Embodiment 1

[0053] The invention will be further understood by reference to the following non-limiting examples. Example 1: Regulating estrus and ovulation experiment design of gilts with estradiol microspheres

[0054] A controlled study in 180-day-old gilts was performed to demonstrate that a single dose injection of the controlled-release microsphere formulation must be able to induce false pregnancy, followed by PG 600 TM Induces estrous ovulation in youth. Four doses of estradiol microspheres were evaluated in this study. Studies have also demonstrated that Lutalyse TM (PGF2α, 10 mg) could induce controlled estrus in pseudopregnant gilts at day 59 after treatment in a controlled manner to induce luteal breakdown and estrus.

[0055] The study was designed as a parallel group scheme. The same number of gilts were randomly divided into the following treatment groups: placebo group; polylactide microspheres group containing 12.5 mg 17β-estradiol; 25 mg 17β-estradiol group; 50 mg 17β...

Embodiment 2

[0082] Controlled-release polylactide microspheres designed to release physiologically effective levels of 17β-estradiol over a period of 16 to 26 days were able to successfully induce pseudopregnancy in gilts for more than 50 days. Following induction of pseudopregnancy, pseudopregnancy can be terminated and estrus induced by administering a commercially available analogue of PGF2[alpha]. The advantage of inducing pseudopregnancy followed by estrus induction is that the reproductive pattern of a large number of gilts can be controlled. Example 2: Overview of biodegradable drug delivery systems for estradiol in sucrose acetate isobutyrate formulations

[0083] The aim of this study was to prepare sucrose acetate isobutyrate (SAIB)-based formulations of estradiol with in vivo pharmacokinetic release profiles similar to those obtained using biodegradable microsphere formulations. The delivery system uses SAIB as a high viscosity matrix compound to provide controlled release of ...

Abstract

Controlled release compositions and methods for inducing pseudopregnancy in gilts and sows are disclosed. In the preferred embodiment, the formulation includes polylactide microspheres releasing estradiol 17 beta at physiologically useful levels over a period of time between five and thirty days. Following induction of pseudopregnancy, the pseudopregnancy can be terminated and estrus induced by administration of a compound such as PGF2 alpha . The advantages of inducing pseudopregnancy followed by induction of estrus are that the breeding patterns of large numbers of gilts and sows can be controlled.

Description

Background of the invention [0001] The invention relates to the field of animal feeding industry, in particular to a controlled-release preparation for regulating estrus and ovulation of gilts. [0002] In fact, gilts (female piglets who have never had piglets) typically reach puberty between 150 and 250 days of age. Once in puberty estrus, they have a regular cycle of ovulation and sexual excitement called "estrus" or "estrus." Most gilts seem to ovulate and conceive during estrus. [0003] Normally, gilts and sows do not come into estrus (estrous) in sync and therefore do not all conceive at the same time. Piglets produced by gilts tend to be smaller and lighter compared to sows. [0004] The main goal of commercial swine products is to maximize reproductive efficiency, especially in gilts. Increased reproductive rates offer producers great opportunities to reduce production costs and increase profitability. Attempts are being made to increase early reproductive rates i...
