[0053] In the present invention, a plant-derived resorcinol lipid is used as an active ingredient of the composition for improving liver function.
[0054] As described above, resorcinol lipids are lipophilic substances obtained by having alkyl groups at positions 2, 4, 5 and / or 6 of the aromatic ring of resorcinol (1,3-dihydroxybenzene). In nature, it is common in plants belonging to Anacardiaceae, Ginkgoaceae, Proteaceae, Myrsinaceae, Primulaceae, Myristicaceae, Iridaceae, Araceae, Leguminosae, Gramineae, etc., and Artemisia argyi of Asteraceae. Among these plants, the edible parts (seeds) of Gramineae plants such as wheat and rye contain relatively abundant resorcinol lipids, so they can be suitably used as the raw material for the resorcinol lipid of the present invention. For example, the bran or whole grains of the edible parts (seeds) of wheat, rye, etc. contain alkyl resorcinol compounds represented by the following formula (I) of about 0.015~0.3 mass %. In addition, as a preferred raw material for the resorcinol lipid of the present invention, fermented products such as miso with wheat or rye as raw materials can also be exemplified.

[0075] The plant-derived resorcinol lipid used in the present invention can be purchased from a commercial product or extracted from a plant by a conventional method. For example, a commercial product of the compound of formula (I) can be purchased from ReseaChem GmbH, SIGMA-ALDRICH, etc.
[0076] As a method for extracting from plants, there can be cited a method of immersing the raw material in a solvent while stirring it in an extraction solvent at room temperature or after heating under normal pressure or pressure as required, or a reflux extraction method, etc. Before adding to the extraction solvent, the raw material can be cut, crushed, squeezed, dried, or a combination thereof as required.
[0077] As the extraction solvent, there can be listed alcohols that are liquid at room temperature, such as methanol, ethanol, n-propanol, isopropanol, n-butanol and other lower alcohols, or 1,3-butanediol, propylene glycol, glycerol and other polyols; ethers such as diethyl ether and propyl ether; esters such as butyl acetate and ethyl acetate; ketones such as acetone and methyl ethyl ketone; hexane; and organic solvents such as chloroform. The above solvents can be used alone or in combination of two or more. Among the above solvents, alcohols that are liquid at room temperature, such as lower alcohols with 1 to 4 carbon atoms, are preferably used from the perspective of operability or the impact on the environment. From the perspective of the safety of the residual solvent, ethanol is more preferably used.
[0078] As the extraction solvent, an aqueous organic solvent containing an aqueous component in the organic solvent may also be further included. From the perspective of maintaining the extraction efficiency high, it is expected that the content of the aqueous component in the aqueous organic solvent is usually 50 volume % or less, preferably 30 volume % or less, and more preferably 20 volume % or less. As the aqueous organic solvent, preferably, an aqueous alcohol containing an aqueous component in the alcohols that are liquid at room temperature as described above can be cited, and more preferably, aqueous ethanol can be cited.
[0079] The conditions such as temperature and time for extraction can be appropriately set according to the type of extraction solvent used or the extraction conditions, and the preferred extraction temperature is 2 to 100 ° C, and the extraction time is about 30 minutes to 60 hours. Relative to 100 parts by mass of the raw material, it is preferred that the amount of the extraction solvent is set to about 200 to 2000 parts by mass. In addition, after the extraction treatment, the mixture containing the extract and the residue can be provided for filtration or centrifugation as needed to remove the solid components as the residue. Furthermore, the removed solid components can also be provided again for the extraction operation using the extraction solvent, and the operation can be repeated many times. The obtained extract can be passed through liquid chromatography, etc., thereby preparing a further purified extract containing resorcinol lipids. When purified in this way, for its degree of purification, in any scheme, based on the content of the above-mentioned plant-derived resorcinol oil, for example, typically can be in the range of 1% by mass or more, more typically can be in the range of 5% by mass or more, and further more typically can be in the range of 10% by mass or more. In addition, for example, the range can be typically 100% by mass or less, more typically 90% by mass or less, and further more typically 80% by mass or less. The obtained extract can be used directly in the present invention, or can be used in the present invention after further treatment such as concentration, freeze drying, hot air drying, pulverization, powderization, classification, dilution, mixing with water, etc. as required.
[0080] As shown in the examples described below, plant-derived resorcinol lipids have the effect of improving liver function. The present invention provides a composition for improving liver function based on the properties of the resorcinol lipids. In addition, "liver function" can include, for example, the improvement of symptoms of hepatitis such as non-alcoholic hepatitis, alcoholic hepatitis, viral hepatitis, etc. In addition, in addition to hepatitis, the improvement of symptoms of liver diseases such as non-alcoholic liver disease, fatty liver, cirrhosis, etc. can also be listed. However, the application of the composition for improving liver function of the present invention is not limited to these symptoms, and can be widely used for the purpose of improving liver function. Whether the symptoms are improved can be evaluated, for example, by measuring the aspartate aminotransferase (AST) value or alanine aminotransferase (ALT) value in plasma as a general indicator of liver function.
[0081] The dosage of the composition for improving liver function of the present invention can be appropriately changed according to the type, symptoms, age, sex, etc. of the applicable individual. Regarding the dosage when a human is used as the subject, 0.01 to 10 g of the above-mentioned plant-derived resorcinol oil can usually be given to an adult per day. The daily dosage can be given once or divided into multiple times. In addition, oral administration is more preferred. In addition, the applicable subject is not limited to humans, and for example, it can also be applied to animals such as dogs and cats.
[0082] The form of the composition for improving liver function of the present invention is not particularly limited as long as it is a form that can ingest the above-mentioned plant-derived resorcinol oil. For example, the form can be solid, semisolid or liquid, or can be various forms such as tablets, chewable tablets, powders, capsules, granules, drinkables, gels, syrups, tube feeding-enteral feeding liquid food, etc. In addition, for example, it can be a health food, a functional diet, a specific health-care diet, a diet for patients, a food additive for the diet, a medicine, a nutritional supplement, an animal feed for livestock, horse racing, ornamental animals, pets, etc. That is, it can be formed into a form that is easy to ingest by containing the above-mentioned plant-derived resorcinol oil as a part of each form. On the other hand, the composition for improving liver function of the present invention can be basically composed of the above-mentioned plant-derived resorcinol oil. Therefore, for example, in any form, the content of the plant-derived resorcinol oil may be, for example, typically in the range of 0.1% by mass or more, more typically in the range of 0.5% by mass or more, and further more typically in the range of 1.0% by mass or more. In addition, for example, typically in the range of 100% by mass or less, more typically in the range of 90% by mass or less, and further more typically in the range of 80% by mass or less.
[0083] As the form of typical medicine, for example, oral agents such as tablets, capsules, granules, powders, syrups, dry syrups, solutions, suspensions, inhalants, transdermal preparations, suppositories, enteral preparations, drips, injections and other non-oral agents can be listed. [0090] Example
[0091] Hereinafter, the present invention will be described in further detail with reference to Examples, but the present invention is not limited to the following Examples.
[0092] <Preparation Example 1> ~Preparation of alkylresorcinol~
[0093] Rye bran (about 9.4 kg) was dried under reduced pressure (80°C, 16 hours), and 5 times the amount of 99% ethanol was added to the dried raw material, and stirred and extracted under heating (60°C, 1 hour). Then, solid-liquid separation was performed to obtain filtrate A, and an equal amount of 99% ethanol was added to the residue, and further rinsed and solid-liquid separation was performed to obtain filtrate B. The obtained filtrate A and filtrate B were combined and concentrated under reduced pressure to obtain 516 g of rye bran ethanol extract.
[0094] 5.8 L of purified water and 5.8 L of 99% ethanol were added to 463 g of rye bran ethanol extract, and stirred on a 60°C water bath for 1 hour, and then 696 g of α-cyclodextrin was added, and further stirred and mixed for 1 hour. This was spray-dried to obtain 649 g of a spray-dried product, and the particle size was further adjusted (passed through a 10-mesh sieve) to obtain 639 g of a dry rye bran ethanol extract.
[0095] The dried product was dissolved in 2L of hexane and purified by normal phase silica chromatography. The chromatographic column used was HI-FLASH COLUMN SILICA 5L catalog number: W007 (chromatographic column inner diameter × chromatographic column length: 60×180mm, filling amount: 250g) manufactured by YAMAZEN CORPORATION. The chromatographic conditions were set to a flow rate of 70mL / min, a detection wavelength of 280nm, and a mobile phase (9 minutes of hexane / ethyl acetate: 90 / 10 (v / v) → 15 minutes of 80 / 20 (v / v) → 16 minutes of 60 / 40 (v / v) step gradient), and the eluent was separated during the elution time of 30 minutes to 40 minutes. The separated product was dried under reduced pressure to obtain 50g of a dry solid.
[0096] <Test Example 1> ~Analysis using high performance liquid chromatography~
[0097] The amount of alkylresorcinol in the dried solid obtained in Production Example 1 was quantitatively determined by an analysis method using high performance liquid chromatography. In addition, alkylresorcinol may be referred to as "ARs" or "AR" hereinafter.
[0098] Specifically, Inertsil ODS4 from GL Sciences Inc. was used as the analytical column, and the analytical conditions were a flow rate of 1 mL / min, a detection wavelength of 275 nm, a column temperature of 40°C, and a mobile phase (89 v / v% methanol → (5 minutes) → 92 v / v% methanol → (25 minutes) → 100% methanol gradient). Commercially available olivetol (AR with a linear alkyl chain of 5 carbon atoms: Sigma-Aldrich) was used as a standard for alkylresorcinol. For the quantification of each AR with a different alkyl chain length from olivetol, the elution position was determined in advance, and the value obtained by dividing the molecular weight of each by the molecular weight of olivetol was used as a coefficient, and the actual measured peak area was multiplied by the coefficient to obtain the result.
[0099] As a result, it was confirmed that 6.8 g of ARs were contained in 50 g of the dried solid obtained in Preparation Example 1.
[0100] <Test Example 2> ~Animal test using hepatitis model mice~
[0101] Nonalcoholic steatohepatitis model mice (manufactured by Charles River Laboratories Japan, Inc., C57BL / 6 NASH male mice) were moved to cages at 6 weeks of age, and then after a 2-day adaptation period, 6 mice were housed per cage. The mice were housed in a constant temperature and humidity chamber with a 12-hour light-dark cycle (light cycle: 8 am to 8 pm), and after the adaptation period, they were allowed to freely consume the feed corresponding to each test group shown below.
[0102] Control group (Group C): control feed without test substance added
[0103] Resveratrol ingestion group (R group): feed blended with 0.4% by mass of resveratrol (Tokyo Chemical Industry Co., Ltd.)
[0104] ARs high intake group (AH group): feed containing 0.9% by mass of ARs
[0105] ARs low intake group (AL group): feed containing 0.3% by mass of ARs
[0106] During the test period, body weight was measured once a day. In addition, blood was collected once a week from the tail vein of the mice to obtain plasma. The plasma was stored at -80°C before being used for measurement. In addition, after body weight was measured on the last day of the test, blood was collected under anesthesia with diethyl ether, and the liver was immediately removed after sacrifice, frozen with dry ice after weighing, divided into two halves, and placed in 1.5 mL tubes, respectively, and stored at -80°C.
[0107] Figure 1 The figure shows the changes in body weight gain of each group during the test period.
[0108] like Figure 1 As shown, during the test period, almost no weight gain was observed in the control group (C group). Since the mice used in this test example are mice in the growth period of 6 weeks old, weight will usually increase, and it is believed that since it is a hepatitis model mouse, the nutritional metabolism is abnormal and weight gain is suppressed. In contrast, in the ARs low intake group (AL group) and the ARs high intake group (AH group), weight gain was confirmed. It is believed that this is because hepatitis is suppressed by taking ARs, and the abnormal metabolism of nutrition is also improved as a result. On the other hand, in the intake group (R group) of resveratrol, which is known to have an inhibitory effect on fat accumulation in organs such as the liver, the increase in weight of the hepatitis model mice has not been improved. It is believed that this is due to the fact that the effect of ARs is not simply the effect of the result of improving the usual fat metabolism, but the effect of ARs peculiar to the symptoms of hepatitis.
[0109] on the other hand, Figure 2 The feed intake of each group during the experimental period is shown in FIG. Figure 2 As shown, no major differences in food intake were observed in any of the groups.
[0110] <Test Example 3> ~Measurement of AST and ALT values in plasma~
[0111] Aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels in plasma were measured as general indicators of liver function. The measurement was performed using Transaminase CII-Test Wako (manufactured by FUJIFILM Wako Pure Chemical Corporation) according to the instructions of FUJIFILM Wako Pure Chemical Corporation.
[0112] Figure 3 The changes in AST values of each group during the test period are shown in FIG.
[0113] like Figure 3 As shown, in the control group (Group C), the AST value increased from week 0 to week 4, confirming the development of hepatitis. In contrast, in the ARs low intake group (Group AL) and the ARs high intake group (Group AH), the increase in AST values was suppressed. The degree of suppression in the high intake group was more significant than that in the low intake group. On the other hand, in the resveratrol intake group (Group R), the level was almost the same as that of the control group (Group C).
[0114] Figure 4 The changes of ALT values in each group during the test period are shown in FIG.
[0115] like Figure 4 As shown, in the control group (C group), ALT values increased from week 0 to week 4, confirming the development of hepatitis. In contrast, in the ARs low intake group (AL group) and the ARs high intake group (AH group), the increase in ALT values was suppressed. The degree of suppression in the high intake group (AH group) was more significant than that in the low intake group (AL group). On the other hand, in the resveratrol intake group (R group), the level was almost the same as that of the control group (C group).
[0116] As above Figure 3 and Figure 4 As shown in the results of , it is known that ARs have an effect of inhibiting hepatitis, and this effect depends on the intake amount. The effect of ARs is a specific effect on hepatitis symptoms that resveratrol, which is known to generally inhibit fat accumulation in organs such as the liver, cannot exert.
[0117] <Test Example 4> ~Measurement of Neutral Fat Amount in Liver Tissue~
[0118] 50 mg of frozen liver tissue was placed in a tube, and 1 mL of a mixture containing chloroform and methanol in a liquid ratio of 2:1 was added thereto. Use a homogenizer to crush it into a paste, and use a shaker to stir the total mixture at room temperature for 15 to 20 minutes. The homogenate was centrifuged at 15000 rpm for 5 minutes and the supernatant was recovered. 0.5 mL of physiological saline was added to the recovered supernatant and mixed for a few seconds. The mixture was centrifuged at 2000 rpm for 1 minute, and the operation of removing the upper phase was repeated twice. The lower chloroform phase was dried under reduced pressure, and the dried solids were suspended with 100 μL of ethanol each, and 45 μL of the suspension was added to the ethanol each and mixed. It was used as a sample to measure the neutral fat content.
[0119] The measurement was performed using Triglyceride E-Test Wako (manufactured by FUJIFILM Wako Pure Chemical Corporation) in accordance with the instructions of FUJIFILM Wako Pure Chemical Corporation. The results are shown in Figure 5 .
[0120] like Figure 5 As shown, in the control group (C group), the neutral fat amount was shown as 130mg / g, which is a clear value of fatty liver. In contrast, in the ARs low intake group (AL group) and the ARs high intake group (AH group), the neutral fat amount was halved to less than 60mg / g. These values are within the range of normal neutral fat amounts. On the other hand, in the intake group (R group) of resveratrol known to have the effect of inhibiting fat accumulation in organs such as the liver, the neutral fat amount in the liver tissue of the hepatitis model mice cannot be corrected. Therefore, it is believed that the effect of the intake of alkylresorcinol is not simply the effect of the result of the improvement of the usual fat metabolism, but the effect peculiar to the symptoms of hepatitis.
[0121] <Test Example 5> ~Pathological evaluation of liver tissue~
[0122] The frozen liver tissue was embedded in paraffin and sliced. The slices were stained with hematoxylin-eosin (HE) using a conventional method. The results of HE staining of the liver tissue of each group are shown in Figure 6 .
[0123] like Figure 6As shown, in the liver tissue of the control group (C group), vacuoles derived from fat droplets were confirmed, and their size and number were relatively large, showing a clear fatty liver image. In contrast, in the ARs low intake group (AL group) and the ARs high intake group (AH group), the number and size of vacuoles were suppressed. On the other hand, almost no effect was observed in the resveratrol intake group (R group). Therefore, the same as the result of Test Example 4, it is believed that the effect of correcting the amount of neutral fat in liver tissue by taking alkylresorcinol is not simply an effect as a result of improving the usual fat metabolism, but an effect specific to hepatitis symptoms.
[0124] <Test Example 6> ~Measurement of PPARγ gene expression level in liver tissue~
[0125] It is known that peroxisome proliferators-activated receptor γ (PPARγ) is a protein belonging to the nuclear receptor superfamily, and plays an important role in lipid metabolism. Pioglitazone (used as a diabetes drug), which is a ligand of PPARγ, is also effective as a non-alcoholic hepatitis therapeutic drug. Accordingly, it is believed that enhancing the function of PPARγ is a useful target for non-alcoholic hepatitis therapeutic drugs. Therefore, in this test example, the expression level of the PPARγ gene in liver tissue was investigated. Specifically, it was measured by quantitative PCR in the manner described below.
[0126] Total RNA from liver tissue was prepared using "NucleoSpin (registered trademark) RNA II Kit (manufactured by Takara Bio Inc.)" according to the instructions of Takara Bio Inc. cDNA synthesis from total RNA was performed using "Protoscript (registered trademark) II First strand cDNA Synthesis Kit (manufactured by NEW ENGLAND BioLabs Ltd)" according to the instructions of NEW ENGLAND BioLabs Ltd.
[0127] Quantitative PCR using the prepared cDNA was performed using "MyGo Mini Real Time PCR Device (manufactured by Funakoshi Co., Ltd.)" and Luna universal LPCR Mix (manufactured by NEW ENGLAND BioLabs Ltd.) according to the instructions of NEW ENGLAND BioLabs Ltd. The following primers were used.
[0128] (For PPARγ)
[0129] Forward: 5'-TGTCGGTTTCAGAAGTGCCTTG-3' (SEQ ID NO: 1)
[0130] Reverse: 5'-TTCAGCTGGTCGATATCACTGGAG-3' (SEQ ID NO: 2)
[0131] (for β-actin)
[0132] Forward: 5'-TGACAGGATGCAGAAGGAGA-3' (SEQ ID NO: 3)
[0133] Reverse: 5'-GCTGGAAGGTGGACAGTGAG-3' (SEQ ID NO: 4)
[0134] The PCR reaction was carried out under the following conditions.
[0135] (95℃ 30 sec → 95℃ 5 sec → 60℃ 30 sec) × 40 cycles → 95℃ 60 sec
[0136] The relative values of the peroxisome proliferator-activated receptor γ (PPARγ) gene expression levels in liver tissue were compared when the control group (group C) was set as 100%. Figure 7 .
[0137] like Figure 7 As shown, the expression of PPARγ gene in liver tissue of the ARs high intake group (AH) increased by about 3.5 times compared with the control group (C group). Therefore, it is suggested that the liver function improvement effect of alkylresorcinol is related to the effect of enhancing the expression of PPARγ gene.
[0138] <Test Example 7> ~Measurement of the expression levels of various genes in liver tissue~
[0139] For various genes of peroxisome proliferators-activated receptor gamma coactivator-1α (PGC-1α), peroxisome proliferators-activated receptor α (Peroxisome Proliferators-Activated Receptor PPARα), and adiponectin, which are known to show important roles in lipid metabolism, the gene expression levels in liver tissue were investigated. The determination was performed by quantitative PCR in the same manner as in Test Example 6. As primers for the determination, the following sequences were used.
[0140] (for PGC-1α)
[0141] Forward: 5'-AAGTGTGGAACTCTCTGGAACTG-3' (SEQ ID NO: 5)
[0142] Reverse: 5'-GGGTTATCTTGGTTGGCTTTATG-3' (SEQ ID NO: 6)
[0143] (For PPARα)
[0144] Forward: 5'-AAGTGCCTGTCTGTCGGGATG-3' (SEQ ID NO: 7)
[0145] Reverse: 5'-CCAGAGATTTGAGGTCTGCAGTTTC-3' (SEQ ID NO: 8)
[0146] (For adiponectin)
[0147] Forward: 5'-GTCAGTGGATCTGACGACACCAA-3' (SEQ ID NO: 9)
[0148] Reverse: 5'-ATGCCTGCCATCCAACCTG-3' (SEQ ID NO: 10)
[0149] (for β-actin)
[0150] Forward: 5'-TGACAGGATGCAGAAGGAGA-3' (SEQ ID NO: 3)
[0151] Reverse: 5'-GCTGGAAGGTGGACAGTGAG-3' (SEQ ID NO: 4)
[0152] The PCR reaction was carried out under the following conditions.
[0153] (95℃ 30 sec → 95℃ 5 sec → 60℃ 30 sec) × 40 cycles → 95℃ 60 sec
[0154] The relative values of each group were compared when the control group (C group) was set as 100% for the gene expression levels in liver tissue. Figure 8A to Figure 8C .
[0155] like Figure 8A to Figure 8C As shown in the figure, the expression levels of PGC-1α, PPARα, and adiponectin genes in liver tissue increased significantly in the ARs intake group. In particular, the gene expression levels of PGC-1α and adiponectin increased further compared to the positive control resveratrol intake group (R group). Fig. 8A , 8C ). Therefore, it is suggested that the liver function-improving effect of alkylresorcinol is related to the effect of enhancing the expression of these genes.
[0156] <Test Example 8> ~Evaluation of the number of mitochondria in liver tissue cells~
[0157] Regarding PGC-1α, whose gene expression level was confirmed to increase in the alkylresorcinol intake group in Test Example 7, it is known that enhanced gene expression leads to an increase in the number of mitochondria, promotion of oxidative phosphorylation, activation of the TCA cycle, promotion of lipid combustion based on β-oxidation, promotion of gluconeogenesis, promotion of ketone body synthesis, etc.
[0158] In this test example, the number of mitochondria in liver tissue cells was investigated. Specifically, the number of mitochondria in liver tissue cells was evaluated by comparing the amount ratio of mitochondrial DNA to genomic DNA measured by quantitative PCR as described below.
[0159] DNA was extracted from liver tissue using "NucleoSpin (registered trademark) Tissue kit (manufactured by Takara Bio Inc.)" in accordance with the instructions of Takara Bio Inc. The number of DNA copies was quantified using "MouseFeeder Cell Quantification Kit (manufactured by Takara Bio Inc.)" in accordance with the instructions of Takara Bio Inc. The following sequences were used as primers.
[0160] (For mitochondrial DNA)
[0161] Forward: 5'-AACCCCGCTCTACCTCACC-3' (SEQ ID NO: 11)
[0162] Reverse: 5'-GTAGCCCATTTCTTCCCATTT-3' (SEQ ID NO: 12)
[0163] (For genomic DNA)
[0164] Forward: 5'-CGCGGTTCTATTTTGTTGGT-3' (SEQ ID NO: 13)
[0165] Reverse: 5'-AGTCGGCATCGTTTATGGTC-3' (SEQ ID NO: 14)
[0166] The PCR reaction was carried out under the following conditions.
[0167] (95℃ 30 sec → 95℃ 5 sec → 60℃ 30 sec) × 40 cycles → 95℃ 60 sec
[0168] The relative values of the measured mitochondrial DNA amount relative to the genomic DNA amount were compared when the control group (C group) was set as 100%. The results are shown in Fig. 9 .
[0169] like Fig. 9 As shown, compared with the control group (C group), no change in the number of intracellular mitochondria was observed in the resveratrol intake group (R group). In contrast, the number of intracellular mitochondria increased in the ARs low intake group (AL group) and the ARs high intake group (AH group). Therefore, it is suggested that the liver function improvement effect brought about by the intake of alkylresorcinol is related to the effect of increasing the number of mitochondria in liver tissue cells.
