Benefits of technology

This patented technology uses techniques like mass spectrometry or protein analysis to identify specific types of molecules called histones (DNA) associated with infertility caused by male fertility disorders such as polycysemia and autism spectrum syndrome. By comparing these data from healthy individuals to those who have symptoms suggestive of impaired reproductivity, this research can help predict which men will develop serious problems later onset due to lacking egg whites at home during pregnancy.

Problems solved by technology

This patent discusses various aspects related with fertile impairment caused by reduced levels of testosterone due to lack therapy. These conditions can cause steroidal hypoplassis syndrome where certain types of cells cannot produce enough estrogen during menstrual cycles leading to decreased sexual functioning. Overall, these issues contribute towards improving diagnosis and prevention efforts aimed toward reducing infertorside symptoms associated with excessive estradiolysis.

Embodiment 1

[0026] 1. Detection method

[0027] In this example, a method for detecting oligoasthenospermia markers based on sperm proteomics is provided. The samples used in the following examples are all from the Affiliated Reproductive Hospital of Shandong University. %-17.3, the sperm concentration is between 3.8-14.8 million / mL, and the detection method includes the following steps:

[0028] (1) Extract the whole protein of sperm cells: Wash the sperm sample with DPBS, add RIPA lysate and sonicate for 1-2 minutes, incubate on ice for 30 minutes to lyse, centrifuge at 14,000g for 20 minutes, take the supernatant protein supernatant, and use the Bradford method to measure the supernatant Medium protein concentration;

[0029] (2) Enzymolyze the protein using the method based on ultrafiltration-assisted sample preparation (FASP): add 300 μg protein solution to a 10KD ultrafiltration tube, shake at 600 RPM for 1 min, centrifuge at 14000 g for 20 min, add 100 μL of 8M Urea, shake at 600 ...

Embodiment 2

[0048] Embodiment 2 Clinical detection verification

[0049] According to the method of Example 1, protein extraction and nanoflow liquid chromatography-tandem mass spectrometry were performed on the collected sperm of 10 groups of patients with oligoasthenospermia. Each set of data was repeated three times, and then the mass spectrometry data was analyzed. Table 1 shows the analysis results. Oligospermia and asthenozoospermia were detected in 9 persons using the above method. The accuracy rate of this group of data is as high as 90%.

Abstract

The invention belongs to the technical field of oligoasthenozoospermia and asthenozoospermia diagnosis, and particularly relates to application of MYH9 in preparation of a diagnostic reagent for severe oligoasthenozoospermia and asthenozoospermia. On the basis of a proteomics method, protein data in normal and severe oligoasthenozoospermia and asthenozoospermia samples are analyzed, and the protein abundance in the samples is detected by taking BSA protein signal intensity as a standard to obtain differential protein in normal and diseased samples. The MYH9 is detected and confirmed to be significantly up-regulated in severe oligoasthenozoospermia and asthenozoospermia samples through the method, has good credibility, is expected to be used as a detection marker of severe oligoasthenozoospermia and asthenozoospermia, and is applied to development of related detection reagents and kits.
