Description

Technical Field

[0001] The present invention relates to the technical field of tyrosinase inhibition, and more particularly to tyrosinase antibodies for inhibiting tyrosinase and uses thereof. Background Art

[0002] The skin color of a person is different according to many factors, and is mainly determined by the amount of melanin, carotene and hemoglobin, wherein melanin is the most decisive factor. Melanin is synthesized in melanocytes present in the skin base, and is transferred to peripheral keratinocytes to present the skin color of a person. It is known that when melanin is abnormally less, skin lesions such as vitiligo can be induced. On the contrary, when melanin is overproduced, spots and blemishes can be formed. Melanin is generated by tyrosinase acting on tyrosine as an amino acid, and tyrosinase is more activated by ultraviolet rays, therefore, if exposed to a large amount of sunlight, the skin will darken. Therefore, the basic mechanism of the medicament for developing whitening cosmetics for preventing pigmentation can include inhibiting the activity of tyrosinase, inhibiting the generation of tyrosinase, inhibiting the intermediate factor (mediator) that promotes the generation of melanin, inhibiting the reduction and photooxidation of the melanin generated, promoting the excretion of melanin and blocking ultraviolet rays, etc. Among them, the method by inhibiting the activity of tyrosinase can be said to be one of the methods that can obtain the simplest and effective skin whitening effect.

[0077] Example 1 Screening of α-Tyrosinase scFv Antibodies

[0078] 1. Perform Biopanning

[0079] Using a 7.6×10 9Biopanning was performed on the OPAL database of diversity. 5 μg of tyrosinase antigen was immobilized on epoxy magnetic beads and reacted with input phage. Phage reacted with antigen were eluted and the output titer was measured. Input and output titers were measured each time to obtain information on biopanning and to confirm whether biopanning was performed normally. 2×10 12 The phage with colony count / ml ≥ was taken as the input phage. It can be seen that the output increased with the progress of bio-panning from 1.7×10 6 Colony count / ml, 2nd time 3.65×10 6 Colony count / ml, 3rd time 4.68×10 8 Therefore, it can be confirmed that bio-panning is proceeding normally.

[0080] 2.ELISA analysis

[0081] ELISA analysis was performed to screen for highly sensitive and highly specific antibodies. The obtained phage was used to infect E. coli and smeared on an LB plate supplemented with antibiotics. After culturing in an incubator at 30°C for 16 hours, the generated colonies were randomly collected. After each colony was cultured in LB medium, it was treated with isopropyl-β-D-thiogalactoside (IPTG) to express scFv, and the E. coli was lysed to extract the water-soluble fraction, and ELISA was performed. First, 30 ng of tyrosinase antigen was fixed on a 96-well ELISA plate and blocked with phosphate buffered saline (PBS) containing 3% bovine serum albumin (BSA). After 1 hour, it was treated with the cell lysate obtained above and reacted at 37°C for 2 hours. After washing the plate three times with PBS containing 0.1% Tween 20, HRP conjugated anti-HA antibody was diluted 1:1000 in blocking solution and reacted at 37°C for 1 hour. The plate was washed five times with PBS containing 0.1% Tween 20, then developed with TMB substrate, and the scFv antibody bound to the antigen was measured using an ELISA reader. 96 antibodies were analyzed, and 11 antibodies (OD 0.4 and OD 0.1) were arbitrarily selected as the positive guideline and the negative guideline, respectively. Figure 1 ).

[0082] Example 2 Sequencing of α-tyrosinase scFv antibody

[0083] For the 11 tyrosinase scFv positive clones screened by ELISA analysis, individual clones were screened by sequencing. Since there may be duplicate clones in the positive clones screened previously, sequence analysis is required. According to the sequence analysis results, 2 of the 11 positive clones were confirmed to be individual clones.

[0084] Example 3 Purification and sensitivity analysis of the screened α-tyrosinase antibodies

[0088]

[0089] 2. Sensitivity analysis of purified α-tyrosinase scFv antibody

[0090] Sensitivity analysis was performed using purified α-tyrosinase scFv antibody. The sensitivity analysis utilized ELISA analysis. First, various concentrations of tyrosinase antigen were fixed to the ELISA plate, and then 10μg / ml of α-tyrosinase scFv antibody was treated. The analysis was also performed using HRP-labeled anti-HA antibody diluted at a ratio of 1:1000. The analysis results were EC50-1368ng / ml, LOD-12.5ng / ml ( Figure 2 ).

[0091] Example 4 Construction of cell-permeable α-tyrosinase scFv

[0092] 1. Construction of TAT-α-tyrosinase scFv construct

[0093] The polymerase chain reaction (PCR) product of the previously screened scFv insertion restriction enzyme site and TAT was produced using primers. 30 μl of each PCR product was treated with 4 μl buffer 3.1, 1 μl EcoRI, 1 μl XhoI, and 4 μl distilled water, and after reacting at 37°C for 1 hour, the DNA was separated to ensure the insert to be inserted into the vector. For the pET 28a (+) vector, after transformation to DH5α competent cells, it was smeared on an LB plate supplemented with kanamycin, cultured at 37°C for 16 hours, and colonies were extracted the next day and inoculated in LB medium supplemented with kanamycin, and then cultured at 37°C for 16 hours. The next day, after isolating the vector using a mini-prep kit, 30 μl of the vector was treated with 4 μl of buffer 3.1, 1 μl of EcoRI, 1 μl of Xho I, 1 μl of CIP, and 3 μl of distilled water, and purified after reacting at 37°C for 1 hour. The concentrations of the purified vector and insert were measured by a spectrophotometer (nanodrop), and T4 ligase was used to connect at room temperature for 16 hours in the ratio of vector and insert. After completing the connection (ligation), after transformation to DH5α, it was smeared on an LB plate supplemented with kanamycin and cultured at 37°C for 16 hours. The colony was extracted the next day and inoculated into an LB medium supplemented with kanamycin and cultured at 37°C for 16 hours. The next day, in order to confirm whether the insert was inserted, the plasmid was isolated and restricted with EcoRI and XhoI, and the band was confirmed by electrophoresis on 1% agarose gel.

[0094] 2. Purification of TAT-α-tyrosinase scFv antibody

[0095] The cloned TAT-α-tyrosinase scFv antibody clone was transformed into a BL21 (DE3) E. coli host, and the transformant was induced with 0.5 mM IPTG. The scFv antibody was suspended in a lysis buffer (50 mM Tris-HCl, pH 7.5, 150 mM NaCl), crushed using an ultrasonic pulverizer, and then centrifuged to obtain a supernatant. The protein was purified using a resin with affinity for Ni-NTA. Good expression was confirmed by SDS-PAGE analysis. The protein size was confirmed to be about 30 kDa ( Figure 3 ).

[0096] Example 5 Cytotoxicity assay of cell permeable anti-tyrosinase scFv

[0097] In order to understand the cytotoxicity of TAT-α-tyrosinase scFv antibody, mouse-derived malignant melanoma cells B16F10 were cultured in a CO2 incubator at 37°C. After culture, TAT-α-tyrosinase scFv antibody was treated with different concentrations (0.1ppm, 1ppm, 5ppm, 10ppm, 50ppm, 100ppm), and then further cultured under the same culture conditions. After the culture was completed, MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) solution was added for culture, and then the culture medium was removed, dimethyl sulfoxide (DMSO: Dimethyl sulfoxide) was added, and after appropriate shaking, the absorbance was measured at 570nm using an ELISA reader system. The values are shown in Table 2. As shown in Table 2, when TAT-α-tyrosinase scFv was treated at concentrations of 0.1ppm, 1ppm, 5ppm, 10ppm, 50ppm, and 100ppm, it was confirmed that the cell shape and cell survival rate of malignant melanoma cells were almost unchanged up to 100ppm. Therefore, it can be seen that TAT-α-tyrosinase scFv is a substance with no cytotoxicity up to a concentration of 100ppm.

[0110] Example 9 Skin penetration analysis of cell permeable anti-tyrosinase scFv

[0111] In order to confirm the skin penetration of TAT-α-tyrosinase scFv, it was confirmed by tissue staining using pig skin. Pig skin was treated with 10μg of TAT-α-tyrosinase scFv antibody and incubated in a CO2 incubator at 37°C for 16h. After fixing the pig skin with 4% formaldehyde, paraffin blocks were prepared and the tissue was cut 5μm using a microtome. After that, the tissue was mounted on a slide, the paraffin was removed and after a hydration process, the tissue section was blocked for 30min. The blocked tissue was reacted with anti-His HRP antibody at room temperature for 1 hour. After washing with PBST, it was reacted with DAB chromogen solution for 5min, then stained by H&E staining method and observed with an optical microscope. Figure 5 The results of comparison of the skin permeability of α-tyrosinase scFv and TAT-α-tyrosinase scFv are shown. Figure 5 As shown, it was confirmed that α-tyrosinase scFv was not delivered to pig skin tissue at all, while TAT-α-tyrosinase scFv had effective skin delivery ability.
