The method comprises the following steps: collecting rana nigromaculata from a natural water body; setting treatment groups and control groups, and performing natural matching treatment on rana nigromaculata; putting all rana nigromaculata in a glass aquarium in a dark room to adapt for 5-15 minutes; recording songs of normal female and male rana nigromaculata in the mating season in advance; playing normal songs of rana nigromaculata, observing response of rana nigromaculata of different treatment groups, and recording corresponding songs and data; and acquiring song parameters, and analyzing experimental data.
