, the supernatant is filtered out, and the ethanol is recovered by a rotary evaporator until there is no alcohol taste and the liquid is concentrated to a final concentration of 1g / mL. 1mol / L sodium hydroxide and 1mol / L hydrochloric acid solution are used to adjust the pH value to 7.4, and Tween-80 (cosolvent), stevioside (flavor corrector) and sodium benzoate (preservative) are added according to the volume ratio to a final concentration of 0.3%, 0.4% and 0.5%, respectively. Finally, the drug solution was divided into 100 mL polypropylene plastic bottles and sterilized under high pressure at 100°C for 15 min for later use.
[0068] 3.2 Evaluation of acute toxicity of oral solution
[0069] Acute toxicity tests showed that mice in each experimental group did not show normal behavioral changes or death, and the LD50 of the oral solution could not be measured, which preliminarily confirmed that the screened composition and oral solution were safe and non-toxic.
[0070] 3.3 Oral liquid taste evaluation - TGI index
[0071] The two-bottle preference test method was used to evaluate the preference of mice for the experimental oral solution and water. The experimental results are shown in Table 1. The oral solution preference index was 1.08 (31.87% / 29.39%), indicating that the oral solution had good palatability, no peculiar smell, and was highly accepted by mice.
