Abstract
Description
Technical Field
[0001] The invention relates to the field of pharmaceutical chemistry, and in particular to a method for preparing butylated hydroxyanisole. Background Art
[0002] Butylated hydroxyanisole (BHA), also known as butylated hydroxyanisole, has been used as an antioxidant in food, cosmetics, feed and pharmaceutical industries since 1941. BHA consists of two isomers, 3-tert-butyl-4-hydroxyanisole (3-BHA) and 2-tert-butyl-4-hydroxyanisole (2-BHA). The phenolic hydroxyl group of 3-BHA is affected by the steric hindrance of the tert-butyl group, and its antioxidant performance is 1.5-2 times higher than that of 2-BHA. BHA has been included in the Chinese, American and European Pharmacopoeia. The Chinese Pharmacopoeia (2020 edition) requires that the 2-BHA content shall not exceed 10.0%, while the European Pharmacopoeia (EP 10.0) requires that the 2-BHA content shall not exceed 10.0% of the 3-BHA content. The Joint Expert Committee on Food Additives (JECFA) under the Food and Agriculture Organization of the United Nations and the World Health Organization stipulates that the 3-BHA content in BHA shall not be less than 85%. The melting point of 3-BHA (99.5%) is 60°C-61.5°C, which is higher than the melting point of 2-BHA (99.0%) of 50-51.5°C. 3-BHA is more conducive to eliminating free radicals, has better antioxidant activity, and has good physical and chemical stability. Its stability is mainly reflected in its good fluidity, not easy to agglomerate, and not easy to change color in the environment.
[0003] At present, the research direction of BHA synthesis is to reduce the generation of 2-BHA and other impurities. There are two main synthesis methods: one is to use p-hydroxyanisole as substrate, tert-butyl alcohol, methyl tert-butyl ether and isobutylene as alkylating agents, and synthesize BHA in the presence of a catalyst. The commonly used catalysts are proton acids or Lewis acids, such as H2SO4, H3PO4, ion exchange resins, BF3, ZnCl2, solid heteropoly acids, etc. US Patent US2410902 uses n-hexane as solvent and H3PO4 as catalyst to react p-hydroxyanisole with tert-butyl alcohol or isobutylene at 50°C to obtain a product containing 75% BHA and 15% impurity 2,5-di-tert-butyl-4-hydroxyanisole. Chinese patent CN108238857 reported the use of p-hydroxyanisole, tert-butyl alcohol, phosphoric acid to synthesize BHA in a ratio of 200:90:60 and 300:110:80. In addition, an appropriate amount of concentrated sulfuric acid was added at a temperature of 30°C-40°C. The product yield and isomer content were not reported, but it can be seen that more proton acid catalysts were used in the reaction. Xia Yingzi et al. used p-hydroxyanisole as a substrate, methyl tert-butyl ether as an alkylating agent, a cation exchange resin as a catalyst, toluene and butanone (volume ratio 5:1) as solvents, and a temperature of 80-90°C to obtain a BHA yield of 47.1% and a 3-BHA content of 89.1%. U.S. Patent No. 2,616,931 uses p-hydroxyanisole and butanol in a molar ratio of 2:1, SiO2-AlO3 as a catalyst, and reacts at 150° C. under pressure to obtain 88% 3-BHA and 12% 2-BHA. This method uses a proton acid as a catalyst, which is used in a large amount and has poor reaction selectivity. The catalyst used is a Lewis acid, which has a small source of catalysts and harsh reaction conditions, and is not suitable for large-scale production.
[0004] The second method for synthesizing BHA is to use tert-butylhydroquinone (TBHQ) as a substrate, dimethyl sulfate, methyl iodide, and dimethyl carbonate as alkylating agents to synthesize BHA, or to react with methanol in the presence of a catalyst to obtain BHA. U.S. Patent No. 2776321 uses TBHQ as a substrate, xylene as a solvent, zinc powder and water, and dimethyl sulfate as a methylating agent, and reacts at 90.5°C-91.5°C to obtain 89.5% BHA, of which the 3-BHA content is 72% and the 2-BHA content is 28%. British Patent No. GB1366441 also uses TBHQ as a substrate, dimethyl sulfate as an alkylating agent, and reacts at 30-40°C to obtain a product BHA yield of 84.1%, of which the 3-BHA content is 92.8% and the 2-BHA content is 5.3%. Chinese patent CN 108314609 uses tert-butylhydroquinone (TBHQ) and dimethyl carbonate (DMC) with low toxicity as raw materials, uses self-made double alkali metal as catalyst, and synthesizes BHA at a temperature of 180°C and a pressure of 1.2MPa. The conversion rate of TBHQ is only 28-40%, and the selectivity of the target product 3-tert-butyl-4-hydroxyanisole (3-BHA) is 84-100%. Chinese patent CN101613260 uses TBHQ as a substrate, methanol as a solvent and an alkylating agent, adds concentrated sulfuric acid and metal chloride for reflux reaction, and extracts to obtain a BHA content of 90-94%, the isomer content is not reported, and the yield is 78-81%. This method uses highly toxic dimethyl sulfate and methyl iodide, or generates methyl sulfate substances during the reaction process, which is not friendly to the human body and the environment; using dimethyl carbonate with low toxicity, the conversion rate is low and the cost is high.
[0005]
[0006] The methods for preparing butylated hydroxyanisole (BHA) disclosed in the above prior art have at least one of the following defects:
[0007] (1) The reaction process uses a large amount of protonic acid or highly toxic alkylating agent;
[0008] (2) The reaction produces toxic substances, which are unfriendly to the human body and the environment;
[0009] (3) Poor reaction selectivity and low conversion rate;
[0010] (4) The reaction product is impure, producing 2-BHA or 2-substituted products.
[0011] Therefore, there is still a need to develop a method for preparing butylated hydroxyanisole (3-BHA) that is environmentally friendly, highly selective and has good product purity. Summary of the invention
