Description

Technical Field

[0001] The present invention relates to the technical field of pet health care, and in particular to a polysaccharide composition and application thereof for targeted inhibition of intestinal odor-causing bacteria in dogs and cats, promotion of short-chain fatty acid production, improvement of intestinal health and reduction of fecal odor. Background Art

[0002] The intestinal health and fecal odor problems of dogs and cats have long been important topics in pet care. Intestinal health directly affects the digestive function, immunity and overall health of pets. The problem of fecal odor not only affects the quality of life of pets, but also causes trouble to the living environment of pet owners. The intestines of dogs and cats contain a rich microbiome, including bacteria, fungi and other microorganisms, which play an important role in the digestion and metabolism process. Intestinal microorganisms not only help digest undigested food, but also participate in the production of various metabolites. The production of fecal odor in dogs and cats is mainly closely related to the metabolic activities of their intestinal microbiota, especially the decomposition of food residues, proteins and other organic matter by bacteria in the intestines. Especially for cats, as obligate carnivores, a high-protein diet is necessary for their health and nutritional needs, but a high-protein diet will produce undigested residues in the intestines. The proteins in these residues (especially sulfur-containing amino acids, cysteine, methionine, tryptophan, etc.) are easily metabolized by odor-causing bacteria in the intestines, producing ammonia (NH3), hydrogen sulfide (H2S), indoles, volatile fatty acids, olefin compounds and other volatile organic compounds (VOCs), which are the main sources of fecal odor. Similarly, a high-protein diet for dogs will also produce similar metabolites, especially in certain dog breeds, which show a stronger fecal odor problem.

[0003] Intestinal odor-causing bacteria refer to bacteria that produce compounds with a foul odor through metabolic activities. The metabolic pathways of odor-causing bacteria usually involve the decomposition of proteins, amino acids and sulfur-containing compounds to produce volatile organic compounds (VOCs), such as ammonia, hydrogen sulfide, methyl mercaptan, indole and volatile fatty acids. These compounds have a strong pungent odor. Common odor-causing bacteria are mainly distributed in several major bacterial phyla, including Desulfobacterota, Bacteroidetes, Firmicutes, Proteobacteria and Clostridia. Desulfovibrio in the Desulfobacterota is a typical odor-causing bacteria that produces H2S with a rotten egg smell through sulfate reduction. Bacteroides in the Bacteroidetes phylum, etc., produce indoles and sulfides by decomposing proteins and amino acids, with a distinct fecal and corrupt odor. Fusobacterium and Clostridium generate sulfide and amine compounds such as cadaverine and putrescine by decomposing proteins and amino acids, which increase the rancid smell of feces. Proteus and Morganella in the Proteobacteria generate a large amount of ammonia by decomposing urea, which has a pungent ammonia smell, which is particularly prominent in urinary tract and intestinal odor. Clostridia, as an important part of the Firmicutes, contains a large number of anaerobic odor-causing bacteria, which can generate odor substances such as sulfide, amines and indole through various metabolic pathways, aggravating intestinal odor.

[0004] At present, many existing patents on the market have proposed to regulate intestinal flora through probiotics, prebiotics (such as inulin, oligofructose, etc.), plant extracts, etc., so as to improve the intestinal health of animals. For example: Patent CN107156840B proposes to regulate intestinal health and improve defecation function through kudzu vine polysaccharides. The polysaccharide composition proposed in this patent has a certain prebiotic effect, but it does not specifically solve the problem of fecal odor, nor does it focus on the inhibition of specific intestinal harmful bacteria. Patent CN104013650B regulates the balance of intestinal microecology through Bacillus licheniformis and a traditional Chinese medicine polysaccharide composition, thereby improving the intestinal health of pets. Patent CN202311210890.4 discloses a pet food containing postbiotics, which regulates intestinal flora through probiotics and their metabolites, and reduces the generation of odor components such as ammonia from the root. Patent CN202410931260.4 proposes a deodorization scheme using a composite bacterial agent of Bacillus subtilis to improve the odor of animal excrement by inhibiting the production of ammonia and hydrogen sulfide. Patent CN201710491827.0 discloses a composite solid probiotic agent containing Phellinus igniarius, which uses multi-stage fermentation technology to enhance the activity of probiotics, and can more effectively decompose complex organic matter in the culture medium and produce polysaccharides, thereby helping to reduce the odor of livestock feces. In addition, patent CN202311399881.4 decomposes high-protein food residues through the encapsulation technology of slow-release enzyme preparations and probiotics, thereby reducing the intestinal burden of pets and reducing odor to a certain extent. Another patent, CN202410904598.0, proposes to use high-calcium vegetable extract to improve the intestinal microecological environment of pets, and reduce the odor of feces by reducing the production of substances such as indole and skatole.

[0005] However, there are currently no relevant studies and programs that focus on inhibiting intestinal odor-causing bacteria to reduce stool odor. Currently, most of these programs focus on the overall regulation of intestinal health or the addition of general probiotics, and fail to achieve targeted inhibition of specific odor-causing bacteria to effectively solve the stool odor problem caused by protein residue metabolism from the root. Some deodorization solutions are suitable for animal environments rather than pets, or fail to take into account the dual improvement of intestinal health and stool odor problems.

[0006] Fucoidan is mainly derived from seaweed plants. Its molecular structure contains sulfate groups, which give it strong antioxidant, anti-inflammatory and immunomodulatory functions. Studies have shown that fucoidan can improve metabolic diseases by regulating the abundance of intestinal flora and its metabolites and improving the intestinal microenvironment. Aloe polysaccharide is a type of high-molecular polysaccharide extracted from aloe plants, which has strong moisturizing, antioxidant, antibacterial and anti-inflammatory properties. Its structure contains rich polysaccharide chains, which can enhance the body's immunity. Aloe polysaccharide can improve intestinal health by regulating intestinal flora, promoting the growth of beneficial bacteria and inhibiting the reproduction of harmful bacteria. Yeast glucan is a polysaccharide extracted from yeast cell walls with good immunomodulatory functions. The β-1,3-glycosidic bond in its molecular structure gives it unique biological activity and significant immune activation properties. It can stimulate the activity of macrophages and lymphocytes and enhance the body's immune defense. Yeast glucan can support the growth of specific beneficial bacteria in the intestines and produce short-chain fatty acids during its fermentation process. These short-chain fatty acids have a positive effect on regulating the acid-base balance of the intestines and reducing the proliferation of harmful bacteria. Studies have also shown that yeast glucan can increase the diversity of intestinal flora, thereby improving the stability and anti-interference ability of the intestines.

[0007] Based on the effects of brown algae polysaccharides, aloe polysaccharides and yeast glucan in regulating intestinal flora and strengthening intestinal immunity, the present invention innovatively utilizes the combination of these three polysaccharides to achieve a more precise targeted regulation of odor-causing bacteria and the production of short-chain fatty acids. Summary of the invention

[0008] In view of the limitations of the prior art, the purpose of the present invention is to provide a polysaccharide composition for targeted inhibition of intestinal odor-causing bacteria in dogs and cats and promotion of the production of short-chain fatty acids, aiming to target the reproduction of various odor-causing bacteria such as Desulfovibrio in the intestines of dogs and cats, reduce the generation of odor molecules from the source, and significantly promote the production of short-chain fatty acids, thereby effectively improving the intestinal health of dogs and cats and reducing stool odor.

[0009] To achieve the above object, the present invention adopts the following technical solution:

[0010] In a first aspect of the present invention, there is provided a polysaccharide composition for targeted inhibition of intestinal odor-causing bacteria in dogs and cats and promotion of short-chain fatty acid production, which is composed of fucoidan, aloe polysaccharide and yeast glucan; the weight proportions of the three raw materials of fucoidan, aloe polysaccharide and yeast glucan in the polysaccharide composition are as follows:

[0011] 35-50 parts of brown algae polysaccharide,

[0012] Aloe polysaccharide 5-15 parts,

[0013] Yeast glucan 35-60 parts.

[0014] Preferably, the polysaccharide composition is composed of the following raw materials in parts by weight:

[0015] 40 parts of brown algae polysaccharide, 10 parts of aloe polysaccharide, and 50 parts of yeast beta-glucan.

[0016] The second aspect of the present invention provides the use of the above-mentioned polysaccharide composition in the preparation of pet feed or pet health products.

[0017] The third aspect of the present invention provides the use of the above-mentioned polysaccharide composition in the preparation of pet feed and pet health products for reducing fecal odor of dogs and cats.

[0018] A fourth aspect of the present invention provides the use of the polysaccharide composition described above in the preparation of pet feed and pet health products for promoting the production of short-chain fatty acids.

[0019] A fifth aspect of the present invention provides the use of the polysaccharide composition described above in the preparation of pet feed and pet health products for improving the intestinal health function of dogs and cats.

[0020] The composition of the present invention achieves its function through the following mechanism of action by using a specific ratio of brown algae polysaccharide, aloe polysaccharide and yeast glucan:

[0021] 1. Targeted inhibition of intestinal odor-causing bacteria: The polysaccharide composition significantly inhibits the proliferation of core odor-causing bacteria in the intestine (such as Fusobacterium, Desulfovibrio, Bacteroides, etc.) through synergistic effects. These bacteria are prone to metabolize protein and sulfur-containing amino acids under high-protein diet conditions, producing volatile organic compounds with foul odors such as sulfides and amines. By inhibiting the reproduction of odor-causing bacteria, the composition effectively reduces the generation of odor components.

[0022] 2. Promote the proliferation and metabolism of beneficial bacteria: The composition of the present invention can significantly promote the proliferation of acid-producing bacteria (such as Megamonas, Megasphaera and Bifidobacterium). These beneficial bacteria can metabolize and produce short-chain fatty acids (SCFAs) such as acetic acid, propionic acid and butyric acid. SCFAs play a key role in maintaining intestinal acid-base balance, inhibiting the growth of harmful bacteria, lowering intestinal pH and enhancing intestinal barrier function, which helps to optimize overall intestinal health.

[0023] 3. Competitive inhibition of metabolic substrate utilization: The polysaccharide composition significantly reduces the availability of metabolic substrates such as proteins and amino acids by preferentially supplying energy to acid-producing bacteria. This competitive inhibition mechanism directly limits the ability of odor-causing bacteria to utilize protein decomposition products, thereby indirectly reducing the generation of odor metabolites.

[0024] Compared with the prior art, the polysaccharide composition of the present invention has the following significant advantages:

[0025] 1. Comprehensive intestinal regulation mechanism: The present invention synergistically regulates intestinal flora through multiple pathways of action, including targeted inhibition of specific odor-causing bacteria, optimization of functional metabolism, and competition for metabolic substrates, thereby forming a more systematic intestinal regulation effect and adapting to the complexity of intestinal ecology.

[0026] 2. Targeting the unique needs of high-protein diet: The protein residues and metabolites brought by high-protein diets of dogs and cats are prone to cause fecal odor and flora disturbance. The composition of the present invention improves the intestinal environment problems and fecal odor problems caused by high-protein diets by effectively regulating intestinal metabolic activities.

[0027] 3. Comprehensive optimization of intestinal microecology: The present invention optimizes the flora structure and reduces the generation of odor components by inhibiting the growth of odor-causing bacteria and improving the metabolic activity of acid-producing bacteria, while promoting the healthy development of intestinal function. [0043] Example 4 Polysaccharide composition promotes lactic acid bacteria fermentation and acid production

[0044] 1. Test methods

[0045] In order to evaluate the growth-promoting effect of the polysaccharide composition on lactic acid bacteria, 7 strains of lactic acid bacteria (including Bifidobacterium, Lactobacillus and Pediococcus) were selected for mixed fermentation experiments to simulate the intestinal microecological environment and reflect the comprehensive effect of the polysaccharide composition on multiple lactic acid bacteria species.

[0046] (1) Preparation of bacterial solution: Bifidobacterium adolescentis, Bifidobacterium animalis, Bifidobacterium longum, Lactobacillus acidophilus, Lactobacillus plantarum, Pediococcus acidilactici N-FE-001 and Lactobacillus reuteri N-FE-002 were provided by Weishi Pet Nutrition Science Research Institute (Jiangsu) Co., Ltd. The above lactic acid bacteria were inoculated into BHI broth respectively and cultured in an anaerobic incubator at 38°C for 24 hours for recovery. After the culture was completed, the bacterial solution was centrifuged to remove the precipitate, and the bacterial solution was resuspended in sugar-free MRS broth, and the concentration of the bacterial solution was adjusted to 1.5 McFarland standard. The lactic acid bacteria were mixed in a ratio of 1:1 (v / v) to prepare a mixed lactic acid bacteria solution.

[0047] (2) Grouping: The experiment was divided into the following groups. Sugar-free MRS broth was used as the basic culture medium. Different polysaccharide components were added to each group according to the following proportions:

[0048] Negative control group (NC): sugar-free MRS broth without added polysaccharides.

[0049] Example 1 Polysaccharide composition group (FAY1): a polysaccharide composition prepared with 35 parts of brown algae polysaccharide, 5 parts of aloe polysaccharide, and 35 parts of yeast glucan, added to sugar-free MRS broth at 2% (m / v), with a final concentration of 0.93% brown algae polysaccharide, 0.13% aloe polysaccharide, and 0.93% yeast glucan (m / v).

[0050] Example 2 Polysaccharide composition group (FAY2): a polysaccharide composition prepared with 50 parts of brown algae polysaccharide, 15 parts of aloe polysaccharide, and 60 parts of yeast glucan was added to sugar-free MRS broth at 2% (m / v), with a final concentration of 0.8% brown algae polysaccharide, 0.24% aloe polysaccharide, and 0.96% yeast glucan (m / v).

[0051] Example 3 Polysaccharide composition group (FAY3): a polysaccharide composition prepared with 40 parts of brown algae polysaccharide, 10 parts of aloe polysaccharide, and 50 parts of yeast glucan was added to sugar-free MRS broth at 2% (m / v), with a final concentration of 0.8% brown algae polysaccharide, 0.2% aloe polysaccharide, and 1% yeast glucan (m / v).

[0052] The comparison group of Example 3 was set up as follows:

[0053] Fucoidan + aloe polysaccharide group (FA): Only fucoidan and aloe polysaccharide were added to sugar-free MRS broth, with the final concentrations of 0.8% fucoidan and 0.2% aloe polysaccharide (m / v).

[0054] Yeast glucan group (YG): Only yeast glucan was added to sugar-free MRS broth, with a final concentration of 1% yeast glucan (m / v).

[0055] (3) Inoculation and culture: 2% of the mixed bacterial solution of lactic acid bacteria (volume ratio) was added to each group and cultured anaerobically at 38°C for 24 hours. The OD value and pH value of the fermentation broth of each group were measured at the beginning of the culture (0 hours) and after the end of the culture (24 hours) to determine the promoting effect of the polysaccharide composition on the fermentation growth and acid production of lactic acid bacteria. The change of OD value reflects the growth of lactic acid bacteria, while the change of pH value indicates the acid production capacity.

[0056] 2. Test results

[0057] Experimental results ( Figure 1) showed that in the control group (NC) without polysaccharide addition, the OD value of lactic acid bacteria was 0.412 and the pH value was 5.97, indicating that the growth and acid production ability were weak. The OD value of yeast glucan (YG group) added alone was 0.433 and the pH value was 5.94, showing that its promoting effect on the growth and acid production of lactic acid bacteria was limited. Adding a combination of brown algae polysaccharide and aloe polysaccharide (FA group) significantly increased the OD value to 1.341, and the pH value decreased to 4.68, indicating that brown algae polysaccharide and aloe polysaccharide are the core components that promote the growth and acid production of lactic acid bacteria. The FAY3 polysaccharide combination (brown algae polysaccharide, aloe polysaccharide and yeast glucan) of Example 3 further enhanced the proliferation and acid production of lactic acid bacteria, with an OD value of 1.394 and a pH value of 4.64, showing the best effect. In addition, the polysaccharide compositions of Example 1 (FAY1) and Example 2 (FAY2) also showed significant proliferation and acid production promoting effects. In summary, the polysaccharide composition of the present invention exhibits a synergistic effect, which is superior to a single component and a two-component combination.

[0058] Example 5 Polysaccharide composition promotes intestinal flora to produce short-chain fatty acids and lowers intestinal pH

[0059] 1. Test methods

[0060] (1) Sample preparation: In order to verify the effect of the polysaccharide composition of the present invention on promoting the production of short-chain fatty acids by the colon flora of dogs and cats, a dog and cat fecal fermentation test was conducted. Four fresh dog and cat fecal samples were collected respectively, and the feces were immediately transferred to anaerobic bags after collection, and mixed at a 1:1 (mass ratio) to obtain a mixed sample of dog and cat feces. The four dogs and four cats involved in the fecal collection were all in good health. They had not consumed probiotics, taken any medications, had no history of vaccination, no history of antibiotic use, no history of drug treatment, and did not suffer from any digestive tract-related diseases within one month before the experiment.

[0061] (2) Preparation of fecal slurry: Weigh 20 g of mixed fecal sample and add 80 ml of GAM medium to prepare 20% (m / v) fecal slurry. Mix the mixed fecal slurry evenly in an oscillator and then filter it with a 150-mesh gauze to obtain 20% fecal slurry for later use.

[0062] (3) Grouping and medium preparation: The polysaccharide combination of Example 3 was selected for testing. The fermentation GAM medium was prepared according to the experimental grouping. The prepared medium was placed in an anaerobic environment and deoxygenated for 24 hours for use. The experimental groups included:

[0063] Negative control group (NC): modified GAM medium without added polysaccharide.

[0064] Example 3 Polysaccharide composition group (FAY): a polysaccharide composition prepared with 40 parts of brown algae polysaccharide, 10 parts of aloe polysaccharide, and 50 parts of yeast glucan was added to the modified GAM medium at 2% (m / v), with a final concentration of 0.8% brown algae polysaccharide, 0.2% aloe polysaccharide, and 1% yeast glucan (m / v).

[0065] The control group of Example 3 was set up as follows:

[0066] Fucoidan + aloe polysaccharide group (FA): Only fucoidan and aloe polysaccharide were added to the modified GAM medium, with the final concentrations of 0.8% fucoidan and 0.2% aloe polysaccharide (m / v).

[0067] Fructo-oligosaccharide group (FOS): 1% Fructo-oligosaccharide was added to the modified GAM medium.

[0068] (4) Fermentation culture and sample collection: Fecal slurry was added to each culture medium at a 10% inoculum rate and fermented in an anaerobic fermentation incubator at 38°C for 24 hours. After fermentation, the fermentation broth of each group was centrifuged and the supernatant was collected for pH determination, short-chain fatty acid determination, etc. The intestinal flora precipitate was collected for subsequent intestinal flora sequencing analysis.

[0069] (5) Determination of short-chain fatty acid content: The short-chain fatty acid content of the supernatant after fermentation of intestinal flora in each group was determined. Liquid chromatography-tandem mass spectrometry (LC-MS / MS) multiple reaction monitoring mode (MRM) was used to analyze short-chain fatty acids, and the specific content of 11 short-chain fatty acids was detected. 11 short-chain fatty acid standards were purchased from Shanghai Zhenzhun Biotechnology Co., Ltd. The analytical instruments used were Thermo Scientific TSQ Altis mass spectrometer and Thermo Scientific VanquishFlex liquid chromatograph. 11 short-chain fatty acid standards were accurately weighed to prepare mixed standard linear mother solutions, and the linear mother solutions were diluted with methanol to obtain a series of working solutions. A certain concentration of isotope internal standard solution was prepared and mixed to obtain an internal standard solution (IS). The mother solutions and working solutions of linearity, internal standard and quality control were all frozen and stored. The concentration series of the standard solution were detected by LC-MS, with the ratio of the standard to the internal standard concentration as the horizontal axis and the ratio of the standard to the internal standard peak area as the vertical axis to examine the linearity of the standard solution. The linear regression equations of each short-chain fatty acid were obtained. Subsequently, all samples were quantitatively analyzed to ensure high sensitivity and accuracy of the detection results.

[0070] (6) Determination of lactic acid content: The lactic acid content in the fermentation supernatant was determined using a lactic acid content kit (Jianglai Biotechnology) in strict accordance with the instructions of the kit.

[0071] (7) pH determination: The pH value of the fermentation supernatant of each group was measured using a pH meter.

[0072] 2. Test results

[0073] The FAY polysaccharide composition of the present invention significantly increases the content of short-chain fatty acids and lactic acid in the fermentation liquid of dog and cat feces, and significantly reduces the pH value of the fermentation liquid (Table 1-2).

[0074] As shown in Table 1, in the intestinal flora fermentation broth of cats, the total acid content of the FAY composition group reached 21488.69 μg / mL, an increase of 19.16% compared with the negative control group (FE_NC). In contrast, the positive control group (FOS) was 19664.94 μg / mL, only 9.05% more than the negative control group, showing that the FAY composition has a more significant promoting effect on total acid production. In terms of core acid (lactic acid, acetic acid, propionic acid and butyric acid) content, the FAY composition group increased by 16.58% compared with the negative control group and 15.84% higher than the FA group. This shows that yeast glucan has a significant synergistic effect in combination with fucoidan and aloe polysaccharide, significantly improving acid production activity. In addition, the pH value of the FAY composition group dropped to 6.26, compared with 6.60 in the FOS group and 6.74 in the FA group, further showing its unique advantages in regulating the acidic environment. As a widely used prebiotic, FOS has a certain promoting effect on the acidic environment, but the FAY composition of the present invention is more significant in increasing acidity and core acid production.

[0075] As shown in Table 2, the experimental results of dogs also showed the advantages of the FAY composition. The total acid content of the FAY composition group was 17376.7 μg / mL, an increase of 4.45% compared with the negative control group (CA_NC), and the core acid content reached 16947.3 μg / mL, which was much higher than the FA group and the FOS group. Compared with the FA and FOS groups, the advantages of the FAY composition in core acid production are more prominent, indicating that the FAY composition also exhibits a synergistic effect in the intestines of dogs. At the same time, the pH value of the FAY composition group dropped to 5.78, which was significantly lower than 6.10 of the FOS group and 6.32 of the FA group, which shows that the FAY composition can effectively reduce the pH value of the intestine and is conducive to the proliferation of probiotics.

[0076] The data in Tables 1 and 2 show that the FAY composition of the present invention has significant advantages in promoting the production of short-chain fatty acids and regulating the acidic environment of the intestinal flora of cats and dogs, especially in promoting the production of lactic acid, propionic acid and butyric acid. Compared with the single fucoidan + aloe polysaccharide group (FA) and the widely used prebiotic oligofructose group (FOS), the FAY composition exhibits a stronger synergistic effect.

[0081] Example 6 Polysaccharide composition regulates the balance of intestinal flora and inhibits the growth of intestinal odor-causing bacteria

[0082] 1. Test methods

[0083] 16S rDNA amplicon sequencing: In order to verify the effect of the polysaccharide composition of the present invention on the intestinal microbial community of dogs and cats, 16S rDNA amplicon sequencing analysis was performed using fecal fermentation precipitate samples. First, total DNA was extracted from fecal fermentation precipitates of dogs and cats according to the QIAamp DNA Stool MiniKit kit. The extracted DNA was used for PCR amplification, and primers 515F and 806R for the bacterial 16S rDNA V4 region were used for amplification to ensure that the bacterial diversity in the intestine can be fully covered and identified. The PCR reaction system included 15 μL of high-fidelity PCR master mix (Phusion High-Fidelity PCR Master Mix), 0.2 μM primers and 10 ng genomic DNA template. The reaction conditions were: pre-denaturation at 98°C for 1 min, followed by 30 cycles, each cycle including denaturation at 98°C (10 s), annealing at 50°C (30 s) and extension at 72°C (30 s), and finally extension at 72°C for 5 min to complete the amplification. After amplification, the PCR products were purified by magnetic beads and mixed in equal amounts according to the product concentration. The mixed PCR products were detected and the target bands were recovered. Finally, the sequencing library was constructed, and the library was sequenced on the machine after passing the Qubit fluorometer and qPCR quantitative detection. Subsequently, the raw data (RawData) obtained by sequencing was spliced and filtered to obtain valid data (Clean Data); then, based on the valid data, DADA2 was used for noise reduction to obtain characteristic amplicon sequences (Amplicon Sequence Variants, ASVs). The representative sequence of each ASV was annotated with species to obtain the corresponding species information and species-based abundance distribution, and principal coordinate analysis (PCoA) was performed based on the weighted UniFrac distance to analyze the diversity of the bacterial community and its abundance changes.

[0084] 2. Test results

[0085] (1) Polysaccharide composition significantly regulates the balance of intestinal flora

[0086] Principal coordinates analysis (PCoA) results ( Figure 2 ) showed that the FAY composition had a significant effect on regulating the intestinal flora structure of dogs and cats. The sample community structure of the FAY group was more aggregated and separated from the control group (NC group) and the positive control group (FOS group), indicating that the FAY composition had a significant effect on adjusting the flora structure.

[0087] The experimental results (Table 3 and Table 4) show that the FAY composition has significant advantages in regulating the balance of intestinal flora in dogs and cats. Compared with the control group (NC) and the positive control group (FOS), the FAY composition can effectively promote the proliferation of beneficial bacteria and significantly inhibit the reproduction of odor-causing bacteria, especially in the proliferation of Firmicutes and the inhibition of Fusobacteriota and Desulfobacterota.

[0088] Cat intestinal flora (Table 3): In the FAY group, the abundance of Firmicutes increased significantly to 57.55%, much higher than that of the NC group (28.92%). At the same time, the FAY composition significantly reduced the abundance of Fusobacteria, which was only 6.43%, while that of the NC group was 44.63% and that of the FOS group was 8.23%. In addition, the FAY group had a particularly prominent inhibitory effect on Desulfobacteria, with an abundance of only 0.19%, significantly lower than that of the NC group (2.53%) and the FOS group (0.60%). This shows that the FAY composition has a significant advantage in reducing intestinal odor components.

[0089] Canine intestinal flora (Table 4): In the FAY group, the abundance of Firmicutes increased to 63.27%, which was significantly higher than that of the NC group (23.98%) and the positive control group FOS (33.83%). At the same time, the FAY composition had a very obvious inhibitory effect on the Fusobacterium, with an abundance of only 0.84%, while the NC group was 47.86% and the FOS group was 4.90%. For the Desulfobacterium, the FAY composition also showed an excellent inhibitory effect, with an abundance of 0.05%, which was significantly lower than that of the NC group (0.59%) and the FOS group (0.10%).

[0090] The effect of the FAY composition was significantly better than that of the control group (NC) and the positive control group (FOS). In the NC group, the abundance of odor-causing bacteria (such as Fusobacteria and Desulfobacteria) was higher, while the abundance of beneficial bacteria (such as Firmicutes) was lower. The positive control group FOS improved the flora structure to a certain extent, but the effect was weaker than that of the FAY composition, which further proved the advantages of the FAY composition in promoting the proliferation of beneficial intestinal bacteria and inhibiting the reproduction of odor-causing bacteria.

[0098] (2) The polysaccharide composition significantly inhibits the growth of odor-causing bacteria and promotes the proliferation of acid-producing bacteria

[0099] In order to verify the inhibitory effect of the polysaccharide composition of the present invention on intestinal odor-causing bacteria, we conducted intestinal flora analysis at the genus level and clarified the regulatory effects of different treatment groups on odor-causing bacteria and acid-producing bacteria. The results showed that the FAY composition significantly inhibited the reproduction of odor-causing bacteria and significantly promoted the proliferation of acid-producing bacteria, showing excellent flora regulation ability.

[0100] As shown in Table 5, in the cat intestine, the FAY composition significantly reduced the overall abundance of odor-causing bacteria, among which the abundance of Fusobacterium decreased from 31.2872% in the NC group to 0.3655%, with an inhibition rate of 98.8%; the abundance of Desulfovibrio decreased from 2.4658% to 0.1763%, with an inhibition rate of 92.8%; the abundance of Bacteroides decreased from 3.7854% to 1.0795%, with an inhibition rate of 71.5%. The overall proportion of odor-causing bacteria decreased from 44.1265% to 4.5178%, with an inhibition rate of 89.8%. At the same time, the FAY composition significantly promoted the proliferation of acid-producing bacteria (producing beneficial ingredients such as short-chain fatty acids). The abundance of Megamonas, a typical short-chain fatty acid-producing bacteria, increased from 10.6024% to 41.7037%, with a promotion rate of 293.3%; the abundance of Megasphaera increased from 0.732% to 5.8629%, with a promotion rate of 701.2%. The proportion of acid-producing bacteria increased from 11.4437% to 47.9821%, with a promotion rate of 319.3%.

[0101] As shown in Table 6, in the canine intestine, the FAY composition also showed a significant inhibitory effect on odor-causing bacteria. The abundance of Fusobacterium decreased from 47.0997% in the NC group to 0.8437%, with an inhibition rate of 98.2%; Desulfovibrio decreased from 0.0027% to 0%, completely inhibited; the abundance of Bacteroides decreased from 7.1847% to 2.1181%, with an inhibition rate of 70.5%. The total proportion of odor-causing bacteria decreased from 71.2367% to 23.976%, with an inhibition rate of 66.3%. In addition, the FAY composition greatly increased the abundance of acid-producing bacteria, among which the abundance of Megamonas increased from 1.5999% to 51.8855%, with a promotion rate of 3144.3%; the abundance of Bifidobacterium increased from 0.0098% to 1.4684%, with a promotion rate of up to 14987.8%. The FAY composition also promoted the abundance of Lactobacillus, Pediococcus, Weissella (Bacillus), Lactococcus, and Leuconostoc. The total proportion of acid-producing bacteria increased from 1.6303% to 53.3915%, with a promotion rate of 3174.9%.

[0102] The above results show that the FAY composition of the present invention can significantly inhibit the reproduction of intestinal odor-causing bacteria, while significantly promoting the proliferation of acid-producing bacteria and improving the intestinal flora structure. The mechanism of action of the FAY composition may be achieved through the following pathways: on the one hand, by promoting the rapid proliferation of acid-producing bacteria, preferentially utilizing nutritional substrates such as proteins and amino acids, resulting in a significant reduction in the metabolic substrates on which odor-causing bacteria depend for survival, thereby achieving competitive inhibition of the growth of odor-causing bacteria; on the other hand, the short-chain fatty acids (such as acetic acid, propionic acid and butyric acid) generated during the metabolism of acid-producing bacteria reduce the pH value of the intestine, thereby further inhibiting the growth and metabolic activity of odor-causing bacteria, and ultimately effectively reducing the generation of intestinal odor substances.

[0111] Example 7 Polysaccharide composition inhibits fecal odor markers produced by intestinal odor-causing bacteria

[0112] 1. Test methods

[0113] To further verify the inhibitory effect of the polysaccharide composition on fecal odor metabolites produced by odor-causing bacteria, non-targeted metabolomics analysis technology was used to quantitatively analyze the metabolites in cat fecal fermentation broth samples. The samples of each experimental group were mixed in equal volumes for testing, and quality control samples (QC) were prepared at the same time. The samples of all experimental groups were mixed in equal amounts to ensure the stability of the analysis and the quality of the data; 53% methanol aqueous solution was used as a blank control (Blank) to eliminate background interference signals. The specific operation included: taking 300 μL of sample, adding an equal volume of pre-cooled 80% methanol aqueous solution for pretreatment, the sample was quickly frozen in liquid nitrogen and then melted on ice, and then ultrasonically treated for 6 minutes to fully extract metabolites. Centrifuge and take the supernatant. Sample analysis was separated by Vanquish UHPLC ultra-high performance liquid chromatography (Thermo Fisher), and combined with QExactive HF high-resolution mass spectrometer (Thermo Fisher) for qualitative and quantitative analysis of metabolites. Chromatographic separation was performed using a C18 reverse phase column, with a mobile phase of 0.1% formic acid in water and 0.1% formic acid in acetonitrile, a flow rate of 0.3 mL / min, and a column temperature of 40 °C. Mass spectrometry acquisition was performed in positive and negative ion mode (ESI+ / -), with a mass range of m / z 70–1050, a resolution of 120,000, and an acquisition mode of Full-MS / ddMS2. The raw data were filtered by retention time and mass-to-charge ratio, and the peak area was normalized and corrected using QC samples to ensure data accuracy. Compounds with a coefficient of variation (CV) of more than 30% in the QC samples were removed, and effective metabolites were identified after background ion signals were removed. The metabolites were qualitatively compared using the mzCloud, mzVault, and Masslist databases, and the identification was completed by combining molecular ion and fragment ion information. The screening criteria for differential metabolites were variable importance projection (VIP) score > 1, statistical significance (P value) < 0.05 (T-test method), and fold change (FC) ≥ 1.2 or ≤ 0.83. The screened differential metabolites were classified and observed by functional classification.

[0114] 2. Test results

[0115] (1) The polysaccharide composition significantly reduces the concentration of odor-causing metabolites and odor-causing precursors

[0116] The FAY composition of the present invention exhibits a significant effect in inhibiting intestinal odor-causing metabolites (as shown in Table 7). Specifically, the following major types of metabolites are reduced:

[0117] Direct markers: The FAY composition significantly reduced the abundance of multiple direct markers of fecal odor, including 2-oxindole (skatole, reduced by 84.1%), hydrocinnamic acid (reduced by 82.4%), 4-ethylphenol (reduced by 76.4%) and styrene (reduced by 75.8%), 5-hydroxyindole (skatole, reduced by 65.5%), 4-hydroxyindole (skatole, reduced by 62.7%), and other uramine and ethylene glycol sulfate compounds. These compounds are produced during intestinal fermentation and have a strong odor. They are the source of pet fecal odor. The significant inhibitory effect of the FAY composition helps reduce fecal odor.

[0118] Amino acids and their derivatives: As substrates and precursors of odor-causing metabolism, some amino acids were significantly reduced under the action of the FAY composition, including Asp-Phe, N-acetyl-L-phenylalanine, S-sulfo-L-cysteine, etc., which were reduced by more than 50%. These benzene rings or sulfur-containing amino acids are converted into odorous volatiles in the metabolism of odor-causing bacteria. The FAY composition effectively inhibits these odor-causing pathways by reducing their abundance.

[0119] Amines: The FAY composition also showed a strong inhibitory effect on amine odor-causing substances, including N-oleyl dopamine (reduced by 80.6%), stearyl ethanolamide (reduced by 68.6%) and other amine compounds. The reduction of these amines can further improve the fecal odor problem.

[0120] (2) The polysaccharide composition significantly increased the beneficial metabolites in the intestine

[0121] While suppressing odor-causing substances, the FAY composition also significantly increased the abundance of beneficial metabolites in the intestine:

[0122] Phospholipid metabolites: The FAY composition significantly promoted the production of phospholipid metabolites, such as LPG 15:0, LPG 16:1, and LPE O-18:2, with growth rates ranging from 810.5% to 2744.0%, respectively. These have potential anti-inflammatory and immunomodulatory effects.

[0123] Vitamins: The FAY composition promotes the synthesis of vitamins such as niacinamide (increase of 259.1%) and pantothenic acid (increase of 262.5%). Vitamins play an important role in supporting intestinal health and enhancing the immune system, which helps to further improve the intestinal environment.

[0124] Prebiotics: The FAY composition promotes the concentration of maltotetraose, α-cyclodextrin, and maltotriose in the intestinal fermentation broth, which are known prebiotic ingredients (e.g., maltotetraose and maltotriose are components of prebiotic oligosaccharides), and contribute to the reproduction of intestinal probiotics.

[0125] Beneficial amino acids and their derivatives: A variety of amino acid derivatives increased significantly under the action of the FAY composition, including histamine (increased by 2879.4%) and N-acetylhistamine (increased by 282.8%). These amino acid derivatives can regulate intestinal immunity, inhibit harmful bacteria, and help maintain a healthy balance of intestinal microecology.

[0126] The above data indicate that the FAY composition significantly inhibits the production of fecal odor markers and odor-causing precursor amino acids, while promoting the production of vitamins and the like.

[0130]

[0131] Note: Metabolite abundance data are expressed as the average of 4 repeated tests, and log2FC represents the logarithm of the abundance change fold between the FAY group and the NC group (log2 transformation). "Reduction / increase ratio" shows the percentage change of the FAY group compared with the NC group, with negative values indicating reduction and positive values indicating increase. P values were calculated by T-test, and the significance levels were as follows: P<0.05 indicated a significant difference, and P<0.01 indicated an extremely significant difference. Metabolite classification was based on chemical classification and biological function in the annotation database.

[0132] Example 8 Dog and Cat Testing

[0133] 1. Test methods

[0134] In order to verify the effect of the polysaccharide composition on the inhibition of fecal odor and regulation of intestinal flora in dogs and cats, we conducted a feeding test on healthy dogs and cats. 15 healthy adult dogs and cats were selected. The animals had not received antibiotics, probiotics or other drug interventions within 1 month before the start of feeding the polysaccharide composition, nor had they been vaccinated, and all were in a healthy state. The animals were randomly divided into three groups: a no-addition control group (NC group), a polysaccharide composition group of Example 2 (FAY2 group), and a polysaccharide composition group of Example 3 (FAY3 group). The experiment collected and analyzed samples at the following time points:

[0135] A. Baseline data collection (Day 0): Fecal samples were collected from all animals before the start of the experiment and recorded as baseline data;

[0136] B. Intervention for 7 days (7 days): The FAY2 and FAY3 groups were fed with a polysaccharide composition in the basal diet once a day, 2 g each time, for 7 days, and stool samples were collected. The NC group was fed only a basal diet without the addition of a polysaccharide composition for 7 days, and stool samples were collected. A handheld hydrogen sulfide and ammonia gas detector (Aeroqual S200-EHT) was used to detect the concentration of volatile H2S and NH3 in the stool samples. The gas probe of the detector was inserted into the gas inlet of the screw-capped bottle containing the stool sample, left to stand for 3 minutes, and the values were recorded.

[0137] 2. Test results

[0138] The experimental results (Table 8) show that the polysaccharide composition has a significant inhibitory effect on volatile hydrogen sulfide and ammonia in dog and cat feces. In the control group (NC group), the concentrations of hydrogen sulfide and ammonia did not change significantly between 0 and 7 days (P>0.05), indicating that the basic diet had no significant effect on the generation of fecal odor. However, after 7 days of intervention, the concentrations of hydrogen sulfide and ammonia in the Example 2 (FAY2) and Example 3 (FAY3) groups were significantly reduced.

[0139] In cat feces, the hydrogen sulfide concentration in the FAY3 group decreased from 2.26±0.77ppm to 1.12±0.29ppm, with an inhibition rate of 50.4%; the ammonia concentration decreased from 2.05±0.88ppm to 1.03±0.60ppm, with an inhibition rate of 49.8%. In dog feces, the hydrogen sulfide concentration in the FAY3 group decreased from 1.56±0.43ppm to 0.73±0.27ppm, with an inhibition rate of 53.2%; the ammonia concentration decreased from 1.95±0.83ppm to 0.33±0.16ppm, with an inhibition rate of up to 83.1%.

[0140] In summary, the polysaccharide composition of the present invention effectively inhibits the metabolic activity of odor-causing bacteria by regulating the balance of intestinal flora, and significantly reduces the generation of odor-causing components such as hydrogen sulfide and ammonia.
