Problems solved by technology

Therefore, the mutation of NF-κB gene is considered to be related to the occurrence and development of MPN, but the mechanism of abnormally high expression of NF-κB in MPN patients is not yet clear
[0005] rs28362491 is an insertion / deletion polymorphic site on the NF-κB gene, and there is no research report on the relationship between rs28362491 and MPN

Embodiment 1

[0028] In this study, 269 patients clinically diagnosed as MPN in Qilu Hospital of Shandong University and 291 normal controls were included, and the distribution of rs28362491 insertion / deletion polymorphism on NF-κB gene in blood samples was detected.

[0029] As shown in Table 1, in the co-dominant model, the ins / ins genotype was positively correlated with the onset of MPN (p=0.033, OR=1.713, 95%CI=1.044-2.810), while the ins genotype was associated with the onset of MPN There was a positive correlation (p=0.040, OR=1.113, 95% CI=1.005-1.232). The results indicated that the carrier type of the rs28362491 locus on the NF-κB gene of individuals was related to their susceptibility to MPN. The ins / ins genotype and ins gene distribution frequency are significantly different between individuals with and without myeloproliferative neoplasms, and individual myeloproliferative neoplasms can be predicted by detecting the carrier type of the rs28362491 locus on the individual NF-κB ge...

Embodiment 2

[0038]At the same time, bone marrow samples from 64 MPN patients and 9 normal controls were collected for mRNA expression detection and protein expression detection.

[0039] The result is as Figure 1-4 As shown, the mRNA expression level of NF-κB in bone marrow mononuclear cells of MPN patients was significantly higher than that of normal controls, and the mRNA expression level of NF-κB in different MPN types was significantly higher than that of normal controls ( figure 1 ). JAK2 V617F Mutation is the most common gene mutation in MPN, and it is one of the criteria for clinical diagnosis of MPN. We found that the mRNA expression level of NF-κB was significantly lower than that of JAK2 V617F Mutation-positive patients were significantly higher than mutation-negative patient samples ( figure 2 ). Different genotypes of NF-κBrs28362491 insertion / deletion polymorphism affect the expression of NF-κB. The expression levels of NF-κB mRNA and protein in bone marrow mononuclea...

Abstract

The invention belongs to the technical field of biomedicine, and particularly relates to application of polymorphism sites to preparation of products for detecting myeloproliterative neoplasms (MPN).According to the invention, through 269 patients clinically diagnosed as MPN in Qilu Hospital of Shandong University and 291 normal controls, statistical analysis is performed on the distribution situation of No. rs28362491 insertion / deletion polymorphism sites on an NF-kappa B gene in blood specimens, and data shows that the gene carrying type of the rs28364491 sites on the NF-kappa B gene is related to the susceptibility of the sites to MPN, thus providing a theoretical foundation for the occurrence mechanism of the myeloproliterative neoplasms, and providing a research foundation for development of a myeloproliterative neoplasms early warning model suitable for the population of China.

Description

technical field [0001] The invention belongs to the technical field of biomedicine, and in particular relates to the application of a polymorphic site in the preparation of myeloproliferative tumor detection products. Background technique [0002] The information disclosed in this background section is only intended to increase the understanding of the general background of the present invention, and is not necessarily taken as an acknowledgment or any form of suggestion that the information constitutes the prior art already known to those skilled in the art. [0003] Myeloproliferative neoplasms (MPNs) are a group of clonal hematopoietic stem cell disorders characterized by the proliferation of one or more lineages of myeloid (granulocytic, erythroid, megakaryotic, or hypertrophic) cells. Classic BCR / ABL1-negative MPNs include polycythemia vera (PV), essential thrombocythemia (ET) and myelofibrosis (MF). The clinical manifestations are one or more types of blood cell incre...
