Embodiment 1

[0051] Example 1 - Analysis of Cell Activation

[0052] Activated leukocyte compositions prepared according to preferred embodiments of the present invention are quantified by analysis of various cell surface markers. Increased aggregation of platelets with monocytes or granulocytes as indicated by P-selectin expression is a marker of monocyte and granulocyte activation. CD62L is a plasma membrane protein that disappears during activation and thus decreases with cell activation. CD42b is a platelet activation marker involved in the coagulation process as a coagulation factor. It interacts with the extracellular matrix as well as adhesion molecules and is also used in the present invention as an indicator of monocyte and granulocyte activation.

[0053] Cells were sampled at 3 time points: fresh buffy coat (Fr. BC), incubated buffy coat (IBC) and final activated leukocyte composition (FP) (Table 1).

Embodiment 2

[0060] Example 2 - Analysis of Activated Leukocyte Compositions

[0061] Tables 2 and 3 describe the cellular composition of the final ALC, as determined by analysis using the Cell Dyn Analyzer. After resuspending the activated leukocytes in serum, the cells were analyzed. Live cells were stained using the trypan blue exclusion test and observed under a microscope.

[0062] Table 2: Composition of Activated Leukocyte Compositions

[0063]

[0064] Table 3: Leukocyte composition in ALC

[0065]

[0066] As shown in Table 2, Cell Dyn results show that ALC contains 46.8+ / -39.2 (103 / μl) platelets, 0.1+ / -.03 (106 / μl) red blood cells and 6.8+ / -3.8 (103 / μl) ) of white blood cells. Based on Cell Dyn analysis, the white blood cell composition of ALC (as shown in Table 3) was also determined to contain 52%-78% neutrophils, 1-2% basophils, 1-9% eosinophils Granulocytes and 6%-12% monocytes and 13%-24% lymphocytes (taking standard deviation into account). The ranges described ...

[0068] Comparison between the method of the invention and the prior art process

[0069] To emphasize the several unexpected advantages associated with the present invention, embodiments thereof are compared to the method disclosed in US Patent No. 6,146,890, namely to Danon ("Danon").

Abstract

Disclosed are a therapeutic blood-derived activated leukocyte composition, a method for preparing the same, and a method for using the composition to repair or promote wound healing.

Description

[0001] Cross References to Related Applications [0002] This application claims the benefit of Application Nos. 61 / 209,298, filed March 5, 2009, entitled "Activated Leukocyte Compositions," and filed April 1, 2009, entitled "Activated Leukocyte Compositions for Vascular Insufficiency" 61 / 211,587, the disclosure of which is hereby incorporated by reference. Background technique [0003] The wound healing process involves the involvement of white blood cells (also known as leukocytes). White blood cells include lymphocytes, granulocytes and monocytes. The three common types of lymphocytes are T cells, B cells, and natural killer cells. T cells and B cells play an important role in antigen recognition in vivo (Parkin, 2001). Natural killer (NK) cells recognize and kill infected cells through changes in major histocompatibility complex (MHC) levels (Moretta, 2008). The involvement of lymphocytes in the healing process is largely related to their production of cytokines and gr...
