Description

[0001] CROSS-REFERENCE TO RELATED APPLICATIONS

[0002] This application claims the benefit of U.S. Provisional Application Serial No. 63 / 355,072, filed on June 23, 2022, under 35 U.S.C. §119(e), the contents of which are hereby incorporated by reference in their entirety. Technical Field

[0003] The present application relates to a synthesis method of a small molecule regulator of eukaryotic initiation factor 2B which can be used as a therapeutic agent, a novel intermediate and a synthesis method thereof. Summary of the invention

[0004] The multi-subunit protein complexes eukaryotic initiation factor 2B (eIF2B) and eukaryotic initiation factor 2 (eIF2) are required for the initiation and regulation of protein synthesis in eukaryotic cells. The interaction of eIF2B with eIF2 plays an important role in the integrated stress response (ISR) pathway. Activation of this pathway leads in part to ATF4 (activating transcription factor 4) expression and stress granule formation. Abnormal ISR activation is found in a variety of neurodegenerative diseases, which has a strong functional connection with the pathology characterized by RNA binding / stress granule protein TAR DNA binding protein (TARDBP) (also known as TDP43). Impairment of eIF2B activity is associated with activation of the ISR pathway involved in a variety of neurodegenerative diseases, including Parkinson's disease, amyotrophic lateral sclerosis (ALS), Alzheimer's disease, and frontotemporal dementia and vanishing white matter disease. On the other hand, activation of eIF2B inhibits the formation of ISR and ISR-dependent stress granules, and has been found to be neuroprotective in these disease models. Small molecule modulators of eIF2B are expected to treat these conditions.

[0005] Disclosed herein are improved methods for synthesizing small molecule modulators of eukaryotic initiation factor 2B. In certain embodiments, provided herein are improved methods for preparing compounds of formula I-1:

In some embodiments, the method comprises contacting a compound of formula X, or a stereoisomer, or a mixture of stereoisomers, or a salt thereof, under deprotection conditions sufficient to provide a compound of formula XI, or a stereoisomer, or a mixture of stereoisomers, or a salt thereof.

[0431] In some embodiments, the deprotection conditions described for the preparation of the compound of formula IX include contacting the compound of formula X with an acid. In some embodiments, the acid is generated in situ from an acid precursor in the presence of a protic compound. In some embodiments, the acid precursor is acetyl chloride. In some embodiments, the protic compound is an alcohol. In some embodiments, the generation of the acid includes generation at a temperature of about -30°C to about 100°C. In some embodiments, the temperature is about -30°C to about 60°C. In some embodiments, the generation of the acid includes lowering the temperature to about 0°C to about 5°C and warming the reaction to a temperature of about 15°C to about 25°C. In some embodiments, the temperature of the reaction mixture is raised to about 30°C to about 60°C, such as about 40°C to about 45°C, and stirred for a duration of at least about 2 hours. In some embodiments, the latter reaction step at a higher temperature (such as at about 40°C to about 45°C) provides significantly improved yields and / or reduced impurity content compared to other methods (e.g., maintaining the temperature below about 30°C).

[0432] In some embodiments, the deprotection conditions include generating hydrogen chloride in situ. In some embodiments, the deprotection conditions also include neutralizing the residual acid generated immediately after the reaction is completed and separating the compound of formula X or its salt. Other deprotection conditions, such as contacting the intermediate X-1a with aqueous hydrochloric acid to remove the BOC group, usually lead to the decomposition of the oxadiazole ring, thereby resulting in a low yield of the intermediate XI. In contrast, by implementing the deprotection and neutralization conditions described herein, ring decomposition is avoided, and the yield and / or purity of the compound of formula XI can be significantly improved. Therefore, the deprotection conditions described herein enable large-scale industrial production that was previously unimaginable.

[0433] In some embodiments, the contact of a compound of formula X or its stereoisomer or mixture of stereoisomers or its respective salt under deprotection conditions provides a free base form of a compound of formula XI or its stereoisomer or mixture of stereoisomers. In some embodiments, the contact of a compound of formula X or its stereoisomer or mixture of stereoisomers or its respective salt under deprotection conditions provides a salt of a compound of formula XI or its stereoisomer or mixture of stereoisomers. In some embodiments, the salt is hydrochloride, oxalate and 4-chlorophenoxyacetic acid salt. In some embodiments, each equivalent of hydrochloride includes two equivalents of chloride ions. In some embodiments, each equivalent of oxalate includes two equivalents of oxalate anions. In some embodiments, each equivalent of 4-chlorophenoxyacetic acid salt includes one equivalent of 4-chlorophenoxyacetic acid anions.

[0826] The selected solvent (ethyl acetate (EtOAc) or toluene) is added to a vial containing about 22 mg of the compound of Formula VIa-1a and an equimolar amount of the selected base to prepare a slurry, as described in Table 2. The slurry is stirred at room temperature (RT) for 2 to 3 days. If a solid precipitates out, the solid is recovered by centrifugation and then vacuum dried at RT for XRPD analysis. The sample without precipitation after the solution-mediated reaction is cooled in a -15°C refrigerator for 1 day.
