Triptonide is a diterpene triepoxide that is triptobenzene K in which the acylhydroquinone moiety has undergone oxidation to the corresponding triepoxyketone derivative. It has been isolated from the roots of Tripterygium wilfordii.
Disclosed herein is a method for preparing triptolide, wherein triptolide is prepared by means of highly stereoselective reduction of triptonide. Specifically, in a solvent, triptonide, as shown in formula II, is mixed with a reductant for reaction to give triptolide, as shown in formula I, wherein the reductant is MRxBHy or MAlHR'm(R"O)n. The method of the present invention features commercially available reagents, a simple process, and ease of industrialization.
Disclosed in the present invention are a derivative of triptolide and triptonide, and a synthesis method therefor and the use thereof. The derivative of the present invention has the structure as shown in general formula I, and the definition of each substituent is as described in the description and claims. The derivative of triptolide and triptonide of the present invention exhibits a high anti-tumor activity, and good development and application prospects.
