[0027] The present invention finds that autonomously replicating acentric extrachromosomal structures called double minichromosomes (DMs) often mediate oncogene amplification in human tumors. According to the present inventors, removal of DMs from the nucleus by micronucleus formation initiated by budding of the nuclear envelope in S phase promotes cell maturation, differentiation and death.

Abstract

The present invention provides methods for screening test substances by which double minutes (DM) or extrachromosomal DNA that inhibit, enhance or eliminate micronucleation formation in cells are screened. The present invention also provides a method for inducing maturation or death of cells capable of producing micronuclei. The invention also provides methods of treating diseased subjects with cells associated with the disease having DM and extra chromosomal DNA and capable of producing micronuclei to capture them. Further provided is a method of detecting chromosomal and extra chromosomal DNA in a cell.
