Embodiment Construction

[0065] The following embodiments apply the present invention to a method for detecting target cells from a biological sample containing target cells and non-target cells. In the following embodiments, the biological sample is blood. The biological sample is not limited to blood, and if it is a body fluid sample, for example, urine, bone marrow fluid, or the like may be used. Target cells are rare cells that circulate in the blood. In the following embodiments, the target cells are circulating tumor cells (CTCs), and the non-target cells are red blood cells, white blood cells and platelets. The target cells are not limited to CTCs, and may be cancer cells other than CTCs, endothelial cells (Circulating Endothelial Cell: CEC), hematopoietic stem cells (Hematopoietic Stem Cell: HSC), and the like. In addition, the target cells may also be abnormal lymphocytes. When the target cells are abnormal lymphocytes, the non-target cells are other white blood cells.

Abstract

A cell detection method and a cell detection system capable of performing target cell detection satisfactorily without interference and reducing the time required for detection of target cells are provided. The cell detection method includes a separation step of step S1 to obtain a sample containing target cells by removing at least a part of the non-target cells from a biological sample includingtarget cells and non-target cells based on a difference in forces acting on target cells and non-target cells; a concentration step of step S3 of obtaining a measurement sample having a target cell concentration increased from the sample containing target cells; a detection of step S4 of detecting target cells by subjecting the measurement sample to an imaging flow cytometer.

Description

technical field [0001] The invention relates to a cell detection method and a cell detection system. Background technique [0002] Circulating Tumor Cells (CTCs), endothelial cells, hematopoietic stem cells in bone marrow, and fetal erythroblasts in the blood of pregnant women exist in body fluids, and their association with various diseases is known. In particular, CTCs leaked from tumors into blood have been reported to be useful as cancer diagnostic factors, and they are expected to be used as non-invasive cancer tests. However, the presence rate of CTCs in blood is several to dozens per 10 mL, and it is very difficult to measure rare cells in body fluids. [0003] The CellSearch system (manufactured by Veridex) is known as a method for measuring CTCs which are one of the rare cells (Non-Patent Document 1). Non-Patent Document 1 discloses a method of recovering and measuring CTCs using magnetic particles immobilized with an anti-EpCAM antibody, targeting a cell adhesio...
