Abstract
Description
[Technical field]
[0001] CROSS-REFERENCE TO RELATED APPLICATIONS This application claims priority from U.S. Provisional Patent Application No. 63 / 316,496, filed March 4, 2022, the contents of which are incorporated by reference in their entirety herein.
[0002] The present disclosure relates to fragrance compositions, and more particularly to musk-containing fragrance compositions that induce improved sleep in a subject upon exposure to the fragrance composition. The present disclosure also relates to consumer products containing such fragrance compositions and methods of use thereof. [Background technology]
[0003] Insufficient sleep and / or poor quality sleep is a modern challenge that negatively impacts health and social productivity worldwide. It is reported that 84% of the world's population has experienced some form of sleep difficulty. The US CDC (Centers for Disease Control and Prevention) recommends that adults aged 18 and older should get at least 7 hours of sleep per night, as good quality sleep correlates with many benefits, such as better mood, appearance, immune health, and cognitive / physical function (e.g., focus, memory, etc.). Unfortunately, in recent years (especially during the COVID pandemic), there has been a sharp increase in the number of people reporting issues with insufficient sleep and / or poor quality sleep. In fact, a study published in the Harvard Business Review found that lost productivity due to sleep-deprived employees costs four companies a combined US$54 million per year.
[0004] Common ways to address this problem include taking prescription (e.g., zolpidem, sold as Ambien) and / or over-the-counter (OTC) medications (e.g., melatonin supplements). Problems with this solution may include: (i) the real risk of abuse and addiction to the sleep aid Ambien, (ii) harmful side effects, (iii) limited availability of melatonin due to differences in regulatory systems (e.g., melatonin sold OTC in the United States is available only by prescription in other countries), and / or (iv) the tendency of these pharmaceutical solutions to address only specific sleep problems (e.g., Ambien treats insomnia, while melatonin promotes sleep-wake cycles).
[0005] Another approach involves the use of aromatherapy solutions to affect the sleep patterns of consumers. Commonly used scents include lavender, rose, ylang-ylang, geranium, clary sage, neroli, marjoram, etc., which are widely known as essential oils with sedative effects (JP Patent No. 5019016B2, Takasago International Corporation). However, the drawbacks of these natural oils may include: (i) they each have a characteristic / unique scent, but the preferences tend not to be shared among different consumers, which may cause others in the vicinity to feel disgust or irritation, etc., and / or (ii) the essential oils tend to have different effects. U.S. Patent Application Publication No. 2021 / 0008082A1 (Takasago International Corporation) discloses a fragrance composition useful for improving sleep, comprising at least one floral compound and at least one musk compound selected from Ambroxan®, Ambretone®, L-Muscone, Habanolide®, and Musk T®. US 2020 / 0048578 A1 (Givaudan SA) describes a method for preparing a fragrance composition adapted to address sleep quality, the composition containing 0.01-50% by weight of a number of fragrance ingredients, including at least one musk component (B15) selected from ambrette, Cashmeran®, Cosmone®, cyclopentadecanolide, ethylene brassylate, Galaxolide®, Galaxolide S, Habanolide®, muscenone, Musk C14, Serenolide®, Silvanone® Supra, Sylkolide® and Velvione®. These attempts have encouraged the indiscriminate use of such musk compounds without considering the type of musk compound and / or their impact on sleep benefits. Thus, while these attempts disclose the use of musk compounds in fragrance compositions to obtain sleep quality, they do not teach the specific types of musk compounds or the amounts of these types of musk compounds that should be included to provide the benefit of improving the quantity and / or quality of sleep. Summary of the Invention [Problem to be solved by the invention]
[0006] Therefore, there remains a need to identify fragrance compositions that effectively improve the quantity and / or quality of sleep of individuals, particularly in a safe, non-pharmacological and / or non-invasive manner. It is desirable to have fragrance compositions that can be used in consumer products to improve the sleep benefits of subjects. The present disclosure addresses these and other needs, as will be disclosed in more detail below. [Means for solving the problem]
[0007] The present invention is based on the discovery that a combination of musk compounds, particularly heteroatomic musk compounds and / or acyclic musk compounds, can be formulated into a fragrance composition (optionally containing MCM and linear musk compounds and / or additional heteroatomic musk compounds) that improves sleep in a subject, and preferably enhances at least one sleep benefit. It has also been found that compounds with specific backbone structures are useful for improving sleep. For example, compounds containing such backbones can improve one or more of the sleep benefits. Such compounds can be formulated into a fragrance composition that improves sleep. Furthermore, the fragrance compositions of the present disclosure have been shown to improve sleep benefits in a novel olfactory direction (i.e., away from traditional lavender), have no adverse effects, and / or have high consumer acceptability when used alone or as part of a total sleep solution, for example, sprayed on a pillowcase as a nightly routine.
[0008] In one embodiment, the backbone structure of formula (I): [ka] wherein each open position of the backbone structure is independently optionally substituted; [ka] is a single bond or a double bond; and the six-membered ring of formula (I) has 0 to 3 double bonds. wherein the one or more compounds having the backbone structure of formula (I) are present in a total concentration of greater than 0.05 wt. % of the total weight of the composition.
[0009] In one aspect, there is provided a fragrance composition for improving sleep comprising one or more heteroatomic musk compounds, wherein the one or more heteroatomic musk compounds are present in a total concentration greater than 0.05 wt. % of the total weight of the composition.
[0010] In one embodiment, a fragrance composition for improving sleep is provided, comprising at least four fragrance ingredients, including (i) at least three heteroatom musk compounds; and (ii) at least one acyclic musk compound, 7,7,8,9,9-pentamethyl-6,7,8,9-tetrahydro-5H-cyclopenta[h]quinazoline or 2,2,6,6,7,8,8-heptamethyl-3a,5,6,7,8,8b-hexahydro-4H-indeno[4,5-d][1,3]dioxole. In one embodiment, a fragrance composition for improving sleep is provided, comprising at least four fragrance ingredients, including (i) at least three heteroatom musk compounds; and (ii) at least one acyclic musk compound or 7,7,8,9,9-pentamethyl-6,7,8,9-tetrahydro-5H-cyclopenta[h]quinazoline. The fragrance composition optionally comprises a solvent selected from dipropylene glycol (DPG), isopropyl myristate, ethanol, triethyl citrate, or combinations thereof.
[0011] In another aspect, the present disclosure relates to a consumer product comprising the fragrance composition described herein, the consumer product being an air care product, a home care product, a personal care product, a hygiene product, a pet care product, a fine fragrance, a hair care product, or a beauty care product. In some embodiments, the air care product containing the fragrance composition releases the fragrance composition in response to a biometric reading, for example, a consumer biometric reading. In this way, for example, the release of the fragrance composition is triggered by biometric data.
[0012] In yet another aspect, the present disclosure relates to a method of improving at least one sleep effect in a subject, comprising providing a fragrance composition as described herein or a consumer product as described herein to the subject in an amount effective to improve at least one sleep effect. Preferably, the fragrance composition or consumer product is (i) sprayed or applied to the subject; (ii) sprayed or applied to furniture upholstery or fabrics, preferably bedding, laundry, blankets or pillowcases; and / or (iii) dispersed or diffused in the air.
[0013] Therefore, advantageously, the present disclosure provides novel design guidelines for formulating fragrance compositions intended to improve sleep. In particular, the design guidelines identify selected musk compounds intended to be incorporated into fragrance compositions for the purpose of providing one or more sleep benefits to a subject in need thereof by administering a fragrance composition disclosed herein in an amount effective to provide one or more sleep benefits to the subject in need thereof. In embodiments, the design guidelines identify compounds having the framework structures described herein for the purpose of providing a fragrance composition for the purpose of providing one or more sleep benefits to a subject in need thereof by administering a fragrance composition disclosed herein in an amount effective to provide one or more sleep benefits to the subject in need thereof. Yet another advantage is the ability to create novel fragrance profiles that improve sleep.

[0051] fragrance composition
[0052] By working with sleep experts to combine neuroscience, consumer perception and the use of IFF's proprietary artificial intelligence tools to improve the performance of biometric measurements in real environments, the applicant has surprisingly discovered a musk fragrance composition that can improve the quantity and / or quality of sleep and positively improve the lives of consumers suffering from sleep problems around the world. In particular, the applicant has surprisingly discovered that fragrance compositions that follow a set of design guidelines for containing specific types of heteroatomic musk and / or acyclic musk fragrance ingredients are useful for improving at least one or more sleep benefits. [0103] In some embodiments, the fragrance composition described herein may be blended with an existing accord, draft or finished fragrance. In some embodiments, the accord, draft or finished fragrance may not have a sleep-improving effect as described herein. In some embodiments, the fragrance composition for sleep improvement described herein may be blended with an accord, draft or finished fragrance that does not have a sleep-improving effect to obtain an accord, draft or finished fragrance with a sleep-improving effect. For example, the fragrance composition for sleep improvement described herein may be added to an existing accord, draft or finished fragrance to obtain an accord, draft or finished fragrance with a sleep-improving effect. Thus, in some cases, the fragrance composition for sleep improvement described herein may impart a sleep-improving effect to a fragrance that does not affect sleep.
[0104] In certain embodiments, the fragrance composition of the present disclosure is substantially free or essentially free of one or more musk compounds, provided that the one or more musk compounds do not include heteroatom musk compounds and / or acyclic musk compounds. In certain embodiments, the fragrance composition is substantially free or essentially free of macrocyclic musk compounds, preferably including Ambretone®, ambrettelide, Cosmone®, cyclopentadecanolide, ethylene brassylate, Habanolide®, L-muscone, muscenone, musk C14, Musk T®, Silvanone® Supra and / or Velvione®.
[0105] The perfume compositions of the present disclosure may contain one or more perfume co-ingredients, generally belonging to, but not limited to, the following chemical classes: alcohols, aldehydes, ketones, esters, ethers, acetals, oximes, acetates, nitriles, terpenes, saturated and unsaturated hydrocarbons of natural or synthetic origin.
[0106] Pure perfume. In certain embodiments, the perfume composition of the present disclosure comprises a pure perfume. "Pure perfume" refers to a perfume ingredient that is free of extraneous substances, is not encapsulated, and / or is not associated with other compounds that would delay the release of the perfume ingredient. When the perfume ingredient is a pure perfume, the perfume ingredient is present in an amount of 0.001% to 100% by weight based on the total weight of the perfume composition.
[0107] In some embodiments, the compound or compounds having the backbone structure of formula (I) are pure perfumes. In some embodiments, the heteroatom musk compound or compounds are pure perfumes. In some embodiments, the acyclic musk compound is pure perfumes.
[0108] Encapsulated perfume. In certain embodiments, the sleep benefits of the perfume composition of the present disclosure can be further improved by utilizing a delivery system for such perfume compounds. Typically, microcapsules are used to encapsulate perfumes and deliver them to target areas in a delayed or controlled manner. The term "encapsulated perfume" as used herein refers to perfume compositions encapsulated in microcapsules to stabilize the release of perfume compounds over an extended period of time. Without wishing to be bound by a particular theory, the applicant proposes that the perfume composition can be applied to pillowcases and / or bedding from various consumer products (e.g., fabric sprays, laundry detergents, fabric softeners, fragrance enhancers, etc.). Then, during the night, the subject rubs the pillowcase and / or bedding, causing the microcapsules to burst and the perfume material to be released from the microcapsules. As a result, the released perfume composition acts to improve the quality and / or quantity of sleep continuously throughout the night.
[0109] The average particle size of the microcapsules is preferably 1 to 100 microns, preferably 1 to 50 microns, more preferably 1 to 20 microns. Microcapsules can be prepared from natural materials such as mycelium chitosan (WO 2016 / 185171 A1), silk fibroin particles (US 2015 / 0164117 A1) and biomolecules used as emulsifiers in microcapsule preparation (WO 2016 / 193435 A1; WO 2017 / 102812 A1; US 2018 / 0078468 A1; WO 2018 / 019894 A1; WO 2018 / 019896 A1; and WO 2017 / 102812 A1). Multilayer coacervates of gelatin / gum arabic capsules can be used in the present invention (U.S. Pat. No. 4,946,624; WO 2012 / 001604 A1; U.S. Pat. App. Pub. No. 2015 / 0250689 A1; and WO 2018 / 002214 A1). Protein microcapsules are also useful within the scope of the present invention (U.S. Pat. App. Pub. No. 2017 / 0189283 A1). Environmentally friendly microcapsules, preferably with a shell biodegradability of at least 60% by weight within 60 days according to OECD 301F, are also useful within the scope of the present invention (WO 2021 / 122633 A1).
[0110] In some embodiments, one or more compounds having a skeletal structure of formula (I) are encapsulated. In some embodiments, one or more heteroatom musk compounds are encapsulated. In some embodiments, the acyclic musk compounds are encapsulated. In certain embodiments of the present disclosure, the perfume ingredients are pure perfumes (i.e., unencapsulated perfumes), encapsulated perfumes, or combinations thereof. One or more compounds having a skeletal structure of formula (I), one or more heteroatom musk compounds, and acyclic musk compounds can be considered as perfume ingredients of a perfume composition.

In some embodiments, the fragrance composition is used in conjunction with a smart device, such as a smart bed, a smart mattress, a smart pillow, a smart wearable device, etc., to provide the fragrance to the subject. In some embodiments, the smart device further collects data, such as biometric data from the subject, and uses the data to regulate (e.g., start, stop, increase, decrease) the delivery of the fragrance. The fragrance composition may be present in any form suitable for use in such a device.
[0123] In certain embodiments, the fragrance composition is administered to the subject in gaseous or volatile form. In certain non-limiting embodiments, the fragrance composition can be administered intranasally or by inhalation. In certain embodiments, the subject can inhale the fragrance composition directly or indirectly. In other specific embodiments, the fragrance composition can be released into the area around the subject, and the subject inhales the composition. In specific embodiments, the composition is released into the air by a consumer product, such as, but not limited to, an air freshener or a candle, or bedding and / or pillowcases treated with one or more fabric care products, such as fabric sprays, laundry detergents, fabric softeners, fragrance enhancers, etc. In some embodiments, the consumer product is a bedtime or sleep aid. [0128] How to monitor your sleep effectiveness
[0129] The sleep effect of the subject exposed to the fragrance composition can be monitored using, but not limited to, telemetry using the SleepScore Max device (SleepScore Labs, Carlsbad, California), a non-contact monitoring device that measures the subject's respiratory signal (i.e., breathing) and movement to detect sleep behavior. The sleep monitoring method of the present disclosure represents a significant advance in technology compared to conventional simple consumer-wearable sleep and health monitoring devices sold under the brand names Fitbit Inspire HR®, Fitbit Charge 5®, Fitbit Versa 2®, Fitbit Ionic®, Jawbone Up3®, OURA Ring Gen 3®, and other similar devices. A major drawback of these wearable sleep monitoring devices is that they may disrupt the subject's sleep patterns and / or may have limited sensors (e.g., heart rate sensors (i.e., sleep heart rate), motion detectors (indicators of restless sleep), etc.) and therefore may only provide minimal and / or vague sleep data and / or indicators. Conclusions drawn based on data generated from these devices may then not be particularly robust and / or may not properly support various sleep effects.
[0130] Therefore, the sleep monitoring of the present disclosure is considered to be comparable to polysomnography (PSG), the gold standard of sleep assessment technology. The SleepScore Max device has been shown to have high performance in terms of sleep / wake measurements (Chinoy, ED et al., Performance of Seven Consumer Sleep-Tracking Devices Compared with Polysomnography, Sleep J., 2021, pp.1-16). The sleep monitoring method of the present disclosure allows the measurement of overall sleep quality indicators such as time spent in each sleep stage (light, deep and REM), total sleep time, wake-after-sleep (WASO), number of awakenings or number of awakening episodes, sleep onset latency, total wake time, time in bed, start time of sleep habit, end time of sleep habit (wake-up time), and sleep efficiency, sleep maintenance, sleep score, mental score and physical score.

To make the pillow spray applications, each fragrance composition (Fragrance Example 1 or Example 2) was first mixed with isopropyl myristate (IPM) and then mixed with PEG-4 hydrogenated castor oil according to the ratios specified in Table 4. The remaining ingredients were then added to make the final formulation. For the fragrance samples, each subject was given 20 mL of the formulation in a 2 oz. white HDPE plastic bottle (supplied by Brad Pak Enterprises, Garwood, NJ) with a fine mist spray cap. Subjects also received an unscented sample in a similar bottle. Both samples were blind coded for testing.
[0140] [Table 4]
[0141] All fragrance compositions used in the study were reviewed by the IFF's Global Regulatory Affairs (GRA) and approved as safe for use in human subjects at the specific dose of 0.5% in pillow mist applications.
[0142] Example 2: Improvement of sleep using a fragrance composition
[0143] The purpose of this example is to demonstrate the sleep benefits of using the fragrance composition of the present disclosure. Two sleep studies were conducted. Study participants were 18-60 years old, generally healthy, self-reported mild sleep difficulties due to lifestyle factors, but had no clinical records of sleep disorders. Additionally, participants did not have irregular sleep patterns or schedules. Study 1 (using fragrance example 1) included 51 participants, including 27 women and 24 men, ages 22-55. Study 2 (using fragrance example 2) included 52 participants, including 29 women and 23 men, ages 20-57. Participants did not take any type of sleep aid or medication that would affect sleep during the study period and for three weeks prior to the start of the study. The study was conducted in the participants' own bedrooms, and participants were encouraged to maintain their habitual sleep schedule throughout the study period. All participants provided fully informed consent prior to participation, and all study procedures, including participant recruitment, were reviewed and approved by the Western Institutional Review Board (WIRB).
[0144] All participants were provided with a SleepScore™ Max device (SleepScore Labs, Carlsbad, California) along with setup instructions and anonymized user account. All participants were asked to download the SleepScore™ Max application from the Apple® Application Store onto their smartphone and then pair the SleepScore™ Max application with the SleepScore™ Max device. Once paired, the device was placed 1.3 to 3.9 feet (i.e., arm's length) away from the participant. The bottom of the device should be just above the top surface of the mattress, with nothing between the device and the participant. To ensure minimal interference, no objects with moving parts (e.g., electric fans) were placed near the device. The bedroom was maintained at room temperature (i.e., 25-28°C) and relative humidity of 38-85%. The SleepScore™ Max device was used to monitor the participants' sleep behavior. Nightly summary data from the system was downloaded using the SleepScore™ data visualization platform.
[0145] The study design is shown in Figure 1. An acclimation period of 3 nights / days was conducted at the beginning of the study to acclimate participants to the fragrance examples, unscented control, and application regime. Each study began on a given Monday of the week and ran for a total of 3 weeks, which was divided into three periods: (i) baseline period ("BL"; week 1); (ii) product use period 1 ("PUP1"; week 2); and (iii) product use period 2 ("PUP2"; week 3). The order in which the unscented control and scented samples were presented was counterbalanced across participants.
[0146] On a given night during the study, participants were asked to spray the entire pillowcase with the spray three times just before bedtime and to complete the questionnaire shown in FIG. 2. Before lights out, participants were asked to start sleep tracking via the SleepScore™ Max application on their smartphone. Participants were asked to stop sleep tracking immediately after getting out of bed in the morning. Participants were also asked to complete the morning questionnaire shown in FIG. 3 after fully waking up and before starting their morning routine. All questionnaires were completed electronically via web-based data collection software (Compusense®). The deadline for completing the pre-bedtime questionnaire was set at 2:00 a.m., and the deadline for completing the morning questionnaire was set at 12:00 noon, after which participants were not allowed to complete the questionnaire. Participants were not allowed to view previous days' responses when completing the questionnaire. Additionally, participants were asked to complete the questionnaire based on their own sleep experience without reference to the sleep measurements displayed in the SleepScore™ Max application. Participants were also asked to complete the Product Use Questionnaire, shown in Figures 4A and 4B. The Product Use Questionnaire explored participants' experiences with products they had used the previous night.
[0147] Continuing with reference to FIG. 1, during the BL period, participants were asked to track their sleep using a SleepScore™ Max device and to complete questionnaires before bedtime and in the morning. During PUP1 and PUP2 periods, participants were asked to track their sleep using a SleepScore™ Max device and to complete questionnaires before bedtime and in the morning. Participants were also asked to use control example 1 or fragrance examples 1 and / or 2 according to the instructions in FIG. 1. The first three nights (Friday-Sunday) were considered as acclimation nights. The PUP period lasted from Friday night to Tuesday night of the following week. The study was conducted in a blinded manner so that participants did not know whether they were receiving the control example or the fragrance example.
[0148] Prior to statistical analysis, the study results were processed to remove outliers. All data from the habituation night were excluded from statistical analysis. Nightly objective sleep data as well as self-reports before bedtime and in the morning were analyzed in R (version 4.1) using multilevel regression analysis to account for the nested structure of the data (night per subject). One-sided tests were performed at the α=0.05 level. The following regression model: Sleep parameters ij = constant 0i +β×Test period ij A log-based regression was used to code observations during control or baseline use as 0 and observations during nights during active scent use as 1. Both self-reported and objective data were analyzed for participants' reports of their sleep and previous night's use, which were tracked during the scent example and control use periods with full adherence to the instructions given, including within-subject comparisons by night or day. Secondary analyses were also performed using the same methodology, except that BL was used as the control condition. When distributions of sleep onset latency (SOL) and / or wake after sleep onset (WASO) quantities were skewed and did not meet the normality assumptions required for statistical tests, log-transformations were performed prior to analysis.
[0149] Table 5 summarizes the biometric parameters measured using the SleepScore™ Max device for Study 1. The results show that participants exposed to Fragrance Example 1 of the present disclosure had improved sleep quantity and / or quality compared to Control Example 1. Similar results were observed for self-reported indicators (data not shown).
[0150] [Table 5]
[0151] The results shown in Table 5 indicate that exposure to Fragrance Example 1 has a beneficial effect on sleep. When exposed to Fragrance Example 1, participants fell asleep in less time (-2.7 minutes, -19%, p<0.01) and experienced fewer nighttime awakenings (-4.9 minutes, 19%, p<0.05) compared to Control Example 1. This resulted in improved sleep efficiency (+1.8%, p<0.01) and sleep maintenance (+1.1%, p<0.05). There was also an increase in the amount of time participants spent in deep sleep, both absolute (+4.9 minutes, +6%, p<0.05) and relative (+1.5%, p<0.05%). This resulted in an overall improvement in sleep scores (+2, +3%, p<0.05) and physical scores (+1.8, +2%, p<0.05) observed using Fragrance Example 1 compared to Control Example 1.
[0152] Table 6 summarizes the biometric parameters measured using the SleepScore™ Max device in Study 2. Table 7 summarizes the self-reported measures in Study 2. The results show that participants exposed to Fragrance Example 2 of the present disclosure had improved sleep quantity and quality compared to Control Example 1.
[0153] [Table 6]
[0154] [Table 7]
[0155] The results, shown in Table 6, indicate that exposure to Fragrance Example 2 had a beneficial effect on sleep. Participants fell asleep in less time (-2.4 minutes, -15%, p<0.05). As a result, participants exposed to Fragrance Example 2 reported an overall improvement in sleep quality (+0.3, +4%, p<0.05) as well as feeling more rested (+0.3, +4%, p<0.01) and more refreshed in the morning (+0.2, +4%, p<0.05) compared to Control Example 1.
[0156] Example 3: Sleep Improvement with Exemplary Fragrance Compositions
[0157] The impact of a third exemplary fragrance, Fragrance Example 3 (see Table 8, formulated as described in Example 1), on sleep was evaluated according to the methods described in Examples 1 and 2. A dose of Fragrance Example 3 was tested in a pillow spray at half the dose used to test Fragrance Examples 1 and 2 (see Example 1) and compared to an unscented control pillow spray (Unscented Example 2, see Table 10). The study enrolled 51 participants, including 32 women and 19 men, ages 20-60.
[0158] [Table 8]
[0159] [Table 9]
[0160] [Table 10]
[0161] Table 11 summarizes the biometric parameters measured in this study using the SleepScore™ Max device. The results shown in Table 11 indicate that lower doses of the exemplary compounds described herein, such as those in Examples 1 and 2, may also beneficially affect sleep.
[0162] [Table 11]
[0163] Exposure to fragrance example 3 improved the physical score compared to unscented example 2 (+1.8, +2%, p<0.05). This suggests that participants' deep sleep time increased. Table 11 shows that participants' deep sleep time showed an increasing trend.
[0164] Fragrance Example 2 (see Table 2) was also tested according to the method described in Example 2 at 0.11% by weight pillow spray (half the dose tested in Examples 1 and 2; see Table 4) and compared to Control Example 1. The study enrolled 32 participants, including 24 women and 8 men, ages 23-60. The results are shown in Table 12.
[0165] [Table 12]
[0166] As shown in Table 12, even low doses of Fragrance Example 2 (compared to the doses tested in Examples 1 and 2) were found to improve sleep efficacy, increasing REM sleep time, sleep efficiency, and sleep maintenance. Increased sleep scores and mental scores demonstrated an overall improvement in sleep quality. Consistent with the objective measures, participants also reported fewer interruptions, improved sleep quality, and a sense of refreshment even at lower doses of Fragrance Example 2 (p<0.05 for all self-report measures).
[0167] These data demonstrate that combinations of the exemplary compounds described herein improve sleep over a wide range of concentrations.
[0168] Example 4: Robustness of sleep-improving compositions
[0169] The ability of a fragrance composition for improving sleep to maintain its sleep-improving effect when formulated with additional ingredients that do not have a sleep benefit was evaluated.
[0170] First, an existing exemplary finished fragrance (EFF1) containing 7,7,8,9,9-pentamethyl-6,7,8,9-tetrahydro-5H-cyclopenta[h]quinazoline was tested according to the method of Example 2 to determine whether it had any effect on sleep. EFF1 was formulated into a pillow spray as shown in Table 13. This resulted in a concentration of 7,7,8,9,9-pentamethyl-6,7,8,9-tetrahydro-5H-cyclopenta[h]quinazoline in the pillow spray of 0.00475% by weight. The study included 54 participants, including 27 women and 27 men, ages 21-60.
[0171] [Table 13]
[0172] Table 14 summarizes the parameters for EFF1 and Control 1 that were biomeasured using the SleepScore™ Max device.
[0173] [Table 14]
[0174] The results shown in Table 14 indicate that EFF1 does not have a beneficial effect on sleep. Next, the effect on sleep when EFF1 is combined with Fragrance Example 1 was evaluated.
[0175] Perfume Example 1 described in Example 1 (see Table 1) was formulated into a pillow spray with EFF1 as shown in Table 15 to form a mixed composition (MXC).
[0176] [Table 15]
[0177] The sleep study was performed similarly to that described in Example 2. The study included 31 participants (2 men and 29 women) aged 18-60 years who self-reported mild sleep difficulties due to lifestyle factors but had no clinical records of sleep disorders. Participants had a normal sense of smell, no allergies, and were not taking medications that affected sleep. Additionally, participants did not have irregular sleep patterns or schedules. Participants did not take any type of sleep aid or medication that affected sleep during the study period or for 3 weeks prior to the start of the study.
[0178] The study design is shown in Figure 5. An acclimation period of 3 nights / days was conducted at the beginning of the study to acclimate participants to MXC or unscented control (Control 1) and application regime. The study was conducted over a 2-week period, with the first 3 days of each week being used for acclimation to MXC or Control 1. Sleep measurements were conducted over the last 4 days of each week. The order of presentation of unscented control and MXC samples was counterbalanced across participants.
[0179] Table 16 summarizes the parameters for MXC and Control 1 as biomeasured using the SleepScore™ Max device.
[0180] [Table 16]
[0181] As shown in Table 16, the addition of Fragrance Example 1 to EFF1 improved a wide range of sleep parameters, including increased total sleep time and REM sleep time. Increased sleep and mental scores demonstrated an overall improvement in sleep quality. Consistent with the objective measures, participants self-reported fewer nighttime interruptions (-0.3, -18%, p<0.05) and longer sleep times (+16.8 min, +4.2%, p<0.01).
[0182] These data indicate that fragrances with sleep-improving properties remain robust when combined with fragrances that do not have sleep-improving properties. Thus, the compounds and compositions described herein can be combined with ingredients that do not exhibit sleep-improving properties to produce fragrance compositions for sleep improvement.
