[0010] Technical Solution

[0011] In one aspect, a porous silica is provided which is impregnated with an extract of a strain of Staphylococcus epidermidis keratin.

[0012] As long as it belongs to the species of Epidermidibacterium keratini, the type of the strain is not limited. For example, the strain may be the strain of Epidermidibacterium keratini in Korean Patent Gazette KR10-1910791, but is not limited thereto. The strain may be a strain deposited with the accession number KCCM11843P. The strain deposited with the accession number may be the strain of Epidermidibacterium keratini EPI-7.

[0013] The strain may include a 16S rRNA having a sequence identity of about greater than or equal to 95%, about greater than or equal to 96%, about greater than or equal to 97%, about greater than or equal to 98%, about greater than or equal to 99%, about greater than or equal to 99.5%, or about greater than or equal to 99.9% with a 16S rRNA sequence of Staphylococcus keratin (Epidermidibacterium keratini) EPI-7 strain (Accession No.: KCCM11843P). The strain may include a 16S rRNA sequence of Staphylococcus keratin (Epidermidibacterium keratini) EPI-7 strain (Accession No.: KCCM11843P).

[0014] The term "sequence identity" refers to the degree of identity of the amino acid residues or bases between sequences when two sequences are matched to maximize consistency in a specific comparison region. Sequence identity can be confirmed by methods known in the art. The percentage of sequence identity can be confirmed by using known sequence alignment programs, for example, BLASTN (NCBI), CLC Main Workbench (CLC bio), MegAlign™ (DNASTAR Inc), etc.

[0015] The extract can be a lysate, culture solution, fermentation product or bacterial extract of the Epidermidibacterium keratini strain.

[0016] The term "lysate" can be used interchangeably with "lysate" and can refer to a product obtained by breaking the cell wall of a strain by chemical or physical forces. The lysate can include the lysate itself, its concentrate or lyophilized product.

[0017] The term "culture solution" may be used interchangeably with "culture supernatant", "conditioned culture solution" or "conditioned medium", and may refer to all culture media including the strain, its metabolites, and remaining nutrients, etc., obtained by culturing the strain for a predetermined period in a culture medium capable of providing nutrients to allow the strain to grow and survive in a test tube. In addition, the culture solution may refer to a culture solution in which bacterial cells are removed from a bacterial culture solution obtained after culturing the strain. On the other hand, the liquid from which bacterial cells are removed from the culture solution is called "supernatant", which can be obtained by obtaining only the upper layer liquid excluding the precipitated portion after leaving the culture solution to stand for a predetermined time, or removing the bacterial cells by filtration, or removing the precipitate at the bottom by centrifuging the culture solution and obtaining only the upper liquid.

[0018] The culture medium and culture conditions for culturing the epidermidis keratin (Epidermidibacterium keratini) strain can be used by those skilled in the art after appropriate selection or modification. For example, the culture solution can be obtained by culturing in a culture medium (e.g., R2A culture medium or TSB culture medium) at any temperature above 10° C. and below 40° C. for a predetermined time (e.g., 4 to 120 hours).

[0019] The culture solution may include the culture solution itself obtained after culturing the strain, a concentrate or a lyophilized product thereof, or a culture supernatant obtained after removing the strain from the culture solution, a concentrate or a lyophilized product thereof.

[0020] The term "fermentation product" may refer to any substance obtained by a metabolic process in which a chemical change occurs in a microorganism due to the action of the microorganism. The fermentation product may refer to a fermentation broth obtained by the fermentation metabolism of the strain or a component thereof. The fermentation product may include the fermentation product itself, a concentrate thereof, or a lyophilized product thereof.

[0021] The term "bacterial body" refers to the strain itself of the present invention, and includes the strain itself screened after separation from a sample, etc., or the strain separated from the culture solution after culturing the strain. The bacterial body can be obtained by taking the sediment at the bottom after centrifuging the culture solution, or by removing the upper liquid after leaving the culture solution for a predetermined time using the principle of precipitation to the bottom of the culture solution due to gravity.

[0022] Regardless of the extraction method, extraction solvent, extraction conditions, extracted components or the form of the extract, the term "extract" includes substances obtained by extracting the components of the strain, and also includes substances obtained by processing or treating the components of the strain by other methods. For example, the processing or treatment may include dilution, concentration, drying, purification, fractionation, filtration, fermentation, enzyme treatment, etc. Therefore, the extract may include an extract, a diluted or concentrated extract, a dry product obtained by drying the extract, or their crude products or purified products, and fractions obtained by fractionating them.

[0023] The extraction method is not limited to a specific extraction method, and for example, it may be stirring extraction.

[0024] The extraction solvent may be oil. By using oil as the extraction solvent, water-insoluble components can be extracted from the strain.

[0025] In a specific embodiment, the oil can be at least one selected from the group consisting of rice bran oil, medium-chain triglycerides (MCT) oil, soybean oil, sunflower seed oil, jojoba oil, mineral oil, squalane, caprylic triglyceride / capric triglyceride, cetyl ethylhexanoate, dodecane, isododecane and hydrogenated polydecene, but is not limited thereto. The medium-chain triglyceride oil is also called liquid coconut oil. The oil can be rice bran oil.

[0026] The extract may be an oil extract of a strain culture solution. Based on the total volume of the culture solution, the amount of oil used as the extraction solvent may be 0.01 to 100 times, 0.01 to 50 times, 0.01 to 20 times, 0.01 to 10 times, 0.01 to 5 times, 0.1 to 100 times, 0.1 to 50 times, 0.1 to 20 times, 0.1 to 10 times, 0.1 to 5 times, 0.1 to 1 times, 0.5 to 100 times, 0.5 to 50 times, 0.5 to 20 times, 0.5 to 10 times, 0.5 to 5 times, or 0.5 to 1 times, but is not limited thereto.

[0027] The extraction conditions refer to the conditions for extracting strain components such as extraction time and extraction temperature. The extraction time can be 30 minutes to 120 hours, 30 minutes to 100 hours, 30 minutes to 80 hours, 30 minutes to 60 hours, 2 hours to 120 hours, 2 hours to 100 hours, 2 hours to 80 hours, 2 hours to 60 hours, 10 hours to 120 hours, 10 hours to 100 hours, 10 hours to 80 hours or 10 hours to 60 hours, but can be appropriately selected according to other conditions such as extraction solvent and extraction temperature. [0035] The porous silica may have a uniform particle size. The porous silica may have excellent pore consistency.

[0036] The surface of the porous silica may be coated with a hydrophobic substance. The porous silica may include an organic coating that imparts hydrophobicity to the surface. When the organic coating is present, it may be applied to the inorganic coating or directly to the silica.

[0037] The porous silica coated with the hydrophobic material can effectively impregnate the water-insoluble component into the inner and outer surfaces of the porous silica, thereby impregnating the water-insoluble component. When the porous silica is coated with a water-soluble polymer, the water-insoluble component may not be impregnated.

[0038] The hydrophobic coating of the porous silica can be, for example, organosilicon, silane, metal soap, titanate, organic wax and mixtures thereof. The hydrophobic coating can alternatively include fatty acids, for example, fatty acids containing 10 to 20 carbon atoms, including lauric acid, stearic acid, isostearic acid and salts of these fatty acids. The fatty acid can be isopropyl triisostearate.

[0039] The silicon hydrophobic coating of the porous silica can be polymethylsiloxane (Methicone), polydimethylsiloxane (Dimethicone) or a copolymer or mixture thereof. The silicon can be an organic silicon compound, for example, polydimethylsiloxane having a main chain of repeating -Me2SiO units ("ME" is methyl, CH3), methyl hydrogen polysiloxane having a main chain of repeating -MeHSiO units, and a RnOSiH (4-n)("R" is an alkyl group, and "N" is an integer of 1, 2 or 3).

[0040] The silane hydrophobic coating of the porous silica may be an alkoxysilane. The alkoxysilane may be triethoxyoctylsilane or a perfluoroalkylethyltriethoxysilane having a linear or branched C3 to C12 alkyl group.

[0043] In a specific embodiment, the surface of the porous silica may be coated with polymethylsiloxane, polydimethylsiloxane or a copolymer or mixture thereof. The surface of the porous silica may be coated with polymethylsiloxane.

[0044] When the silica particles are coated or "surface treated," the amount of the coating or treatment substance can be about 0.1 wt % to 35 wt %, about 0.1 wt % to 30 wt %, about 0.1 wt % to 20 wt %, about 0.1 wt % to 10 wt %, about 0.1 wt % to 5 wt %, or about 0.1 wt % to 1 wt % of the silica, but is not limited thereto.

[0045] The epidermidis Staphylococcus keratin keratini) extract can be impregnated into porous silica with an oil absorption amount of 10Ml / 100g to 500Ml / 100g, 10Ml / 100g to 400Ml / 100g, 10Ml / 100g to 300Ml / 100g, 20Ml / 100g to 500Ml / 100g, 20Ml / 100g to 400Ml / 100g, 20Ml / 100g to 300Ml / 100g, 50Ml / 100g to 500Ml / 100g, 50Ml / 100g to 400Ml / 100g, 50Ml / 100g to 300Ml / 100g, or 60Ml / 100g to 270Ml / 100g, but is not limited thereto.

[0046] The silica particles coated with a hydrophobic substance can effectively and stably store active ingredients with high skin absorption rates such as essential fatty acids and lipids due to their microporous structure. When water-insoluble ingredients are impregnated into the porous silica particles, other compositions in storage are less affected, so that they can be stably formulated. In addition, a refreshing and lighter feeling can be provided by eliminating the sticky feeling peculiar to water-insoluble ingredients.

[0047] In one example, it was confirmed that the porous silica impregnated with the keratin oil extract of Staphylococcus epidermidis provides a refreshing and lighter feeling by eliminating the slimy feeling peculiar to water-insoluble substances, thereby providing excellent skin feel such as refreshing feeling, moist feeling, and tight feeling when used.

[0048] In addition, due to the characteristics of the silica particles themselves, the porous silica can have additional effects. For example, when silica is applied to the skin, the particles easily disintegrate and fill fine lines and pores, thereby providing a moist and close skin feel. In addition, due to the low refractive index of silica itself, it can cover curved surfaces while providing a translucent appearance. Further, when the water-insoluble components impregnated into the porous silica are absorbed by the skin, the oil absorption capacity of the porous silica is restored, so that the over-secretion of the cortex can be absorbed, thereby preventing stickiness.

[0049] Another aspect provides a cosmetic composition comprising the porous silica impregnated with an extract of an Epidermidibacterium keratini strain according to one aspect.

[0050] The cosmetic composition has a skin improvement effect. Specifically, the skin improvement may include improving skin condition, improving skin beauty, preventing, improving or treating skin diseases. The skin improvement may be any one or more of skin anti-aging, improving skin wrinkles, enhancing skin barrier, skin moisturizing, enhancing skin elasticity, improving skin keratin, improving skin gloss, improving skin transparency and improving skin brightness. Therefore, the cosmetic composition can be used for skin anti-aging, improving skin wrinkles, enhancing skin barrier, skin moisturizing, enhancing skin elasticity, improving skin keratin, improving skin gloss, improving skin transparency or improving skin brightness.

[0051] The term "skin aging" refers to the visible and invisible changes that occur in the skin with age, such as thinning of the epidermis, decreased number of cells or blood vessels in the dermis, decreased ability to repair DNA damage, shortened cell renewal cycle, decreased wound recovery ability, decreased skin barrier function, decreased ability of the epidermis to retain moisture, decreased sweat secretion, decreased sebum secretion, decreased ability to produce vitamin D, decreased protection against physical damage, decreased ability to eliminate chemicals, decreased immune response, decreased sensory function, and decreased ability to regulate body temperature.

[0052] The composition can be used to prevent skin aging or anti-aging caused by exogenous or endogenous factors. The exogenous factors refer to various external factors (e.g., ultraviolet rays (light)), and the endogenous factors are also called timing factors, which mainly refer to factors caused by the passage of time. That is, the skin aging not only includes early aging symptoms caused by external stimuli such as ultraviolet rays, pollution, cigarette smoke, and chemical substances, but also includes the natural aging phenomenon caused by the decrease in skin cell proliferation with age. It is a concept including wrinkles, decreased elasticity, sagging skin, and dryness. In addition, wrinkles may include wrinkles caused by changes in internal and external factors that cause stimuli to change the components that constitute the skin tissue.

[0053] The aging can be photoaging. The term "photoaging" is a phenomenon caused by external environmental factors, the most typical of which is ultraviolet rays. Ultraviolet rays cause damage to biological components, for example, activating protease, chain breakage of matrix proteins, and abnormal cross-linking, and the repetition of this mechanism can lead to obvious skin aging in appearance. Therefore, the composition can be used to prevent or improve photoaging caused by ultraviolet B (UVB).

[0054] The term "improving skin wrinkles" is interpreted as meaning an effect of reducing the number of skin wrinkles or reducing the depth of wrinkles, and also includes inhibiting skin aging by improving wrinkles.

[0055] The term "enhancing the skin barrier" may refer to any effect in which the function of the skin barrier located in the outermost layer of the skin to prevent the loss of water and nutrients is enhanced.

[0056] The term "skin moisturizing" may refer to any effect that retains moisture in the skin or prevents moisture loss. Skin moisturizing effects may help improve skin wrinkles and enhance skin elasticity.

[0057] The term "enhancing skin elasticity" may refer to any action in which skin elasticity decreased due to aging or the like is enhanced or in which a decrease in skin elasticity is prevented.

[0058] The term "skin keratin" refers to foreign matter produced when aged cells die and fall off from the epidermis of the skin, etc.

[0059] The term "exfoliating the skin" is used interchangeably with "exfoliating the skin" and may mean removing or reducing the amount of skin cell.

[0060] The term "improving skin gloss" may refer to any effect of increasing skin gloss, or restoring reduced gloss. For example, the skin gloss may be measured using a skin gloss index (Gloss Value) evaluation device such as a glossymeter.

[0061] The term "improving skin transparency" may refer to any effect of increasing skin transparency or restoring reduced transparency. For example, the skin transparency may be measured using a skin transparency evaluation device such as a translucency meter.

[0062] The term "improving skin brightness" may refer to any effect of increasing skin brightness or improving dull skin brightness. For example, the skin brightness may be measured using a skin brightness evaluation device such as a Chromameter.

[0063] The term "skin disease" may be a disease caused by skin aging or damage to the skin barrier function. The term "prevention" may include inhibiting the occurrence of a disease. The term "treatment" includes inhibiting, alleviating or eliminating the development of a disease. The term "improvement" may refer to parameters related to the alleviation or treatment of symptoms, for example, any action that at least alleviates the degree of a symptom.

[0064] According to one aspect, the composition can increase the biosynthesis of collagen or reduce the biosynthesis of collagenase. For example, the composition can increase the expression of COL1A1 or reduce the expression of MMP-1. Therefore, the composition can have the effect of preventing skin aging and improving wrinkles.

[0065] According to one aspect, the composition can improve hyaluronic acid synthesis, or improve filaggrin synthesis. For example, the composition can improve the expression of HAS and filaggrin. In addition, the composition can reduce transepidermal water loss or increase skin moisture content. Therefore, the composition can have the effect of enhancing skin barrier and skin moisturizing.

[0066] The results of the evaluation of epidermal elasticity by a cutometer after use confirmed that the composition according to one aspect improves epidermal elasticity. Therefore, the composition may have an effect of enhancing skin elasticity.

[0067] The results of corneum index evaluation by a transparent polyester film tape (Corneofix) and a skin wrinkle tester (Visioscan) after use confirmed that the composition according to one aspect reduced the corneum index of the skin. Therefore, the composition can have the effect of improving the corneum of the skin.

[0068] The skin gloss index evaluation result of the glossymeter after use confirmed that the composition according to one aspect improves the skin gloss index. Therefore, the composition can have the effect of improving skin gloss.

[0069] The skin transparency evaluation result of the translucency meter after use confirmed that the composition according to one aspect reduced the reduction ratio of the amount of transmitted light. Therefore, the composition can have the effect of improving skin transparency.

[0070] The skin brightness evaluation result of the skin color tester (Chromameter) after use confirmed that the composition according to one aspect improves skin brightness. Therefore, the composition can have the effect of improving skin brightness.

[0071] The composition may include 0.001 wt % to 90 wt %, 0.001 wt % to 80 wt %, 0.001 wt % to 70 wt %, 0.001 wt % to 60 wt %, 0.001 wt % to 50 wt %, 0.001 wt % to 40 wt %, 0.001 wt % to 30 wt %, 0.001 wt % to 20 wt %, 0.001 wt % to 10 wt %, 0.01 wt % to 50 wt %, 0.01 wt % to 40 wt %, 0.01 wt % to 30 wt %, 0.01 wt % to 20 wt %, 0.01 wt % to 10 wt %, 0.01 wt % to 5 wt %, 0.01 wt % to 3 wt %, or 0.01 wt % to 1 wt % of porous silica according to an aspect, based on the total weight of the composition.

[0072] The term "included as an active ingredient" means that the porous silica of the present specification is added to such an extent that the above-mentioned results can be exhibited. In addition, this may include adding various ingredients as sub-ingredients and formulating in various forms for the delivery and stabilization of drugs.

[0073] The cosmetic composition can be prepared in any conventional dosage form. For example, the cosmetic composition can have a toner, a face cream, an essence, a cleansing foam, a makeup remover, a facial mask, an ampoule, a moisturizer, a moisturizing oil, a body lotion, a shampoo, a conditioner, a hair serum, a hair spray, a foundation, a lipstick, a mascara, a primer, or a skin-friendly cosmetic dosage form.

[0109] Hereinafter, the present invention will be further described in detail by way of examples. However, these examples are only used to exemplify the present invention, and the present invention is not limited thereto.

[0110] Example 1. Preparation of oil extract of keratinized Staphylococcus epidermidis strain

[0111] The oil was used as an extraction solvent to prepare an extract of the keratinogenic strain of Staphylococcus epidermidis.

[0112] Specifically, after culturing the keratinized Staphylococcus epidermidis strain in TSB medium, rice bran oil was added in an amount of half the volume of the culture medium and vigorously stirred. After standing at room temperature for 24 hours, the rice bran oil layer was recovered to obtain the rice bran oil extract of the keratinized Staphylococcus epidermidis strain.

[0113] Example 2. Preparation of porous silica impregnated with keratin strain extract of Staphylococcus epidermidis

[0114] The rice bran oil extract of the keratin strain of Staphylococcus epidermidis obtained in Example 1 was impregnated into porous silica. Silica (silicone dioxide) coated with polymethylsiloxane (methicone) was used as the porous silica. The extract was impregnated into the porous silica at an oil absorption of 100Ml / 100g. The obtained porous silica impregnated with the keratin extract of Staphylococcus epidermidis was used in the following experimental examples.

[0115] On the other hand, in the following experimental examples, the same porous silica as in Example 2 was used as a control group except that it was not impregnated with the Staphylococcus epidermidis keratin extract.

[0116] Preparation Example 1. Preparation of cosmetic composition

[0117] An air cushion type cosmetic composition was exemplarily prepared, the cosmetic composition comprising porous silica impregnated with the keratin extract of Staphylococcus epidermidis in Example 2. The ingredients and contents (weight %) of the cosmetic composition are shown in Table 1 below.

[0123] Experimental Example 1. Sensory evaluation of improving skin feel

[0124] When a water-insoluble extract or water-insoluble substance derived from a microorganism is applied to the skin, there is often an uncomfortable feeling such as sliminess. Therefore, in order to evaluate the feeling of the porous silica impregnated with the keratin extract of Staphylococcus epidermidis in Example 2 on the skin during use, a sensory evaluation was conducted.

[0125] Specifically, a total of 5 evaluators evaluated the refreshing feeling, moist feeling, and close feeling of each sample using a 5-point scale. After scoring the skin feeling of each sample from 1 to 5 points, the proportion of respondents who scored 3 points or higher was calculated (1 point: very poor, 2 points: poor, 3 points: average, 4 points: good, 5 points: very good). The results are shown in Table 2 below.

[0126]

Table 2

[0127]

[0128] As shown in Table 2 above, the porous silica in Example 2 impregnated with the keratin rice bran oil extract of Staphylococcus epidermidis has a more excellent feel in use than the keratin rice bran oil extract of Example 1. In addition, the feel of use of the untreated porous silica is similar to that of Example 2. This means that the negative feel of use of the keratin rice bran oil extract of Staphylococcus epidermidis is offset by the positive feel of use of the porous silica. Therefore, it is confirmed that the porous silica impregnated with the keratin rice bran oil extract of Staphylococcus epidermidis in Example 2 provides a refreshing and lighter feel by eliminating the slimy feel peculiar to the non-water-soluble components derived from microorganisms, and has an excellent effect of improving the feel of use on the skin.

[0129] Experimental Example 2. Evaluation of anti-aging and wrinkle improvement effects

[0130] The effects of the porous silica impregnated with the keratin rice bran oil extract of Staphylococcus epidermidis in Example 2 on factors related to skin aging and wrinkle formation caused by ultraviolet (UV) irradiation were analyzed.

[0131] Specifically, human dermal fibroblasts (Hs68) were seeded into a 6-well plate at 3.5×105 and cultured in a culture machine at 37°C and CO2 for 24 hours. The culture medium was removed and DPBS was added, and then irradiated with 12 mJ / cm 2UVB or no irradiation. After UVB irradiation, DPBS was removed and replaced with a medium without FBS, and then the porous silica of Example 2 or the untreated porous silica (control group) was treated with 1% (w / v) and further cultured for 24 hours. Then, RNA was isolated from the cells of each sample using TRIZOL (RNAiso, DAKARA, Japan), and the RNA was quantified at 260nm using NanoDrop, and then 2μg of RNA was used to synthesize cDNA in an amplification instrument (C1000 Thermal Cycler, Bio-Rad, USA). Using the synthesized cDNA as a template, SYBR Green (SYBR Green supermix, Applied Biosystems, USA) was added to the target genes collagen synthase (COL1A1) and collagenase (MMP-1) together with primers and cDNA, and real-time polymerase chain reaction was performed in a real-time PCR instrument (Step One Plus, Applied Biosystems, USA). The expression levels of the genes were finally analyzed by correcting the expression levels of the genes based on the β-actin gene.

[0132] Figure 1 The results showing the relative expression level of collagen synthase (COL1A1) in fibroblasts after adding the porous silica of Example 2 or the control group.

[0133] Figure 2 The results are shown for the relative expression level of collagenase (MMP-1) in fibroblasts after adding the porous silica of Example 2 or the control group.

[0134] like Figure 1 and Figure 2 As shown, it was confirmed that the porous silica impregnated with the Staphylococcus epidermidis keratin rice bran oil extract in Example 2 significantly increased the expression of COL1A1 decreased by ultraviolet rays, and significantly decreased the expression of MMP-1 increased by ultraviolet rays.

[0135] This result indicates that the porous silica impregnated with the Staphylococcus epidermidis keratin rice bran oil extract has anti-aging (eg, photoaging) and wrinkle-improving effects.

[0136] Experimental Example 3. Evaluation of skin barrier enhancement and skin moisturizing efficacy

[0137] The effects of the porous silica impregnated with the Staphylococcus epidermidis keratin rice bran oil extract in Example 2 on skin barrier enhancement and skin moisturizing activity were analyzed.

[0138] Specifically, human keratinocyte HaCaT cells were cultured in DMEM medium (Dulbecco'smodified Eagle's Medium, Gibco 1210-0038) containing 10% bovine serum (fetal bovin serum, FBS). The cultures were carried out in a 37°C, 5% CO2 incubator. The cultured cell lines were further cultured for 24 hours after being treated with 1% (w / v) of the porous silica of Example 2 or untreated porous silica (control group). In addition to using primers for HAS3 (hyaluronate synthase), a factor related to skin barrier enhancement and skin moisturizing, and filaggrin, the expression levels of HAS3 and filaggrin were detected by the same method as in Experimental Example 2.

[0139] Figure 3 The results showing the relative expression level of hyaluronan synthase (HAS3) in keratinocytes after adding the porous silica of Example 2 or the control group.

[0140] Figure 4 The results showing the relative expression level of filaggrin in keratinocytes after adding the porous silica of Example 2 or the control group.

[0141] like Figure 3 and Figure 4 As shown, it can be seen that the porous silica impregnated with the keratin rice bran oil extract of Staphylococcus epidermidis in Example 2 significantly increased the expression levels of HAS3 and filaggrin.

[0142] This result indicates that porous silica impregnated with Staphylococcus epidermidis keratin rice bran oil extract has the effect of strengthening the skin barrier and skin moisturizing.
