Problems solved by technology

Due to the strong subjectivity of these identification methods, it is difficult to grasp the specific standards, and there is often a lot of uncertainty—the observation results of different people are often different, which often causes great controversy

Embodiment 1

[0021] (1) Variation information: It is located on the mitochondrial COI gene of Sniper anchovy and Brachygnathus chinensis, the specific location is at 236 bp of the mitochondrial COI gene, and the base variation information is T>C.

[0022] (2) Discovery of mutation sites

[0023] The shortjawed anchovy collected from Dongting Lake and the Yangtze anchovy collected from the Nanjing section of the Yangtze River were taken as the research objects (the sampling amount was more than 100), and the samples were identified and determined by referring to the methods and ecological types described in "Fauna of China". Primers were designed according to the complete mitochondrial sequence (NC026912.1) of Anchovy brevis to amplify the COI gene,

Embodiment 2

[0040] The sample size was increased to 100 silver anchovy and 100 short-jawed anchovy, and the detection was carried out according to the method in Example 1. The result showed that the accuracy rate was 100%, and the mark was still reliable.

[0041] This method overcomes the shortcomings of identifying the Anchovy chinensis and Anchovy shortjaw only by empirical observation, and provides a rapid and reliable molecular biological identification technology for the accurate identification of Anchovy cylindrophyllus and Anchovy shortjaw.

Abstract

The invention discloses a molecular marker for identifying coilia nasus and C. brachygnathus, and application of the molecular marker. The molecular marker is positioned on a COI gene of a mitochondrion of the coilianasus or the C. brachygnathus, and is specifically positioned at the part of 236bp of the COI gene of the mitochondrion; base variation information shows that T is more than C; the variation site of the coilianasus is T, and the variation site of the C. brachygnathus is C. A method for identifying the coilianasus and the C. brachygnathus can be used to identify the coilianasus and the C. brachygnathus by detecting the molecular marker. The method overcomes the disadvantages of only using experiential observation to identify the coilianasus and the C. brachygnathus, and provides a rapid and reliable molecular biological identification technology for the accurate identification of the coilianasus and the C. brachygnathus.

Description

technical field [0001] The invention belongs to the technical field of molecular biology detection, and relates to a molecular marker for distinguishing anchovy and anchovy shortjaw and an application thereof. Background technique [0002] Coilianasus, commonly known as saury, is an important economic fish that people in the lower reaches of the Yangtze River like to eat. Sword anchovy is a migratory fish that usually lives in the sea. It enters the river from the sea every February to March, and goes upstream for reproductive migration. It is mainly distributed in the main stream of the middle and lower reaches of the Yangtze River. C. brachygnathus is a species established by German Kreyenberg et al. (1908) based on specimens collected from Dongting Lake. It grows and reproduces in rivers and lakes in the middle and lower reaches of the Yangtze River, and belongs to freshwater fish. Shortjaw anchovy is closely related to sword anchovy, but because it does not undergo repr...
