[0023] 1.1 Drug target proteins and MRSA-related genes
[0024] Find the chemical constituents of dandelion, Polygonum cuspidatum, Beibaijiang, and Scutellaria barbatae in RYN from "Dictionary of Traditional Chinese Medicine (Second Edition)"; , Metabolism, Excretion) properties to determine the druggable active ingredients; retrieve the human target proteins corresponding to these active ingredients from the PubChem database. 103, 256, 45, and 97 target proteins of Dandelion, Polygonum cuspidatum, Beibaijiang, and Scutellaria barbata were found. Retrieve the target proteins of Van and FOX in ChEMBL, ETCM and PubChem databases, and exclude non-empirical acquisitions and duplicates, and select 134 Van target proteins and 65 FOX target proteins; search for MRSA pathogenicity in Ensembl Bacteria, Uniprot and PubMed databases A total of 80 genes were selected by removing duplicates.

experiment example 2
[0055] Objective: To explore the synergistic effect of combined action of RYN and LNZ on MRSA.
[0056] METHODS: The minimum inhibitory concentrations (MICs) of RYN and LNZ against free MRSA strains were determined by broth microdilution method. The antibacterial effect of RYN and LNZ was studied by chessboard method. Detection of MRSA in biofilms by 2,3-bis-(2-methoxy-4-nitro-5-sulfophenyl)-2H-tetrazole-5-carboxanilide (XTT) reduction method. The anti-biofilm effects of the two drugs were observed in the development and formation of biofilms. Viable cells stained with Syto9 in biofilms after drug administration were visualized using confocal scanning microscopy.
[0057] Results: The MRSA strain was sensitive to RYN, the MIC value was 1 / 2, while the MIC value of LNZ was 4 μg / ml. RYN significantly enhanced the activity of the tested antibiotics by reducing their MICs. The antibacterial effect of LNZ was more significant than that of RYN at 0h, and the antibacterial effect ...
