Problems solved by technology

[0003] At present, the most commonly used method to establish gastric cancer cell lines is to directly culture patient tissue blocks (or ascites). Due to the limitation of surgical resection specimen size, tumor cells and normal cells exist alternately, and it is difficult to obtain a large number of tumor cells. The growth of cells in vitro requires strict culture conditions, and most tumor cells cannot be stably and continuously passaged in vitro

Abstract

The invention discloses a gastric cancer cell line with high transfer potential and its establishment method and application. After the gastric cancer cell line is transplanted into nude mice from clinical tumor specimens of gastric cancer, the tumor tissue is cultured in vitro to establish the line. After the tumor cell line was subcutaneously transplanted into nude mice, liver and lung metastases occurred. The karyotype of the tumor cells was hyperdiploid, which was different from the diploid normal cells, and the chromosomal modal number was concentrated between 50-57. The cell population doubling time was 48 hours. Cell migration experiments showed that the tumor cells had enhanced cell migration ability with time. Cell invasion experiments showed that the tumor cells had obvious invasiveness. Therefore, the gastric cancer cell line established in the present invention provides, for example, a good experimental tool for the treatment of gastric cancer and the study of metastasis mechanism.

Description

technical field [0001] The invention belongs to the field of medicine and biotechnology, and relates to a gastric cancer cell line with high metastasis potential and a method for its establishment and application. Background technique [0002] Gastric cancer (GC) is one of the most common malignancies, and despite improvements through surgery and chemotherapy, the prognosis of advanced gastric cancer is still poor, with a 5-year survival rate approaching 20%. Invasion and metastasis are the main causes of death of the disease and greatly affect the therapeutic effect. Since the occurrence of gastric cancer metastasis is closely related to many tumor-related genes, different patients should have their own characteristics. Therefore, it is urgent to establish an individualized metastasis model that is closer to the clinical biological and behavioral characteristics, to identify the key genes of gastric cancer metastasis and to understand the molecular mechanism of its prognos...
