The method comprises the following steps of: firstly, determining the layout positions of drill holes according to a field investigation condition, analyzing and determining the connectivity between each drill hole and a reservoir water level through water level change correlation, and preliminarily determining a permeation channel between the drill holes through a tracing test; and further measuring the flow rates corresponding to different elevation points in each drill hole by adopting a single-hole dilution underground water flow rate method, drawing a flow rate-elevation curve, finally selecting two drill holes with permeation channels determined by a tracing test, comparing the flow rate-elevation curves of the two drill holes, and finally obtaining the specific leakage part of the dam and the leakage strength of the dam.
