[0121] Example 2 Screening of compounds

[0122] The present invention first takes primary liver cells from 6-8 week old C57B6 mice, and after plating at an appropriate density, uses a 0.3 μM mixture of palmitic acid and oleic acid to induce and stimulate the aggregation of lipid droplets in the cells. After the lipid droplets are aggregated, fresh culture medium mixed with small molecule compounds is added to the cells respectively, and the culture medium is removed after culturing for 36 hours. The cells are rinsed with PBS, and after fixation with a fixative, the cells are incubated with a Nile red solution for 15 minutes, then the Nile red solution is removed, and the cells are rinsed with PBS three times and then incubated with a DAPI solution for 5 minutes, and then the solution is removed, and the cells are rinsed three times with PBS, leaving part of the PBS on the cell surface to cover the cells. The cell well plate is placed in a high content instrument, the appropriate parameters are adjusted, and the instrument is waited for to automatically image and take pictures of the wells continuously. Figure 2 After the high-content instrument takes the photo, the parameters are set. After the instrument automatically analyzes the fluorescence parameters, the multi-substituted 3,3-dicarboxylate-indole-2-thione compounds and their derivatives (as shown in Table 1 below) as shown in the above formula (1) with better Nile red fluorescence attenuation effect are screened out compared with the control group. Among them, the small molecule compounds suitable for the present invention include but are not limited to TOIDC shown in formula (1a) and the compound shown in formula (1k).

[0130] Example 3 In vitro cell experiment

[0131] In the in vitro cell experiment of the present invention, a mixture of palmitic acid and oleic acid was used to stimulate the accumulation of lipid droplets in liver cells, and then the cells were treated with increasing gradient concentrations of the small molecule drug TOIDC screened in Example 2 of the present invention. Nile red staining was used to find that the accumulated lipid droplets in the cells were gradually reduced. Figure 3 .according to Figure 2 Image J software was used to calculate the fluorescence parameters of Nile red staining after cells were treated with different concentrations of TOIDC. The calculation showed that the half inhibition rate (IC50) of TOIDC in reducing lipid droplet accumulation was 14.17 μM. Figure 4 B. The present invention also used a triglyceride detection kit to find that the level of triglyceride accumulated in primary liver cells treated with TOIDC decreased as the concentration increased. Figure 5 .

[0132] Then, the present invention repeated the experiment in vitro using another cell line HepG2 cell line, and found that the small molecule compound TOIDC also reduced the fluorescence intensity in the HepG2 cell line in the Nile red staining experiment. The left picture shows only DMSO, the middle picture shows DMSO and OA / PA, and the right picture shows TOIDC and OA / PA. After calculation using Image J software, it was found that TOIDC had a significant effect in inhibiting the accumulation of cellular lipid droplets (see Figure 6 ), and the small molecule compound TOIDC also significantly reduced the triglyceride content in the HepG2 cell line in the triglyceride quantification experiment. The left column is only DMSO, the middle column is DMSO and OA / PA, and the rightmost column is TOIDC and OA / PA (see Figure 7 ).

[0133] In the qPCR primary liver cell experiment, the present invention verifies that the small molecule compound TOIDC reduces the gene transcription of SREBP1 and reduces the transcription level of the downstream key enzymes of lipid synthesis. In each group of bar graphs, the left side is DMSO, the middle is OA / PA+DMSO, and the right side is OA / PA+TOIDC (see Figure 8 ).

[0134] In the WB liver primary cell experiment, the present invention verifies that the small molecule compound TOIDC reduces the protein level of SREBP1 and downstream key enzymes of lipid synthesis and inhibits the lipid synthesis pathway in the liver to relieve fatty liver (see Fig. 9 ).

[0135] In the qPCR primary liver cell experiment of the present invention, after TOIDC was administered, the levels of SREBP1 and downstream lipid synthesis pathway genes decreased. After adding the SREBP1-activating compound T0901317, the levels of SREBP1 and downstream synthesis pathway genes recovered, proving that the small molecule compound TOIDC inhibits the lipid synthesis pathway in the liver and relieves fatty liver by reducing the protein levels of SREBP1 and downstream key enzymes of lipid synthesis (see Fig.10 ).

[0136] In the WB liver primary cell experiment of the present invention, after administration of TOIDC, the levels of SREBP1 and downstream lipid synthesis pathway protein FASN decreased. After adding drugs that activate SREBP1, the levels of SREBP1 and downstream synthesis pathway protein FASN recovered, proving that the small molecule compound TOIDC inhibits the lipid synthesis pathway in the liver and relieves fatty liver by reducing the protein levels of SREBP1 and downstream key enzymes of lipid synthesis (see Fig.11 ).

[0137] In the Nile red staining experiment of primary liver cells of the present invention, the fluorescence of Nile red weakened after TOIDC was administered, and the fluorescence of Nile red increased after the addition of T0901317, a compound that activates SREBP1 transcription, which proves that the small molecule compound TOIDC inhibits the lipid synthesis pathway in the liver and relieves fatty liver by reducing the protein levels of SREBP1 and downstream key enzymes of lipid synthesis (see Fig.12 ).

[0138] Similarly, in another cell line qPCR HepG2 cell experiment, it was verified that the small molecule compound TOIDC reduced the gene transcription of SREBP1 and reduced the transcription level of downstream key enzymes of lipid synthesis. In each group of bar graphs, the left side is DMSO, the middle is OA / PA+DMSO, and the right side is OA / PA+TOIDC (see Fig.13 In the WB HepG2 cell experiment, it was verified that the small molecule compound TOIDC reduced the protein levels of SREBP1 and downstream key enzymes of lipid synthesis, thereby inhibiting the lipid synthesis pathway in the liver and alleviating fatty liver (see Fig.14In the qPCR HepG2 cell experiment, the levels of SREBP1 and downstream lipid synthesis pathway genes decreased after TOIDC was administered. After adding the SREBP1-activating compound T0901317, the levels of SREBP1 and downstream synthesis pathway genes recovered, proving that the small molecule compound TOIDC inhibits the lipid synthesis pathway in the liver and relieves fatty liver by reducing the protein levels of SREBP1 and downstream key enzymes of lipid synthesis (see Fig.15 ). In the WB HepG2 cell experiment, the levels of SREBP1 and the downstream lipid synthesis pathway protein FASN decreased after TOIDC was administered. After adding drugs that activate SREBP1, the levels of SREBP1 and the downstream synthesis pathway protein FASN recovered, proving that the small molecule compound TOIDC inhibits the lipid synthesis pathway in the liver and relieves fatty liver by reducing the protein levels of SREBP1 and downstream key enzymes of lipid synthesis (see Fig.16 ).

[0139] The above experimental data show that the small molecule drug TOIDC screened in Example 2 of the present invention can reduce the accumulation of fat droplets in the liver, reduce the content of triglycerides in the liver, and treat non-alcoholic fatty liver disease. The present invention further illustrates through in vitro cell experiments that the small molecule compound screened in Example 2 of the present invention has the potential to treat non-alcoholic fatty liver disease.

[0140] Example 4 In vivo animal experiment

[0141] In the in vivo animal experiment of the present invention, after determining that obese mice fed with high-fat for 16 weeks had fatty liver, they were randomly divided into two groups. One group of mice was intraperitoneally injected with 10 mg / kg of the small molecule TOIDC solution selected in Example 2 of the present invention every day (administration group), and the other group was intraperitoneally injected with the same concentration of DMSO solvent every day (control group). The mice were killed after 3 weeks of the experiment. The gross anatomical diagram shows that the liver of the mice in the administration group is bright red (such as Fig.17 A), the liver of mice in the control group was whiter and larger (as shown on the right side of Fig.17 The results showed that the drug group could alleviate fatty liver (as shown on the left side of A). Fig.17 Then the present invention tested the lipid accumulation in the liver by TG kit and found that the triglyceride level in the liver of the drug administration group decreased (as shown in Fig.18 At the same time, after staining the tissue sections, it was found that the fatty liver of the mice in the drug-treated group was significantly alleviated (as shown in Fig.19 At the same time, the weight of the two groups of mice was measured within three weeks of the experiment. It was found that the average weight of the mice in the drug group was 36.0g, which was significantly lower than the average weight of the mice in the control group (40.2g). Fig. 20 At the same time, the mice were killed three weeks after the experiment. It can be seen that the mice in the drug-treated group were significantly smaller than those in the control group (as shown in Fig.21A), the fat tissue was taken and the fat tissue volume of mice in the drug administration group ( Fig.21 B right) was significantly smaller than that of the control group mice ( Fig.21 B shown on the left).

[0142] The present invention also found that TOIDC inhibited the lipid synthesis pathway in the liver (such as Fig. 22 , Fig.23 shown).

[0143] Next, in the cytotoxicity experiment, the present invention treated primary liver cells with different concentrations of TOIDC and found that there was no difference in cell activity when the concentration was less than 60 μM, indicating that the small molecule TOIDC did not affect cell viability at the working concentration (such as Fig.24 shown).

[0144] In the animal experiment of the present invention, the average serum ALT aminotransferase levels of high-fat mice three weeks after intraperitoneal injection of TOIDC and high-fat mice three weeks after injection of solvent DMSO were 172.3U / L and 179.1U / L, respectively, and the average serum aminotransferase AST levels were 33.8U / L and 32.7U / L, respectively, indicating that intraperitoneal injection of TOIDC has no liver damage toxicity to mice (such as Fig.25 shown).

[0145] The above experimental data show that the small molecule drug TOIDC screened in Example 2 of the present invention can inhibit the lipid synthesis pathway in the liver, reduce the level of triglycerides in the liver, alleviate and relieve fatty liver, and the small molecule drug TOIDC is non-toxic.

[0146] Example 5 Insulin tolerance test

[0147] Five mice in the administration group (10 mg / kg of the small molecule TOIDC screened in Example 2 of the present invention was intraperitoneally injected daily) and five mice in the control group (the same amount of DMSO solvent was intraperitoneally injected daily) were taken respectively. After 3 weeks, the two groups of mice were first starved for 4 hours on an empty stomach. After 4 hours, the tail vein blood of the two groups of mice was taken to test the blood sugar of the mice after starvation with a blood glucose test strip. Then, the two groups of mice were intraperitoneally injected with insulin at a dose of 1.75 IU / kg. The tail vein blood of the two groups of mice was taken to test the blood sugar with a blood glucose test strip and a blood glucose meter at 15 minutes, 30 minutes, 1 hour, 1.5 hours, and 2 hours after the injection of insulin. Finally, it was found that there was no significant difference in blood sugar between the two groups 15 minutes, 1 hour, and 1.5 hours after the injection of insulin. However, 2 hours after insulin injection, the average blood glucose of mice in the TOIDC group (the drug group) was 4.80 mM, and the average blood glucose of mice in the control group was 7.32 mM. There was a significant difference in blood glucose between the two groups of mice, indicating that the blood glucose recovery of mice in the drug group was slower than that of the control group 2 hours after intraperitoneal injection of insulin. In the presence of insulin resistance, intraperitoneal injection of TOIDC can improve insulin sensitivity and improve insulin resistance (such as Fig.26 shown).

[0148] The present invention further illustrates through in vivo cell experiments that the small molecule compounds screened in Example 2 of the present invention have the potential to treat non-alcoholic fatty liver disease and / or type 2 diabetes.
