[0089] Example
[0090] The lipid used in the following examples (Examples 1 to 17) is a lipopeptide, specifically surfactin, and the preparation process is as follows:
[0091] Pick a single colony of Bacillus subtilis (for the construction method of a single colony of Bacillus subtilis, see CN201810865295.7), inoculate it into LB liquid culture medium, and culture it at 37°C and 200rpm for 16 hours to obtain a seed solution, which is inoculated into a fermentation medium shake flask at a ratio of 5%. After culturing at 37°C and 200rpm for 2 to 6 hours, 1mM IPTG is added, and the culture is continued for 2 days to obtain an initial fermentation liquid containing surfactin.
[0092] The composition of the initial fermentation medium used was: 30-100 g / L of sugars, 10-50 g / L of inorganic nitrogen source, 0.5-3 g / L of organic nitrogen source, KH 2 PO 4 0.1~1g / L, Na 2 HPO 4 12H 2 O 0.5~0.3g / L, CaCl 2 0.002~0.01g / L, MnSO 4 ·H 2 O0.002~0.01g / L, FeSO 4 7H 2O 0.002~0.01g / L, pH 6.5~7.5, fermentation additives (silicon defoamer, polysaccharide, amino acid or its derivatives, oligopeptide, phospholipid, glycolipid, and / or fatty acid or its derivatives) 0.01~20wt%.
[0093] Adjust the pH of the initial fermentation broth to between 6.0 and 6.5, let it stand overnight at 4°C to allow the substances in the solution to completely precipitate or accelerate its sedimentation by slight stirring, and finally centrifuge to obtain the product, which is the fermentation broth. The method disclosed in CN105400784A was used to detect the content of surfactant in the fermentation broth. Specifically: take 1mL of fermentation broth and centrifuge it at 12000rpm for 1 minute, take 100μL of supernatant and add 1900μL of deionized water, mix well, and analyze it by HPLC after filtering through a 0.22μm filter membrane. The mobile phase of HPLC analysis is methanol and water in a ratio of 85 / 15, the flow rate is 1mL / minute, the chromatographic column is a C18-ODS reverse phase chromatographic column, the column temperature is 40°C, the ultraviolet detector, and the detection wavelength is 205nm.
[0094] The test showed that the content of surfactin in the fermentation broth was 30%. The pH of the fermentation broth was adjusted to acidic, and the surfactin formed a precipitate. After solid-liquid separation, the solid part was taken and dried to obtain a surfactin-containing product. The powder product was obtained by cold purification and freeze drying, and the content of surfactin was measured to be 95wt%-99.99wt%.
[0095] The inorganic compound fertilizer was purchased from Stanley general compound fertilizer (inorganic nitrogen, phosphorus and potassium compound fertilizer (15-15-15)); the organic fertilizer livestock and poultry manure was purchased from natural pure chicken manure (Hebei Rundong Fertilizer Co., Ltd.), the crop straw was purchased from Su Rui Agricultural Products Deep Processing Store, the compost fertilizer was purchased from Hebei Jingwo Fertilizer Co., Ltd., and the biological fertilizer was a purchased bacterial fertilizer containing EM bacteria of Grass subtilis (Hebei Rundong Fertilizer Co., Ltd.).

[0341]
[0342]
[0343] Note: Different lowercase letters in the same row indicate significant differences among treatments (P<0.05).
[0344] As shown in Table 5, the plant height, bulk density, 1000-grain weight and yield of corn were all increased after applying lipid-added fertilizers, among which the plant height increased by 3.26%-8.71%, and the plant height of treatment 8 was significantly increased by 8.71% compared with treatment 2; the bulk density increased by 0.60%-2.41%, and the bulk density of treatment 7 was significantly increased by 2.41% compared with treatment 2; the 1000-grain weight increased by 0.74%-2.04%, and the 1000-grain weight of treatment 9 was significantly increased by 2.04% compared with treatment 2; the yield increased by 3.50%-6.28%, and the yield of treatment 8 was significantly increased by 6.28% compared with treatment 2. This shows that the application of lipid-added fertilizers can promote the growth of corn and increase corn yield; at the same time, there is no significant difference between treatment 6 and treatment 2, which shows that the application of lipid-added fertilizers can reduce the amount of fertilizer, but does not affect the growth and yield of crops. It has also been further confirmed that the growth-promoting and yield-increasing effects of lipids are applicable to crops such as corn, rice, and cucumbers, and that the role of lipids in improving fertilizer utilization is also applicable to organic fertilizers.

[0356] The measurement results are shown in Table 6.
[0357] Table 6: Stem diameter, chlorophyll content and yield of strawberries under different treatments
[0358]
[0359]
[0360] Note: Different lowercase letters in the same row indicate significant differences among treatments (P<0.05).
[0361] As shown in Table 6, the stem diameter, chlorophyll content and yield of strawberries increased after applying lipid-added fertilizers, among which the stem diameter increased by 3.44%-26.78%, and the stem diameter of treatment 15 increased significantly by 26.78% compared with treatment 2; the chlorophyll content increased by 17.17%-23.23%, and the chlorophyll content of treatment 14 increased significantly by 25.18% compared with treatment 2; the yield increased by 4.14%-11.45%, and the yield of treatment 5 increased significantly by 7.59% compared with treatment 2. This shows that the application of lipid-added fertilizers can promote the growth of strawberries and increase the yield of strawberries, and also shows that fertilizers with different lipid components have a certain effect of promoting growth and increasing yield on strawberries.
[0362] Soil samples from treatment 1, treatment 2, treatment 5, treatment 11 and treatment 16 were collected according to the five-point sampling method with a sampling depth of 5-20 cm. The soil physical and chemical properties of the retrieved soil samples were tested, including soil nitrogen, phosphorus, potassium, organic matter, pH, and electrical conductivity.
[0363] The test results are shown in Table 7.

[0365]
[0366]
[0367] Note: Different lowercase letters in the same row indicate significant differences among treatments (P<0.05).
[0368] As shown in Table 7, the total nitrogen, available phosphorus and available potassium content of the soil will be significantly reduced when applying fertilizers with lipids compared with applying fertilizers alone. This may be because lipids have a certain promoting effect on the absorption and utilization of fertilizers by plants. Adding lipids can help plants better absorb and transport nutrients, which is beneficial to improving fertilizer utilization. At the same time, adding lipids significantly increases the organic matter content of the soil, which is beneficial to improving soil quality. Moreover, adding lipids to fertilizers can improve the problem of soil pH decrease caused by fertilizer application to a certain extent. In short, the application of fertilizers with lipids can not only promote plant growth and increase crop yields, but also improve soil quality and fertilizer utilization.
