Embodiment 1

[0067] Example 1. Serum MMP-8 and CRP concentrations associated with CVD and AMI in subjects.

[0068] Concentration levels of serum MMP-8 and CRP concentrations were measured in subjects without CVD at baseline as described above, and 1-year follow-up was used to determine the association of these concentrations with the likelihood of having CVD or AMI. The results showed that the sum of serum CRP and MMP-8 concentrations was higher in people who experienced AMI or any CVD event compared to those who did not experience AMI or any CVD event—both above the corresponding threshold / mean values. This combined difference between these groups was significant (Tables 1 and 2). As shown in Table 1, the mean concentration sum was 1.97 in patients with CVD compared to 1.55 in patients without CVD (p<0.001). The mean concentration of MMP-8 alone was not significant, while the concentration of CRP was also significant when considering CVD alone (p<0.001). In Table 2, it is shown that th...

Embodiment 2

[0079] Example 2. Serum MMP-8 and CRP Concentrations Associated with Inflammatory Bowel Disease (IBD) and Cancer

[0080] Inflammatory bowel disease (IBD) is a group of inflammatory disorders of the colon and small intestine. Crohn's disease and ulcerative colitis are the main types of inflammatory bowel disease. It was early shown that IBD is significantly associated with elevated CRP, as this protein can have differential effects in affected patients (Henriksen et al., 2008). As an inflammatory marker, CRP helps to predict, monitor and evaluate the presence, severity and treatment of IBD.

[0081] Danish researchers, for example, have shown that people with high blood CRP levels have a 30% increased risk of any cancer later in life compared with people with low CRP levels, and are associated with a risk of developing lung cancer and possibly colorectal cancer (Cancer. Net, ASCO's patient website). The researchers also found that among people with cancer, those with high p...

Embodiment 3

[0094] Example 3. Correlation data on MMP-8 concentration determined by IFMA or ELISA.

[0095]Knowing the differences between the different antibodies and methods known in the art for the detection of MMP-8 and the activating part of MMP-8, the inventors wanted to investigate whether the relevant data obtained depended on the measurement method. MMP-8 concentrations were measured by IFMA and ELISA using different MMP-8 antibodies as previously described. Measurements were performed on patients and control subjects (343 patients with acute coronary syndrome (ACS) (Pussinen et al., 2013). The control group was matched for age ± 2 years, sex and parish. Inclusion criteria were: None History of definite or suspected coronary artery disease or stroke, nor history of surgery or chemotherapy within the previous 4 weeks. They had no positive history of angina, that is, chest pain in any position associated with exercise and relieved by rest. None of them were taking any medications ...

Abstract

The present invention relates to a novel method for determining risk of cardiovascular diseases comprising detecting of MMP-8 and CRP in a sample, and comparing the detected amounts with respective predetermined values of MMP-8 and CRP, wherein the detection of elevated levels of MMP-8 and CRP is indicative of presence or risk of cardiovascular event or disease. The present invention relates alsoto the use of detection of MMP-8 and CRP for predicting a risk for getting a cardiovascular event, for monitoring the effect of therapy on cardiovascular event or on cardiovascular disease, or for detecting the presence of a subclinical cardiovascular disease. Also, a method for constructing a risk prediction model for a presence of CVD disease or a risk of CVD events is presented.

Description

technical field [0001] The present invention describes methods for improving the prediction and estimation of prognosis of cardiovascular disease. The method is based on the identification and subsequent combination of biomarkers that are particularly suitable for differentiating subjects at risk of cardiovascular disease events from healthy subjects. The biomarkers identified herein can also be used to detect subclinical cardiovascular disease and to monitor the effects of treatments or drugs on cardiovascular disease. The present invention encompasses the use of matrix metalloproteinase-8 (MMP-8) and C-reactive protein (CRP) for the prediction and estimation of prognosis of cardiovascular disease events, and for monitoring the effects of treatments and drugs on cardiovascular disease events . In addition, MMP-8 and CRP concentration measurements can be used to detect subclinical cardiovascular disease. Background technique [0002] Cardiovascular disease (CVD) is a grou...
