Description

Technical Field

[0001] The present invention belongs to the field of biomedicine and biotechnology, and relates to an active ingredient delivery system for a new type of lipid compounds. Background Art

[0002] Nucleic acid drugs are various oligoribonucleotides (RNA) or oligodeoxyribonucleotides (DNA) with different functions. They can regulate genes expressing related proteins based on the principle of base complementarity, and can directly act on pathogenic target genes or target mRNAs to play a role in treating diseases at the genetic level. The sequence design of nucleic acid drugs is very easy, and it can also avoid the limitation of non-drugable targets faced by traditional small molecule drugs and antibody drugs. In addition, they can play a regulatory role on intracellular and extracellular and cell membrane proteins. However, the development of nucleic acid drugs mainly faces three major problems: 1. Instability of nucleic acid molecules, especially RNA, in vivo; 2. Potential side effects; 3. Difficulties in drug delivery systems, etc. With the development of new technologies, some problems have been better solved, among which breakthroughs in chemical modification and delivery system technology have played a vital role in the development of nucleic acid drugs. Although chemical modification can improve the stability and immunogenicity of nucleic acid drugs, nucleic acid drugs need to enter cells to work. Because nucleic acid drugs have a large molecular weight and are usually negatively charged, the efficiency of cell uptake and endosome escape is relatively low, so they need to rely on the power of the delivery system.

[0003] Lipid nanoparticles (LNPs) are a new type of nucleic acid biomolecule delivery technology. LNPs are usually composed of four components: (1) ionizable lipids, which can self-assemble with mRNA into virus-sized particles and release mRNA from endosomes into the cytoplasm; (2) PEGylated lipids, which can increase the half-life of LNPs in the blood; (3) cholesterol, which can increase the stability of nanoparticles; and (4) phospholipids, which help form lipid bilayer structures. LNPs can protect mRNA from being decomposed by RNases and protect mRNA molecules from being recognized by TLRs, thereby avoiding overactivation of the innate immune system; and ionizable lipids can promote cellular uptake and help drug molecules escape from endosomes to achieve therapeutic effects.

[0004] At present, the LNP delivery system is still regarded as one of the key technologies to promote the entry of nucleic acid drug molecules into therapeutic applications. Lipid compounds with novel structures and high efficiency and stable delivery performance are still to be developed. Summary of the invention

[0005] Based on this, it is necessary to detect novel ionized lipid compounds in response to existing technical problems to improve delivery efficiency and stability.

[0006] The present invention provides a novel lipid compound, a synthesis method thereof, and a mixed encapsulation of drug molecules with a PEGylated lipid compound, a structural lipid cholesterol, and a natural phospholipid to form a nanoparticle delivery system, which can be used for in vitro cell delivery and in vivo organ-targeted cell delivery, as follows:

[0099] The term "heteroaryl" refers to a group of a monocyclic or bicyclic 4n+2 aromatic ring system (e.g., having 6 or 10 π electrons shared in a cyclic arrangement) having ring heteroatoms, wherein each heteroatom is independently selected from nitrogen, oxygen and sulfur. In heteroaryl groups containing one or more nitrogen atoms, the point of attachment can be a carbon or nitrogen atom as long as the valence permits. Heteroaryl bicyclic ring systems can include one or more heteroatoms in one or both rings. Heteroaryl also includes ring systems in which the above-mentioned heteroaryl rings are fused to one or more cycloalkyl or heterocyclic groups, and the point of attachment is on the heteroaryl ring, in which case the number of carbon atoms continues to represent the number of carbon atoms in the heteroaryl ring system. Exemplary 5-membered heteroaryl groups containing one heteroatom include, but are not limited to, pyrrolyl, furanyl and thienyl. Exemplary 5-membered heteroaryl groups containing two heteroatoms include, but are not limited to, imidazolyl, pyrazolyl, oxazolyl, isoxazolyl, thiazolyl and isothiazolyl. Exemplary 5-membered heteroaryl groups containing three heteroatoms include, but are not limited to, triazolyl, oxadiazolyl (e.g., 1,2,4-oxadiazolyl) and thiadiazolyl. Exemplary 5-membered heteroaryl groups containing four heteroatoms include, but are not limited to, tetrazolyl. Exemplary 6-membered heteroaryl groups containing one heteroatom include, but are not limited to, pyridinyl. Exemplary 6-membered heteroaryl groups containing two heteroatoms include, but are not limited to, pyridazinyl, pyrimidinyl and pyrazinyl. Exemplary 6-membered heteroaryl groups containing three or four heteroatoms include, but are not limited to, triazinyl and tetrazinyl, respectively. Exemplary 7-membered heteroaryl groups containing one heteroatom include, but are not limited to, azacycloheptatrienyl, oxacycloheptatrienyl and thiacycloheptatrienyl. Exemplary 5,6-bicyclic heteroaryl groups include, but are not limited to, indolyl, isoindolyl, indazolyl, benzotriazolyl, benzothienyl, isobenzothienyl, benzofuranyl, benzisofuranyl, benzimidazolyl, benzoxazolyl, benzisoxazolyl, benzoxadiazolyl, benzothiazolyl, benzisothiazolyl, benzothiadiazolyl, indazinyl, and purinyl. Exemplary 6,6-bicyclic heteroaryl groups include, but are not limited to, naphthyridinyl, pteridinyl, quinolyl, isoquinolyl, cinnolinyl, quinoxalinyl, phthalazinyl, and quinazolinyl.

[0100] The term "isomer" refers to compounds having the same molecular formula, including enantiomers, diastereomers, stereoisomers, etc. Stereoisomers are particularly preferred in the present invention, and the term "stereoisomer" refers to isomers that differ only in the spatial arrangement of atoms.

[0101] In some cases, the compounds of the present invention can form salts, which are also within the scope of the present invention. The term "salt(s)" refers to acidic and / or basic salts formed with inorganic and / or organic acids and bases. Pharmaceutically acceptable salts are particularly preferred in the present invention.

[0102] The term "pharmaceutically acceptable salt" means those carboxylates, amino acid addition salts of the compounds of the invention which are suitable for use in contact with patient tissues within the scope of sound medical judgment, do not produce undue toxicity, irritation, allergic response, etc., are commensurate with a reasonable benefit / risk ratio, and are effective for their intended use, including (where possible) zwitterionic forms of the compounds of the invention.

[0103] Pharmaceutically acceptable base addition salts are formed with metals or amines, such as alkali metals and alkaline earth metal hydroxides or organic amines. Examples of metals used as cations are sodium, potassium, magnesium, calcium, etc. Examples of suitable amines are N, N'-dibenzylethylenediamine, chloroprocaine, choline, diethanolamine, ethylenediamine, N-methylglucamine and procaine.

[0104] Base addition salts of acidic compounds may be prepared by contacting the free acid form with a sufficient amount of the desired base to form the salt in a conventional manner. The free acid may be regenerated by contacting the salt form with an acid and isolating the free acid in a conventional manner. The free acid forms differ from their respective salt forms somewhat in certain physical properties, such as solubility in polar solvents, but for purposes of the present invention, the salts are equivalent to their respective free acids.

[0105] Salts can be sulfates, pyrosulfates, bisulfates, sulfites, bisulfites, nitrates, phosphates, monohydrogenphosphates, dihydrogenphosphates, metaphosphates, pyrophosphates, chlorides, bromides, iodides prepared from inorganic acids, such as hydrochloric acid, nitric acid, sulfuric acid, hydrobromic acid, hydroiodic acid, phosphoric acid, etc. Representative salts include hydrobromide, hydrochloride, sulfate, bisulfate, nitrate, acetate, oxalate, valerate, oleate, palmitate, stearate, laurate, borate, benzoate, lactate, phosphate, toluenesulfonate, citrate, maleate, fumarate, succinate, tartrate, naphthoate, methanesulfonate, glucoheptonate, lactobionate, laurylsulfonate and isethionate, etc. Salts can also be prepared from organic acids, such as aliphatic mono- and dicarboxylic acids, phenyl-substituted alkanoic acids, hydroxyalkanoic acids, alkanedioic acids, aromatic acids, aliphatic and aromatic sulfonic acids, etc. Representative salts include acetate, propionate, octanoate, isobutyrate, oxalate, malonate, succinate, suberate, sebacate, fumarate, maleate, mandelate, benzoate, chlorobenzoate, methylbenzoate, dinitrobenzoate, naphthoate, benzenesulfonate, toluenesulfonate, phenylacetate, citrate, lactate, maleate, tartrate, methanesulfonate, etc. Pharmaceutically acceptable salts may include cations based on alkali and alkaline earth metals, such as sodium, lithium, potassium, calcium, magnesium, etc., as well as non-toxic ammonium, quaternary ammonium, and amine cations, including but not limited to ammonium, tetramethylammonium, tetraethylammonium, methylamine, dimethylamine, trimethylamine, triethylamine, ethylamine, etc. Also contemplated are salts of amino acids, such as argininate, gluconate, galacturonate, and the like (see, e.g., Berge SM et al., "Pharmaceutical Salts," J. Pharm. Sci., 1977; 66: 1-19, incorporated herein by reference).

[0106] The term "acceptable carrier" refers to a suitable carrier that can be used to utilize the existing material for the purposes of the present invention without excessive toxicity, irritation, allergic response, etc., equivalent to a reasonable benefit / risk ratio.

[0347] Example 19 - Transfection Efficiency

[0348] Various cationic lipid compounds and luciferase mRNA nanoparticles were encapsulated according to Example 18, and the fluorescence intensity or total photon number of luciferase mRNAs encapsulated by different LNPs were tested.

[0349] Experimental animals: SPF-grade BALB / c mice that passed the quarantine, female, 7-9 weeks old, purchased from Beijing Weitonglihua Experimental Animal Technology Co., Ltd., were randomly divided into groups according to their weight, with 3 mice in each group. All animals were adaptively fed for more than 7 days before the experiment, and had free access to food and water during the experiment. The light was alternating between light and dark for 12 / 12h, the indoor temperature was 20-26℃, and the humidity was 40-70%.

[0350] Experimental method: Female BALB / c mice were given different LNP-encapsulated luciferase mRNA by intramuscular injection (tibialis anterior muscle of the right leg of mice); bioluminescence detection was performed using a small animal in vivo imaging system (brand: Bruker, model: XTREME) at 3, 6, 24, 48, and 72 hours after administration. The specific steps are as follows: Substrate preparation: Take an appropriate amount of substrate Luciferin (brand: Promega) and add physiological saline to prepare a 10 mg / mL solution, which is kept away from light for standby use. The test substance dose is 5 μg / mouse, 50 μL / leg. After the mice are given the substrate, they move freely for 5-10 minutes, and then they are placed in an anesthesia box and anesthetized with 2.5% isoflurane. The anesthetized mice are placed in the machine, the bioluminescence parameters are set and photos are taken. After collecting the image, the upper and lower limits of the image are adjusted according to the fluorescence intensity of different groups, and data collection (such as fluorescence intensity, average photon number and total photon number) and data processing are performed on the parts where the fluorescence is concentrated. Statistical analysis: The results of in vivo imaging are expressed as the mean of the fluorescence intensity or total photon number of different animals in the same test group, so as to determine the fluorescence intensity or total photon number of luciferase mRNA encapsulated by different LNPs. The data of luciferase expression induced by ionizable lipid nanoparticle preparations in mice 6 hours after injection are shown in Tables 2-1 and 2-2.

[0351] The fluorescence intensity and total photon count reflect the transfection efficiency of LNP. The higher the value, the higher the efficiency of LNP to deliver the encapsulated substance into the cells.

The negative control group was given 100 μL PBS buffer.

[0365] 14 days after the second immunization, spleens of 4 mice were taken to isolate spleen lymphocytes, and T lymphocytes secreting IFN-γ and IL-4 in mice were detected by ELISPOT method. The specific operation of ELISPOT test was carried out according to the instructions of Mouse IL-4precoated ELISPOT kit (Da Kewei, 221 0402) and Mouse IFN-γprecoated ELISPOT kit (Da Kewei, 2210005).

[0366] 14 days and 35 days after the first immunization (i.e. 14 days after the second immunization), blood was collected from the orbits of the remaining 6 mice (to avoid hemolysis), and serum was collected for detection of BA.2.75.2 type Spike (S) protein-specific IgG antibodies by indirect ELISA. The specific operation of indirect ELISA for detecting BA.2.75.2 type S protein-specific IgG antibody titer is as follows:

[0367] Coating antigen: BA.2.75.2S protein (ACROBiosystems, Cat#SPN-C522r) was diluted to 2 ng / uL, 100 uL / well in coating buffer, and coated overnight at 4°C.

[0368] Wash the plate three times with 1× PBST, 5 min each time.

[0369] Blocked with 1% BSA blocking solution, 200uL / well, and allowed to stand at 37°C for 1 h.

[0370] Wash the plate three times with 1× PBST, 5 min each time.

[0371] The serum was diluted with dilution buffer, added at 100 ul / well, incubated at 37°C for 1 h, and a negative serum control and a blank control well without serum were set.

[0372] Wash the plate three times with 1× PBST, 5 min each time.

[0373] Anti-IgG secondary antibody was diluted 1:000, 100 ul / added, and incubated at 37°C for 1 h.

[0374] Wash the plate three times with 1× PBST, 5 min each time.

[0375] Add freshly configured TMB substrate chromogenic solution, 100 ul / well, and incubate for appropriate time at 37 °C.

[0376] Add stop solution 2mol / L sulfuric acid, 50ul / well.

[0377] The absorbance at OD450nm was measured by an enzyme-labeled instrument.

[0378] The results of IgG antibody titer are as follows Figure 2 , 3 As shown in the figure, the three mRNA vaccine preparations can induce humoral immune responses in BALB / c mice and produce S protein-specific IgG antibodies. At low and high doses, the antibody IgG level produced by L2 was basically the same as that of L1, regardless of the first or second immunization. The IgG antibody titer reached more than 4Log10 after the first immunization and 6Log10~7Log10 after the second immunization. The antibody IgG level produced by L3 was lower, but the IgG antibody titer reached more than 6Log10 after the second immunization. The ELISPOT results are shown in the figure. Figure 4As shown in the figure, after vaccination with low and high doses of vaccine preparations, the T lymphocytes secreting INF-γ in the L1, L2, and L3 groups increased significantly, and the frequency of T lymphocytes secreting INF-γ in the L3 group was the highest, followed by the L2 group, and the worst was the L1 group; after vaccination with low and high doses of vaccine preparations, the T lymphocytes secreting IL-4 in the L1, L2, and L3 groups increased. At low doses, the frequency of T lymphocytes secreting IL-4 in the L3 group was the highest, followed by the L2 group, and the worst was the L1 group; at high doses, there was no significant difference between the groups. In summary, compared with L1, L2 and L3, L3 can induce the strongest cellular immune response. The humoral immune response induced by L2 is equivalent to that of L1, but the cellular immune response is stronger.
