Problems solved by technology

The production of three-dimensional bacterial nets by Arthropodium oligosporum is affected by many factors, and the effect of producing three-dimensional bacterial nets is unstable. The small number of traps often affects its biocontrol effect
[0004] The method provided by the present invention to increase the number of prey organs of Arthrobotrys oligospora (Arthrobotrys oligospora) has not been found to be published in the same literature as the content of the present invention through literature search

Embodiment Construction

[0018] The present invention will be described in further detail below in conjunction with the accompanying drawings.

[0019] The medium used in this example is a conventional medium.

[0020] The implementation steps of the method for improving oligospore Arthrobotrys oligospora (Arthrobotrys oligospora) predator organ quantity of the present invention are as follows:

[0021] 1. Preparation of nematodes for experiments

[0022] Inoculate Caenorhabditis elegans on sterilized oat water medium, and culture at 28°C for 7 days or 8 days. Filter the nematodes with the Behmann funnel method, centrifuge the collected nematodes at 2000 rpm for 2 min, discard the supernatant, resuspend with sterile deionized water, and repeat three times. Finally, dilute the nematodes to 15 / ul.

[0023] 2. Preparation of spores

[0024] Put 50ml of CMA solid medium into a 250ml Erlenmeyer flask, sterilize at 121°C for 20 minutes, inoculate Arthrobusia oligospora blocks after the medium is cooled,...

Abstract

The invention relates to a method for increasing the amount of trapping organs of arthrobotrys oligospora serving as a nematode-trapping fungus, belonging to the field of applied microbiology. The final concentration of a spore suspension of the arthrobotrys oligospora is up to 20mM through adding sodium nitrate into the spore suspension of the arthrobotrys oligospora, so that the amount of the trapping organs of the arthrobotrys oligospora is increased, and the nematode killing capacity of the trapping organs of the arthrobotrys oligospora is improved. The method has the beneficial effects that a great number of trapping organs are generated by the arthrobotrys oligospora after NaNO3 is added in the spore suspension of the arthrobotrys oligospora, so that the nematode killing capacity is remarkably improved, and the death rate of nematodes is up to 85.88% when the nematodes are added for 12 hours; and when the nematodes are added for 24 hours, the amount of the trapping organs generated on a flat plate on which sodium nitrate is added is more than six times as much as the amount of the trapping organs generated on a flat plate on which no sodium nitrate is added, and the death rate of the nematodes is up to 99.80%.

Description

Technical field: [0001] The invention relates to a method for increasing the number of prey organs of Arthrobotrys oligospora, which belongs to the field of applied microbiology. Background technique: [0002] Crop pathogenic nematodes are a serious hazard, and the direct economic losses caused by crop pathogenic nematodes are as high as 157 billion U.S. dollars every year worldwide. In my country, diseases caused by pathogenic nematodes have become the second largest type of diseases in agricultural production after fungal diseases, seriously threatening the safety of grain and economic crops in our country. For a long time, the control of nematodes has mainly relied on chemical pesticides, but due to the high toxicity and high residue of nematode chemical pesticides, it seriously affects the safety of agricultural products. In recent years, research on nematode microorganisms has provided people with another way—biological control. Nematode microorganisms refer to a grou...
