Tissue culture process of arrowroot

Abstract

The invention discloses an arrowroot tissue culture process which comprises the following steps: treating an explant, disinfecting, carrying out inoculated culture, cutting off a newly grown terminal bud when the terminal bud of the explant germinates and grows, removing the top of the terminal bud, inoculating the terminal bud into a starting induction culture medium to carry out adventitious bud induction, and removing useless parts of a block mass obtained in the previous step, so as to obtain the arrowroot. The method comprises the following steps of: inoculating the tissue culture seedlings obtained in the previous step into a proliferation and propagation culture medium for proliferation and propagation culture, removing useless parts of the tissue culture seedlings obtained in the previous step, inoculating the tissue culture seedlings into a hormone-reducing culture medium for hormone-reducing culture, then inoculating the tissue culture seedlings into a transition culture medium for transition culture, directly screening and planting the seedlings which reach the seedling standard, and performing rooting culture on the seedlings which do not reach the seedling standard. The method has the advantages that the proliferation rate can be greatly increased, the yellow leaf rate is reduced to 27.7%, the rooting rate is increased to 95.2%, and the seedling rate is increased to 91.6%. The productivity of the arrowroot is greatly improved, and great economic benefits are brought.
