Problems solved by technology

Although the delivery of this type of radiation has been improved, the fact that it cannot be precisely controlled in many areas of the body has produced certain uncertain biological side effects, including destruction of healthy tissue, radiation burns and vomiting, and other similar injuries such as Destruction of tissue and cellular architecture, structural changes within the cytoskeletal organization and disruption of the structural organization of actin, and effects on various degenerations of blood, blood vessels, microvasculature, healthy tissues and organs secondary to radiation therapy , immune and other damage
In particular, the efficacy of radiation therapy in cancer patients and other patients is currently limited by significant damage to surrounding healthy tissue, including increased inflammatory responses and the release of toxic mediators, including inflammatory cells Factors, chemofactors, eicosanoids, and metabolites that limit the dose of ionizing radiation effects in patients

Embodiment Construction

[0008] According to one embodiment, the present invention generally relates to the use of peptides, thymosin beta 4 (thymosin beta 4 or Tβ4), or Tβ4 fragments such as LKKTET, conservative variants thereof (sometimes, these are also referred to as LKKTET peptides or polypeptides, also including such as N-terminal or C-terminal variants of KLKKTET and LKKTETQ) to treat, prevent or reverse the physical, cognitive, and biological damage caused by ionizing radiation exposure.

[0009] More than 50% of all cancer patients receive radiation therapy to reduce tumor size. Damage to surrounding tissues due to toxic side effects of radiation and damage to normal tissue structures, as well as direct effects of X-rays or gamma-rays, and side effects due to release of toxic substances from tumor tissue and cell debris Inflammatory, degenerative and immunological effects make the effect of radiation therapy dose-limiting. Since up to 10% of the protein in tumors is actin, and 50% of this pr...

Abstract

A method of treatment or prevention of damage due to ionizing radiation exposure involves administering to a subject in need of such treatment an effective amount of a composition containing 1) a compound including a radiation damage-inhibiting polypeptide containing amino acid sequence LKKTET (such as Thymosin beta4), a conservative variant of LKKTET, an actin-sequestering agent, an anti-inflammatory agent; 2) an agent which stimulates production of the compound in the subject; 3) an agent which regulates the compound in the subject; or 4) an antagonist of the compound, so as to inhibit radiation damage in the subject.

Description

[0001] This application claims the benefit of US Provisional Application Serial No. 60 / 488,097, filed July 18,2003. technical field [0002] The present invention relates to the field of treatment or prevention of radiation-induced damage. Background technique [0003] Ionizing radiation has been frequently used as a form of treatment for many types of cancer and tissue abnormalities for decades. Although the delivery of this type of radiation has been improved, the fact that it cannot be precisely controlled in many areas of the body has produced certain uncertain biological side effects, including destruction of healthy tissue, radiation burns and vomiting, and other similar injuries such as Destruction of tissue and cellular architecture, structural changes within the cytoskeletal organization and disruption of the structural organization of actin, and effects on various degenerations of blood, blood vessels, microvasculature, healthy tissues and organs secondary to radia...
