The bonded-phase chromatography-tandem mass spectrometry is characterized by comprising the following steps: drying, smashing and sieving smoking cessation drug samples; after the samples are soaked by sodium hydroxide solution, extracting the solution with methyl tertiary butyl ether and a methanol solution; after matrix solid-phase dispersion extraction and purification are performed on extract liquor, making an analysis with two chiral columns by a tandem bonded-phase chromatography-tandem mass spectrometry; quantitatively detecting a proportion of S-(-)-nicotine and R-(+)-nicotine in the smoking cessation drug by adopting a peak area normalization method.
