[0039] (5) Assembly of the kit

[0040] 4. Clinical application of ELISA kit

[0041]The ELISA kit prepared by the inventor was used to detect scoliosis patients to be diagnosed and compared with the actual clinical detection to determine the effectiveness of the ELISA kit. Specifically, it includes measuring the expression levels of LACC1 and FHL2 proteins in the blood samples of the subjects, and comparing them with the expression levels of LACC1 and FHL2 proteins in normal blood samples, so that clinicians can quickly and accurately grasp the patient's disease state and severity, and take timely and more effective measures. Individualized prevention and treatment plan provides support.

[0042] The full-length nucleotide sequences or fragments of the LACC1 and FHL2 genes of the present invention can usually be obtained by PCR amplification, recombination or artificial synthesis. Once the relevant sequences are obtained,...

Embodiment 1

[0046] Example 1 High-throughput sequencing screening of differentially expressed genes

[0047] 1. Sampling

[0048] From October 2012 to December 2015, patients with scoliosis who were treated in the Department of Orthopedics of Peking Union Medical College Hospital were selected. A total of 25 cases were collected in the case group. All patients had typical clinical manifestations and were confirmed by X-ray examination. Controls were collected from patients with other diseases who were hospitalized in orthopedics at the same period, and a total of 15 cases were collected. The patients were all teenagers aged 10-18. Blood samples of all subjects were collected and numbered and stored in a -80°C freezer. All clinical samples in this study were informed to the patients and approved by the Hospital Ethics Committee.

[0050] use LS (Invitrogen: 10296-010) carried out RNA extraction on the collected blood samples, and t...

Embodiment 2

[0061] Example 2 qRT-PCR verification of the expression of LACC1 and FHL2 genes in patients with scoliosis

[0062] 1. Materials

[0063] A total of 10 patients with scoliosis who were treated in the Department of Orthopedics of Peking Union Medical College Hospital between October 2012 and December 2015 were selected. All patients had typical clinical manifestations and were confirmed by X-ray examination. Controls came from patients with other diseases who were hospitalized in orthopedics at the same period, and a total of 8 cases were collected. The patients were all teenagers aged 10-18. Blood samples of all subjects were collected and numbered and stored in a -80°C freezer. All clinical samples in this study were informed to the patients and approved by the Hospital Ethics Committee.

[0064] 2. Method

[0065] 2.1 Total RNA was extracted from the blood sample, which was the same as the extraction method in Example 1.

[0066] 2.2 Synthesis of cDNA by reverse transcr...

Abstract

The invention discloses an application of LACC1 (laccase domain containing 1) and FHL2 (four and a halfLIM domains 2) genes in preparation of a scoliosis detection product, also discloses a biological reagent for detecting scoliosis and further discloses an ELISA reagent for detecting scoliosis. By detecting scoliosis with the LACC1 and FHL2 genes, early diagnosis can be rapidly and effectively performed, and a treatment target and important evidences are provided for predictive diagnosis of occurrence and development of AIS (adolescent idiopathic scoliosis) and even treatment of AIS on the biological level in the future.

Description

technical field [0001] The invention relates to the field of biomedicine, in particular to the application of LACC1 and FHL2 genes in the preparation of scoliosis detection products. Background technique [0002] Scoliosis, also known as scoliosis, is a complex three-dimensional spinal deformity characterized by lateral curvature of the spine accompanied by vertebral body rotation. According to the cause of the disease, it can be divided into 3 categories: congenital scoliosis, syndromic scoliosis and idiopathic scoliosis. Congenital scoliosis is caused by abnormalities in the development of the spine; syndromic scoliosis is caused by neuromuscular diseases, bone diseases, connective tissue diseases, and neurofibromatosis; the exact cause of idiopathic scoliosis is not yet known. Clearly, relevant researches believe that genetic factors, growth and development and hormones, connective tissue lesions, abnormalities of the muscular system, abnormalities of the central nervous...
