Description

Technical Field

[0001] The invention relates to the technical field of green ecological prevention and control, and in particular to a light-hipped eight-toothed beetle repellent, a repellent slow-release bottle and an application thereof. Background Art

[0002] Ips nitidus Eggers belongs to the genus Ips, subfamily Scolytinae, family Curculionidae, order Coleoptera, and is a species of Ips endemic to my country. It is mainly distributed in spruce forests in the Qinghai-Tibet Plateau and surrounding high-altitude areas, causing serious damage to natural spruce (Picea spp.) in the western plateau of my country, especially the spruce species Picea crassifolia Kom. and P. purpurea Mast. endemic to Qinghai. This beetle is a secondary pest that harms natural spruce forests, mainly harming wind-fallen trees, wind-broken trees, and dying trees caused by natural disasters or mechanical damage; however, when conditions are suitable and the insect population density is too high, it will directly attack healthy standing trees, leading to an outbreak. The smooth-hipped eight-toothed beetle often cooperates with many other beetles such as the Shangri-La toothed beetle Ips shangrila Cognato & Sun, the Oriental pseudo-toothed beetle Pseudips orientalis Wood & Yin, the spruce four-eyed beetle Polygraphus poligraphus Linnaeus and the black-striped wood beetle Xyloterus lineatus Olivier to cause damage in spruce forests, causing large areas of spruce forests to die. This type of beetle feeds on the phloem of spruce and mainly gathers and causes damage under the bark, so it is difficult to control its outbreak with conventional chemical control agents.

[0003] The main components of the aggregation pheromone attractants that have been identified and used to lure and kill the smooth-hipped eight-toothed beetle are: 2-methyl-3-butene-2-ol, (±) beetle dienol and (S)-cis-verbenol. Although aggregation pheromones are helpful for monitoring, trapping and prevention of smooth-hipped eight-toothed beetle populations, they can reduce the damage caused to spruce forests to a certain extent. However, when the population breaks out, the high-density population will spread from the damaged spruce forest to the surrounding healthy forests, causing a large number of spruce to be damaged and die. In addition, the management strategy of smooth-hipped eight-toothed beetle pests only from the perspective of large-scale trapping still has some disadvantages. For example, the high cost of purchasing aggregation pheromones, the cost of matching traps, and the labor cost required in the forest. In view of the characteristics of the toothed beetle's aggregation and damage in the forest, finding and developing pollution-free population repellents to break the toothed beetle's aggregation behavior can effectively prevent the large-scale spread and outbreak of the smooth-hipped eight-toothed beetle, which is of great ecological and economic significance for protecting the spruce forest ecosystem.

[0004] The invasion behavior of bark beetles is not only affected by the aggregation pheromones they produce, but plant-derived volatiles are also very important host location signal recognition substances. It is feasible to use plant-derived volatiles as behavioral regulatory substances for increasing the amount of bark beetles attracted or avoiding the aggregation behavior of bark beetles. At present, some plant volatiles have been found to have synergistic or inhibitory effects on the aggregation pheromone components of bark beetles. Such substances have the characteristics of wide sources, many types, environmental friendliness, and low prices. Therefore, focusing on the regulatory effects of volatile components in host and non-host plants on the behavior of bark beetles, and developing behavioral repellents with application value based on this, is an important measure for the comprehensive management of bark beetles.

[0005] The advantages of using repellents to control borer damage are: 1) Using repellents alone to apply them over a large area in the forest can reduce the use of chemical agents, reduce labor costs, break the aggregation behavior of bark beetles, improve the control effect of subcutaneous bark beetles, and ultimately effectively control the occurrence and spread of bark damage in spruce forests. 2) Combining repellents with existing aggregation pheromones and using a "push-pull" control strategy for bark damage management can completely achieve comprehensive control of the key damage areas of the light-hipped eight-toothed bark, thereby achieving the healthy development of the spruce forest ecosystem in high-altitude cold regions. [0025] The present invention provides a kind of light hip eight tooth beetle repellent, including cis-3-hexene-1-ol and / or 1-hexene-3-ol. In the present invention, when the hip eight tooth beetle repellent is preferably cis-3-hexene-1-ol and 1-hexene-3-ol, the mass ratio of the cis-3-hexene-1-ol and 1-hexene-3-ol is preferably 1:1. The present invention does not specifically limit the sources of the cis-3-hexene-1-ol and 1-hexene-3-ol, and commercially available products can be used. In the present invention, the cis-3-hexene-1-ol (Sigma-Aldrich, purity> 98%), 1-hexene-3-ol (Acros Organics, purity 98%).

[0026] The present invention also provides the use of the light-hipped eight-toothed beetle repellent described in the above technical solution in repelling the light-hipped eight-toothed beetle.

[0027] The present invention also provides a light-hipped eight-toothed beetle repellent slow-release bottle, comprising a slow-release bottle, an absorbent cotton placed in the slow-release bottle, and the light-hipped eight-toothed beetle repellent described in the above technical solution adsorbed on the absorbent cotton. In the present invention, the content of the light-hipped eight-toothed beetle repellent in each slow-release bottle is preferably 20mg to 2g. In the present invention, the content of the light-hipped eight-toothed beetle repellent in each slow-release bottle is preferably 200 to 800mg. In the present invention, the content of the light-hipped eight-toothed beetle repellent in each slow-release bottle is preferably 400mg. In the present invention, when the light-hipped eight-toothed beetle repellent is preferably cis-3-hexene-1-ol and 1-hexene-3-ol, the content of cis-3-hexene-1-ol in each slow-release bottle is 200mg, and the content of 1-hexene-3-ol is 200mg. In the present invention, the sustained-release bottle is preferably a low-density polyethylene plastic bottle with a capacity of 16 ml, a thickness of 0.6 mm, a height of 70 mm, a bottom diameter of 20 mm, and a screw-capped bottle cap. In the present invention, the content of the absorbent cotton in each sustained-release bottle is preferably 0.5 g, and the absorbent cotton is preferably medical absorbent cotton. In the present invention, the preparation method of the repellent sustained-release bottle preferably comprises the following steps: placing the absorbent cotton in a sustained-release bottle, then dripping the repellent on the absorbent cotton, sealing the screw-capped bottle, and then placing it in an aluminum foil bag for sealed storage to obtain a repellent sustained-release bottle.

[0028] The present invention also provides the use of the repellent slow-release bottle described in the above technical solution in repelling the light-hipped eight-toothed beetle. In the present invention, the application preferably includes: placing the repellent slow-release bottle on the tree trunk with a spacing of 50 to 100 m. The light-hipped eight-toothed beetle repellent slow-release bottle provided by the present invention is a long-lasting, environmentally friendly repellent, with a simple preparation method, and is particularly suitable for large-scale promotion in production, with broad industrial prospects. The light-hipped eight-toothed beetle repellent slow-release bottle is easy to process and produce, with low cost, and the two components are stably released in the slow-release bottle, and the long-lasting effect can be maintained for 2 to 3 months.

[0029] When the repellent is applied to a large area in the forest to control the light-hipped eight-toothed bark beetle, the repellent slow-release bottle is preferably nailed directly on the spruce trunk at an interval of 80 to 100 meters, in order to prevent the bark beetle from gathering and causing harm in the forest. This application technology is simple to operate, low in cost, and has significant prevention effect.

[0030] For forest stands seriously damaged by borers, it is preferred to nail repellent slow-release bottles to the trunks of spruce trees in the forest at intervals of about 50m, hang aggregation pheromones at the edge of the forest and use cross-plate bark beetle traps for trapping and killing, and use the "push-pull" control technology to control the serious damage of the smooth-hipped eight-toothed bark to the spruce forest. The purpose is to improve the trapping efficiency of aggregation pheromones and improve the control effect. This kind of use technology is cumbersome and costly, but the prevention effect is significant, and it is suitable for the comprehensive management of forest stands seriously damaged by borers.

[0031] In the present invention, the trap is purchased from Beijing Zhongjie Sifang Biotechnology Co., Ltd. The large plate and the small plate are crossed to form a cross shape (50 cm high and 30 cm wide), connected to the upper cover through a hook at the top, and connected to the funnel through a hook at the bottom. The insect collecting bucket is directly connected to the interface at the lower edge of the funnel.

[0032] In the present invention, the attractant core of the aggregation pheromone attractant of the light-hipped eight-toothed beetle preferably includes: 2-methyl-3-butene-2-ol, (±)-beet dienol, (S)-cis-verbenol, an antioxidant and a slow-release bottle. In the present invention, the content of 2-methyl-3-butene-2-ol in each attractant core is preferably 4g; the content of (±)-beet dienol is preferably 40mg; the content of (S)-cis-verbenol is preferably 20mg; the content of the antioxidant is 10mg. In the present invention, the antioxidant is preferably butylated hydroxytoluene (BHT).

[0033] In the present invention, the method for preparing the attractant core of the aggregation pheromone attractant of the light-hipped eight-toothed beetle preferably comprises: dissolving the (±)-beetle dienol in 2-methyl-3-butene-2-ol according to the required mass, and the concentration thereof is preferably 40 mg / 4 g; dissolving the (S)-cis-verbenol in 2-methyl-3-butene-2-ol according to the required mass, and the concentration thereof is preferably 20 mg / 4 g; dissolving the antioxidant in 2-methyl-3-butene-2-ol according to the required mass, and the concentration thereof is preferably 10 mg / 4 g; after uniform mixing, adding to 0.5 g of absorbent cotton in a polyethylene plastic bottle, tightening the bottle cap, and then putting it into an aluminum foil bag, and sealing it to obtain the aggregation pheromone attractant core. When used in the field, take out the polyethylene plastic bottle and hang it directly in the central part of the cross plate of the trap.

[0034] In the present invention, in order to comprehensively evaluate the repellent effect of the repellent components of the light-hipped eight-toothed beetle, the repellent slow-release bottle prepared by cis-3-hexene-1-ol and / or 1-hexene-3-ol is used simultaneously with the light-hipped eight-toothed beetle aggregation pheromone attractant lure core. The purpose of using the light-hipped eight-toothed beetle aggregation pheromone attractant lure core alone is for comparison. When in use, the repellent slow-release bottle and the attractant slow-release bottle lure core are simultaneously hung in the middle position of the cross plate of the trap. The trap is fixed with 2 bamboo poles and iron wire, and is set at the edge of the spruce forest or in an open, sunny, ventilated area in the forest, with a spacing of about 50m, and the insect collecting bucket is about 80cm above the ground. The spacing between the traps in the block group is preferably 200m. The deterrent effect of the slow-release bottle was evaluated by the number of trapped Oxyconoptera in the insect collecting bucket of the trap.

[0035] The technical solution provided by the present invention is described in detail below in conjunction with the embodiments, but it is not intended to limit the protection scope of the present invention.

[0036] Special note: In the preliminary experiment, it has been confirmed that 2-methyl-3-butene-2-ol, (±)-dienol, (S)-cis-verbenol, S-(–)-β-pinene, (–)-limonene, 1-octen-3-ol, cis-3-hexen-1-ol and 1-hexen-3-ol alone in the forest cannot attract the light-hipped eight-toothed beetle. The data is zero, so the corresponding chart is not provided in the text.

[0037] Example 1

[0038] The preparation of the slow-release bottle of the scaly beetle in this embodiment uses commercially available cis-3-hexen-1-ol, whose chemical purity is greater than 98%. 200 mg of cis-3-hexen-1-ol is added to 0.5 g of absorbent cotton in a low-density polyethylene plastic bottle, the bottle cap is tightened, and the bottle is placed in an aluminum foil bag and sealed to obtain a slow-release bottle.

[0039] Preparation of the aggregation pheromone attractant core of the light-hipped eight-toothed beetle: 40 mg (±)-beetle dienol, 20 mg (S)-cis-verbenol and 10 mg antioxidant (butylated hydroxytoluene) are directly added to 4 g 2-methyl-3-butene-2-ol, and mixed evenly to obtain the aggregation pheromone attractant. The attractant is added to 0.5 g of absorbent cotton in a low-density polyethylene plastic bottle, the bottle cap is tightened, and then placed in an aluminum foil bag and sealed to obtain the aggregation pheromone attractant core of the light-hipped eight-toothed beetle. The prepared attractant core is sealed with aluminum foil and refrigerated at 4°C.

[0049] Comparative Example 3

[0050] Only the aggregation pheromone attractant core of the light-hipped eight-toothed beetle was prepared, and the attractant composition, preparation method and dosage were the same as those in Example 1. The results are shown in Figure 1 .

[0051] Comparative Example 4

[0052] No ingredients are added, only low-density polyethylene sustained-release bottle + 0.5g of absorbent cotton, the results are shown in Figure 1 .

[0053] The slow-release bottles and aggregation pheromone attractant cores prepared by Examples 1 to 2 and Comparative Examples 1 to 3 for avoiding the light-hipped eight-toothed beetle were hung on the traps in the forest at the same time. The trap with the blank core (Comparative Example 4) was used as a blank control to evaluate the biological activity of the four plant-derived volatiles against the light-hipped eight-toothed beetle. The specific steps are as follows:

[0054] a) Production of the trap: The trap is mainly a cross-plate type bark beetle trap purchased from Beijing Zhongjie Sifang Biotechnology Co., Ltd. The large plate and the small plate are crossed to form a cross shape (50 cm high and 30 cm wide), connected to the upper cover through the hook on the top and connected to the funnel through the hook on the bottom. A small insect collecting bucket is connected to the bell mouth at the lower edge of the funnel.

[0055] b) Suspension of the repellent slow-release bottle and / or the attractant core: One repellent slow-release bottle and one attractant core were suspended simultaneously in the middle of the cross-plate bark beetle trap 20 cm from the top. The trap with only one attractant core was used in Comparative Example 3.

[0056] c) Field setting of traps: Each experimental treatment was set up with 3 replicates. The traps were set up at the edge of the spruce forest, and the distance between the traps was about 50m. Two 2m long and 5cm diameter bamboo poles were inserted into the soil layer to fix them, with a distance of 30cm between the two bamboo poles. Then the trap was fixed between the two bamboo poles with wire, and the insect collection bucket of the trap was about 80cm above the ground. The distance between the traps between replicates was about 200m.

[0057] d) Data survey and trap emptying: According to the above specific usage steps, the traps were correctly laid out as required at the beginning of the first flight period of the adult beetles of the light-hipped eight-toothed bark. In this experiment, each treatment was repeated 3 times, and the number of bark beetles trapped in each treatment was the average of the three traps. Depending on the weather conditions, the number of bark beetles in the insect collection bucket was observed every 2 to 3 days. After counting, the trapped bark beetles were placed in a plastic box and brought to the laboratory.

[0058] from Figure 1The analysis of the insect attractant effect in the results shows that the aggregation pheromone attractant of the light-hipped eight-toothed beetle (Comparative Example 3) has a good trapping effect. The blank control (Comparative Example 4) has no attractant activity to the light-hipped eight-toothed beetle. When the S-(–)-β-pinene slow-release bottle preparation (Comparative Example 1) or the (–)-limonene slow-release bottle preparation (Comparative Example 2) is used together with the light-hipped eight-toothed beetle aggregation pheromone attractant core, although the number of light-hipped eight-toothed beetles trapped in the trap is slightly increased (Comparative Example 1) or decreased (Comparative Example 2) compared with Comparative Example 3, there is no significant synergistic effect or inhibitory effect compared with Comparative Example 3. There is no statistically significant difference in the number of light-hipped eight-toothed beetles trapped by the three treatment cores. When the cis-3-hexene-1-ol slow-release bottle preparation or the 1-hexene-3-ol slow-release bottle preparation is used together with the light-hipped eight-toothed beetle aggregation pheromone attractant lure core, the trapping efficiency of the light-hipped eight-toothed beetle is significantly reduced, and the trapping amount of the light-hipped eight-toothed beetle aggregation pheromone attractant lure core can be reduced by 5.6 times (Example 2) to 6.6 times (Example 1), proving that cis-3-hexene-1-ol and 1-hexene-3-ol have a strong inhibitory effect on the light-hipped eight-toothed beetle aggregation pheromone attractant lure core (Examples 1 and 2), and both ingredients have a strong repellent effect on the light-hipped eight-toothed beetle. The inhibition rate of cis-3-hexene-1-ol reached 84.8%, and the inhibition rate of 1-hexene-3-ol reached 82.2%, and there was no statistically significant difference in the inhibitory effect of the two. The experimental conclusion shows that either of the two components, cis-3-hexen-1-ol and 1-hexen-3-ol, can be used as an effective repellent for the smooth-hipped eight-toothed beetle. Their application in the forest can reduce the aggregation behavior of the smooth-hipped eight-toothed beetle, and can comprehensively prevent and control the damage of the smooth-hipped eight-toothed beetle, thereby better protecting the spruce forest.

The sample purity, the preparation method of the repellent sustained-release bottle, the storage of the repellent sustained-release bottle, and the implementation steps are the same as those in Example 1.

[0061] Preparation of the attractant core of the aggregation pheromone attractant of the light-hipped eight-toothed beetle: the attractant ingredients are 2-methyl-3-butene-2-ol, (±)-beetle dienol and (S)-cis-verbenol, and the preparation method and dosage are the same as those in Example 1.

[0066] Example 5

[0067] The repellent component in the slow-release bottle of the light-hipped eight-toothed beetle repellent is 400 mg of cis-3-hexen-1-ol; the rest is the same as in Example 1, and the results are shown in Figure 2 .

[0069] Example 6

[0070] The repellent component in the slow-release bottle of the light-hipped eight-toothed beetle repellent is 800 mg of cis-3-hexen-1-ol; the rest is the same as in Example 1, and the results are shown in Figure 2 .

[0071] Preparation of the attractant core of the aggregation pheromone attractant of the smooth-hipped eight-toothed beetle: the attractant ingredients, preparation method and dosage are the same as those in Example 1.

[0072] Example 7

[0073] The repellent component in the slow-release bottle of the light-hipped eight-toothed beetle repellent is 2g of cis-3-hexen-1-ol; the rest is the same as in Example 1, and the results are shown in Figure 2 .

[0074] Preparation of the attractant core of the aggregation pheromone attractant of the smooth-hipped eight-toothed beetle: the attractant ingredients, preparation method and dosage are the same as those in Example 1.

[0075] Comparative Example 5

[0076] Only the aggregation pheromone attractant core of the light-hipped eight-toothed beetle was prepared, and the preparation method and dosage were the same as those in Example 1. The results are shown in Figure 2 .

[0077] According to the specific implementation steps in Example 1, the forest avoidance effect test of Examples 3 to 7 of the light-hipped eight-toothed beetle and Comparative Example 5 was carried out from May 17 to June 16, 2023. The specific results are shown in Figure 2 In this experiment, each treatment was repeated three times, and the number of bark beetles trapped in each treatment was the average number of the three traps. Depending on the weather conditions, the number of bark beetles in the insect collection bucket was observed every 2 to 3 days.

[0078] from Figure 2The analysis of the insect attractant effect shows that the aggregation pheromone attractant of the light-hipped eight-toothed beetle (Comparative Example 5) has a good trapping effect. When the cis-3-hexene-1-ol sustained-release bottle preparation is used together with the light-hipped eight-toothed beetle aggregation pheromone attractant core, it shows a significant inhibitory effect at 5 test doses of 20mg, 200mg, 400mg, 800mg and 2g. When the dosage of cis-3-hexene-1-ol in each repellent slow-release bottle is 20mg, the inhibition rate reaches 66.3% compared with Comparative Example 5 (Example 3). When the dosage of cis-3-hexene-1-ol in each repellent slow-release bottle is between 200mg and 2g, the inhibition rate exceeds 83.9%, and the inhibitory effect difference between the four treatments is not significant, and is significantly lower than the trapping amount at 20mg (Examples 4 to 7). [0096] Only the aggregation pheromone attractant core of the light-hipped eight-toothed beetle was prepared, and the preparation method and dosage were the same as those in Example 1. The results are shown in Figure 3 .

[0097] from Figure 3 The analysis of the insect attractant effect shows that the aggregation pheromone attractant of the light-hipped eight-toothed beetle has a good trapping effect, with an average total trapping amount of 112 heads (Comparative Example 6). When the 1-hexene-3-ol sustained-release bottle release preparation is used together with the light-hipped eight-toothed beetle aggregation pheromone attractant core, it shows a significant inhibitory effect at 5 test doses of 20mg, 200mg, 400mg, 800mg and 2g, and the inhibition rate reaches more than 60.7% (Examples 8 to 12). When the dosage of 1-hexene-3-ol in each repellent sustained-release bottle is 200mg, the inhibition rate reaches 80.1% (Example 9). When the dosage is greater than 200mg, compared with Example 9, the difference in the inhibitory effect is not significant, and it is significantly less than the trapping amount when the dosage is 20mg. The experimental results show that 1-hexen-3-ol can be used as a repellent for the light-hipped eight-toothed beetle in the forest alone or in combination with aggregation pheromones to comprehensively prevent and control the damage of the light-hipped eight-toothed beetle. Considering the attractant core's attractant effect in the forest, its effective period, and its economic use, 200 mg of 1-hexen-3-ol was selected as the optimal repellent dose in practical applications.

[0098] Example 13

[0099] The repellent ingredients in the repellent sustained-release bottle of the light-hipped eight-toothed beetle are cis-3-hexen-1-ol and 1-hexen-3-ol, and the dosage is 200 mg. The sample purity, the preparation method of the repellent sustained-release bottle, the storage of the repellent sustained-release bottle, and the implementation steps are the same as those in Example 1.

[0100] Preparation of the attractant core of the aggregation pheromone attractant of the light-hipped eight-toothed beetle: the attractant ingredients are 2-methyl-3-butene-2-ol, (±)-beetle dienol and (S)-cis-verbenol, and the preparation method and dosage are the same as those in Example 1.

[0101] The use of the repellent slow-release bottle and the aggregation pheromone attractant core is the same as in Example 1, and the rest is the same as in Example 1. The results are shown in Figure 4 .

[0102] Comparative Example 7

[0103] Only the aggregation pheromone attractant core of the light-hipped eight-toothed beetle was prepared, and the preparation method and dosage were the same as those in Example 1. The results are shown in Figure 4 .

[0104] According to the specific implementation steps in Example 1, traps were set up in the forest from May 17 to June 16, 2024, and the aggregation pheromone attractant core and repellent slow-release bottle were hung to test the repellent effect of the light-hipped eight-toothed beetle. Comparative Example 7 was used as a control, and the specific results are shown in Figure 4 In this experiment, each treatment was repeated three times, and the number of bark beetles trapped in each treatment was the average number of the three traps. Depending on the weather conditions, the number of bark beetles in the insect collection bucket was observed every 2 to 3 days. After counting, the trapped bark beetles were placed in a plastic box and brought to the laboratory.

[0105] from Figure 4 The analysis of insect trapping data shows that the aggregation pheromone attractant of the light-hipped eight-toothed beetle has a good trapping effect. When used alone, the average total number of traps is 78 (Comparative Example 7). When the repellent slow-release bottle and the aggregation pheromone attractant core are used at the same time, the average total number of traps of the light-hipped eight-toothed beetle is only 4.33, and the inhibition rate is 94.4% (Example 13), indicating that cis-3-hexene-1-ol and 1-hexene-3-ol can be used as repellents for the light-hipped eight-toothed beetle in the forest at the same time to prevent and control the light-hipped eight-toothed beetle; or the two can be used in combination with the aggregation pheromone attractant core to comprehensively prevent and control the damage of the light-hipped eight-toothed beetle.
