[0001] SYNERGISTIC PRESERVATIVE BLEND AND COMPOSITIONS CONTAINING

[0002] SAME

[0003] BACKGROUND

[0004]

[0001] Microbial contamination of personal care products, home care products and other similar products is a matter of great importance to the industry and can become a major cause of product losses. Preservative agents and preservative compositions for protecting and preserving end-use formulations against microbial attack from bacteria or fungi are known in the art. These preservative agents and compositions have a wide variety of applications in fields such as personal care products, cosmetics, home care products, and health and hygiene products. Conventional preservative agents and compositions have used traditional active ingredients, such as benzisothiazolinone, methylisothiazolinone, and methylchloroisothiazolinone, which provide good bacterial and fungicidal properties against microbial attack. However, because of growing governmental scrutiny regarding the use of benzisothiazolinone, methylisothiazolinone, and methylchloroisothiazolinone, alternative preservative compositions which are similarly low cost and broad spectrum have been desired.

[0005]

[0002] One problem with antimicrobial agents, such as pyrithione, is that they often require high concentrations to be effective in preservative compositions. Because such high concentrations are known to be required, the use of pyrithione in personal care products and home care products can be cost prohibitive.

[0006]

[0003] Ideally, a desired preservative agent or composition has broad-spectrum activity against all types of microorganisms at various pH levels. The preservative agent or composition should also have high efficacy so that a minimum amount of the preservative can be used to save cost and to avoid or reduce any possible adverse effects caused by the preservative agent or preservative agents in the preservative composition. Also, it is desirable that the preservative agent or composition is stable to any changes in temperature encountered during manufacturing, packaging, and shipping as well as during storage of the end-use formulation in which the preservative agent or compositions is placed. Further, ideal preservative agents are physically and chemically compatible with ingredients present in various end-use formulations so that one preservative agent or preservative compositions can suitably be incorporated in various products.

[0007]

[0004] In recent developments in the preservative arts, there has been considerable effort given to find compounds that will synergistically interact with the preservative agent(s) in order to boost the efficacy of the preservative agent(s) and / or to effectively reduce the amount of preservative agent(s) needed to have a desired preservation effect. This is a result of continued regulatory pressures to reduce the amount of the preservative agent(s) in end-use formulations.

[0008]

[0005] Similarly, in the field of personal care formulations containing an active agent to treat specific conditions, such as dandruff, there is ongoing regulatory pressures to reduce the amount of the active ingredient in personal care formulations. One way to reduce the amount of the active ingredients is to increase the active ingredients efficacy in the personal care formulation, forming a synergistic mixture of an active ingredient with a potentiator, which increases the efficacy of the active ingredient.

[0009]

[0006] The present disclosure provides an answer to that need, by providing a preservation composition comprising a synergistic mixture of a pyrithione, an organic acid or salt thereof, and an aromatic alcohol which can be used in various compositions as a preservative, including personal or home care formulations.

[0027] DETAILED DESCRIPTION

[0028]

[0024] It is to be understood by one of ordinary skill in the art that the present discussion is a description of exemplary embodiments only and is not intended as limiting the broader aspects of the present disclosure.

[0029]

[0025] In general, the present disclosure is directed to a preservative composition. As used herein, the term “preservative” means a biocidal agent or biocidal composition which is intended to be blended into an end-use formulation, such as personal care formulations, cosmetics, home care formulations, and health and hygiene products, to prevent microorganisms from destroying the end-use formulation or making the end-use formulation unusable for the particular purpose the end-use formulation was developed.

[0030]

[0026] It has now been surprisingly found that the combination of a pyrithione, an organic acid or salt thereof, and an aromatic alcohol in a preservative composition produce synergistic effects and can be incorporated into end-use formulations at very low levels while providing excellent antimicrobial properties. As used herein, a “synergistic interaction” refers to the fact that the combination of pyrithione, organic acid or salt thereof, and aromatic alcohol has a total antimicrobial / antifungal effect that is greater than the antimicrobial / antifungal properties of the combination of only an organic acid or salt thereof and an aromatic alcohol. In other words, the pyrithione of the present disclosure operates synergistically with an organic acid or salt thereof and an aromatic alcohol so as to have greater antimicrobial / antifungal activity in the presence of each other against certain microorganisms than in comparison to the antimicrobial activity of the pyrithione alone or the antimicrobial activity of the combination of organic acid or salt thereof and aromatic alcohol at the same concentrations. Due to the synergistic effect, the amount of the pyrithione present in the preservative composition can be reduced while still producing the desired efficacy. This effect is also known as “potentiation” of the pyrithione in the preservative composition. This potentiation of the pyrithone is also referred to herein as a “synergistic effect” between the pyrithione and the combination of an organic acid or salt thereof and an aromatic alcohol for boosting the efficacy of the pyrithione.

[0031]

[0027] As used herein, the term “salts thereof” are intended to include cations of sodium, potassium, calcium, magnesium, ammonium and ethanolamines and other similar cations, anions of chloride, bromine, acetate, sulfates and the other similar anions.

[0032]

[0028] The preservative composition disclosed herein comprises (i) a pyrithione; (ii) an organic acid or salt thereof; and (iii) an aromatic alcohol. The weight ratio of the pyrithione to the organic acid or salt thereof and to the aromatic alcohol is from about 1 :1 :1 to about 1 : 15630: 12500, such as from about 1 :1 :1 to about 1 : 10000: 10000, such as from about 1 :1 :1 to about 1 :5000:5000.

[0033]

[0029] Pyrithione compounds are preservative agents known as iron chelators. Pyrithione is known by several names, including 2 mercaptopyridine-N- oxide; 2- pyridinethiol-1 -oxide (CAS Registry No. 1 121 -31 -9); 1 -hydroxypyridine-2- thione and 1 hydroxy-2(1 H)-pyridinethione (CAS Registry No. 1 121 -30-8); 2-pyridinol-1 - oxide (HPNO) and N-hydroxy-6-octyloxypyridine 2(1 H)-one and -hydroxy-6- octyloxypyridine 2(1 H)-one ethanolamine salt. Pyrithione salts are commercially available from Arxada AG such as Sodium OMADINE® or Zinc OMADINE®.

[0030] The pyrithione present as the preservative agent can be present in a water insoluble form or in a water soluble form. Exemplary pyrithione compounds include, for example, sodium pyrithione, zinc pyrithione, barium pyrithione, strontium pyrithione, copper pyrithione, cadmium pyrithione, and / or zirconium pyrithione. Other pyrithiones that may be present in the composition include sodium pyrithione, bismuth pyrithione, potassium pyrithione, lithium pyrithione, ammonium pyrithione, calcium pyrithione, magnesium pyrithione, silver pyrithione, gold pyrithione, manganese pyrithione, and / or an organic amine pyrithione. In a preferred embodiment, the pyrithione is a sodium pyrithione. A single pyrithione may be present as the preservative agent or a combination of any of the above may be included as the preservative agent.

[0034]

[0031] The pyrithione particles can have a particle size such that 100% of the particles have a particle size of less than about 10 microns and at least 70% of the particles have a particle size less than 5 microns, such as at least about 50% of the particles can have a particle size of 1 micron or less. Particle size can be measured using a laser scattering particle size analyzer, such as a HORIBA LA 910 particle size analyzer.

[0035]

[0032] The pyrithione particles can be produced by reacting pyrithione or a water- soluble salt of pyrithione, and a water-soluble polyvalent metal salt in a pressurized, turbulent flow reactor that generates pulverizing forces. The pulverizing forces produced by the pressurized, turbulent flow reactor efficiently generate pyrithione salt particles of micron size. The micron-sized pyrithione salt particles made by the method have a narrow and uniform size distribution, and have excellent surface deposition properties due to the large surface area provided by the population of micron particles.

[0036]

[0033] The pyrithione may be used in the end-use preservative composition formulation in amounts up to about 1 % by weight, such as up to about 0.75% by weight, such as up to about 0.50% by weight, such as up to about 0.25% by weight depending on the use of the end-use preservative composition.

[0037]

[0034] Pyrithione may also have other advantages in the end-use preservative composition, including antidandruff properties in hair care products.

[0038]

[0035] The preservative composition also comprises an organic acid or salt thereof. Exemplary salts include, for example, sodium benzoate and potassium sorbate. Other salts may also be used. In a preferred embodiment, the organic acid is lactic acid, levulinic acid, citric acid, or itaconic acid, their salts, or any mixture thereof. In another embodiment, the organic acid is itaconic acid. In an alternative embodiment, the organic acid comprises lactic acid, levulinic acid, citric acid, or mixtures thereof.

[0039]

[0036] The preservative composition may include from about 1 % by weight to about 5% by weight of an organic acid such as citric acid, succinic acid, phosphoric acid, lactic acid, or any mixture thereof. In another aspect, the composition may include from about 1 % by weight to about 5% by weight of an organic acid such as citric acid, succinic acid, phosphoric acid, lactic acid, or any mixture thereof, in combination with from about 0.05% by weight to about 5% by weight of another organic acid. In another embodiment, the composition may include from about 2% by weight to about 4% by weight of an organic acid such as citric acid, succinic acid, phosphoric acid, lactic acid, or any mixture thereof, in combination with from about 0.1 % by weight to about 4% by weight of another organic acid.

[0040]

[0037] Organic acid compounds can be present as preservatives in end-use preservative formulations in amounts up to about 3% by weight, depending on the particular organic acid compound. Similar amounts may be used for the salts. In most cases, the amount of the acid or salts thereof are used in amounts up about 1 % by weight, more typically up to about 0.6% by weight. Mixtures of organic acids may also be used in the preservative composition.

[0041]

[0038] The preservative composition also comprises an aromatic alcohol. Exemplary aromatic alcohols include, for example, phenoxyethanol, 2,4- Dichlorophenyl)methanol, benzyl alcohol, 1 -Phenoxypropaneol, chlorphenesin, and benzyl hemiformal. In a preferred embodiment, the aromatic alcohol is a phenoxyethanol, a benzyl alcohol, or mixtures thereof.

[0042]

[0039] The aromatic alcohols can be present in end-use formulations in amounts up to about 1 .5% by weight based on the total weight of the end-use formulation, depending on the particular alcohol compound. In most cases, the amount of the alcohol compounds are typically used in amounts up to about 1 % by weight, more typically up to about 0.5% by weight, based on the weight of the end-use formulation.

[0040] In a one embodiment of the preservative composition, the pyrithione comprises sodium pyrithione, the aromatic alcohol comprises phenoxyethanol, and the organic acid or salt thereof comprises citric acid, lactic acid, sodium benzoate, or potassium sorbate. In an alternative embodiment of the preservative composition, the pyrithione comprises sodium pyrithione, the aromatic alcohol comprises benzyl alcohol, and the organic acid or salt thereof comprises citric acid, lactic acid, sodium benzoate, or potassium sorbate.

In the preservative concentrate, the amount of the preservative agent will be higher than typical final use amounts. The preservative concentrate may have only the pyrithione, the organic acid or salt thereof, and the aromatic alcohol. Alternatively, the preservative concentrate may contain the pyrithione, the organic acid or salt thereof, the aromatic alcohol and a carrier that is compatible with end-use formulations. [0117]

[0120] The following examples illustrate the invention without limitation. All parts and percentages are given by weight unless otherwise indicated.

[0118]

[0121] To demonstrate the synergistic effect the following examples were performed.

[0119] Example No. 1

[0120]

[0122] The synergistic activity of pyrithione, organic acids or salts thereof, and aromatic alcohols are demonstrated as part of three-component composition.

[0121]

[0123] The sodium pyrithione used was Sodium OMADINE® obtained from Arxada AG.

[0122]

[0124] A wide range of concentrations were tested against: Pseudomonas aeruginosa ATCC 9027 / DSM 1128, and Candida albicans ATCC 10231 / DSM 1386.

[0123]

[0125] The lowest concentration of each mixture or single compound which prevents an observable level of growth was determined to be the minimum inhibitory concentration (MIC) value. The MIC value was determined for each microorganism.

[0124]

[0126] Serial dilutions of each mixture or single compound were prepared. Microorganisms were inoculated into media: Nutrient Broth (bacteria) or Malt Extract Broth (yeast). The final concentrations of micro-organisms in media were approximately 105 CFU / ml. The inoculated media was introduced to each of the serial dilutions at a 1 :1 ratio and incubated at 30±1 °C for 24-48 hours.

[0127] The MICs for each mixture were compared with the MIC of each single compound alone.

[0125]

[0128] Synergy was determined using a synergy index (“SI”) value, calculated using a method based on the Kull equation (F.C. Kull et. al., “Mixtures of Quaternary Ammonium Compounds and Long-chain Fatty Acids as Antifungal Agents”, Appl Microbiol. 1961 Nov; 9(6): 538-541 ):

[0126] SI = Qa / QA + Qb / QB + Qc / QC wherein

[0127] QA= concentration of compound A in ppm, acting alone produced an end point;

[0128] Qa = concentration of compound A in ppm, in the mixture, which produced an end point;

[0129] QB = concentration of compound B in ppm, acting alone produced an end point;

[0130] Qb = concentration of compound B in ppm, in the mixture, which produced an end point;

[0131] QC = concentration of compound C in ppm, acting alone produced an end point; and Qc = concentration of compound C in ppm, in the mixture, which produced an end point.

[0132]

[0129] In this study the end point is measured by the MIC. Synergy is demonstrated by an SI value of less than 1 . An additive effect is demonstrated by an SI value of equal to 1 . Antagonism is demonstrated by an SI value of greater than 1 .
