Abstract

The method includes: sample genome DNA extraction, single copy gene primer design, real-time quantitative PCR standard preparation and standard curve establishing of nuclear genome and mitochondrial genome single copy gene, detection of mitochondrial genome copy number in a sample and content calculation, wherein a sample DNA extraction method includes: adding sepiella maindroni tissue into polybutylene succinate powder and alkyl ether ammonium sulfate, grinding together,and performing extraction, organic solvent separation and precipitation to obtain sample DNA.
