The method has the advantages that SXY36 is subjected to irradiation mutation breeding by ultraviolet rays, microwave and gamma rays, no hemolysis exists, the HA fermentation yield is high, and the heredity is stable, in the optimal fermentation condition, the fermentation yield of shaking flask is up to 0.731 g/L, and the yield of fermentation by a fermentation cylinder can reach 4.78 g/L; a two-stage variable temperature fermentation and stable pH method is adopted, high molecular-weight HA can be prepared by fermentation, and the molecular weight can reach 1.89 MDa, which is two times the molecular weight by a conventional fermentation method; HA with the molecular weight in the range of 0.035-1.89 MDa can be produced by using a fermentation broth self-enzymatic degradation method to prepare different molecular-weight HA and by controlling the time of enzymatic hydrolysis.
