Problems solved by technology

[0003] At present, attenuated vaccines and inactivated vaccines are mainly used to prevent IBD. With the emergence of IBDV mutant strains and super-virulent strains, immune failure often occurs.
Although the VP2 protein expressed by recombinant subunit vaccines and live vector vaccines can induce neutralizing antibodies to varying degrees, it still fails to solve the problem of IBDV antigenic variation

Abstract

The invention discloses a DNA (deoxyribonucleic acid) vaccine for expressing infectious bursal disease virus polyprotein gene VP243, as well as a construction method and application thereof. A polyprotein gene (VP243) of an infectious bursal disease virus virulent strain (HLJ0504) is optimized by chicken bias codons, and cloned to a eukaryotic expression vector pCAGGS so as to construct a recombinant expression vector pCAGoptiVP243. An initially immunized 14-day SPF chick is injected with 100mu g of the prepared pCAGoptiVP243 though two points of crureus in, a 28-day chick is subjected to the secondary immunization in same dosage and way, and IBDV (infectious bursal disease virus) virulent strain (HLJ0504) strain is attacked in 14 days after secondary immunization. Results indicate that the DNA vaccine can be used for inducing antibody with higher level and providing immunized protection to the virulent strain.

Description

technical field [0001] The invention relates to a DNA vaccine and its construction method and application, in particular to a DNA vaccine expressing infectious bursal disease virus polyprotein gene VP243 and its construction method and application. The invention belongs to the field of biomedicine. Background technique [0002] Infectious Bursal Disease Virus (IBDV) mainly infects central immune organs such as chicks' bursal disease, leading to immunosuppressive diseases characterized by lymphocyte exhaustion. IBDV is a double-stranded, double-segment RNA virus that belongs to the genus Avian BiRNAvirus of the family DiRNAviridae. Its genome consists of two segments, A and B, and the B segment (about 2.8 kb) encodes the VP1 protein, which is RNA-dependent. RNA polymerase. The A segment (about 3.3kb) has two partially overlapping open reading frames (ORFs). ORF1 encodes a precursor fusion protein (VP243) with a molecular weight of about 110kDa. The precursor fusion protein ...
