The visual observation culture medium comprises, by mass percentage, 45-60% of gypsum, 5-10% of activated carbon and the balance of distilled water. The preparation method includes uniformly mixing the gypsum with the activated carbon to pour into the distilled water, uniformly stirring to obtain mixture; pouring the mixture into a twelve-hole cell culture plate 0.1-0.2 centimeter in thickness; and standing on a horizontal platform till solidifying and obtaining the visual observation culture medium for the nematode acute toxicity test. The visual observation culture medium is easy to count, obvious in dead nematode judgment, easy for behavior observation and higher in test result correctness and accuracy, and can be applied to the nematode toxicity test.
