Embodiment 1

[0023] (1) Using the upland cotton dwarf variety AS98 as the male parent and the normal tall upland cotton LHF10 as the female parent, crossed to obtain F 1 Generation, and then selfed to get F containing 225 individual plants 2 .

[0024] (2) Use CTAB method to extract parent, F 2 Genomic DNA of the young leaves of a single plant.

[0025] (3) Amplify the two parents AS98 and LHF10 with the designed 7 pairs of primers (Table 1). The amplification results show that the amplified products of 14k-2 and 14k-3 in the 7 pairs of primers have a single band in AS98. And there is no amplification product in LHF10 ( figure 2 ). Then use 14k-2 primer as molecular marker in F 2 Genotypic analysis in the isolated population showed that the phenotypic traits of the individual plants that were amplified were all dwarfed, and the phenotypic traits of the individual plants that failed to be amplified were all tall plants. 2 Only 9 of the 225 individual plants in the population do not conform to t...

Abstract

The invention belongs to the field of molecular biology and discloses a molecular marker closely linked to dwarfing traits of upland cotton AS98 and application. The molecular marker is located on a D12 chromosome, a specific primer pair using the molecular marker can amplify 120 bp. products, a forward primer sequence of the specific primer pair is shown as SEQ ID No.1, and a reverse primer sequence of the specific primer pair is shown as SEQ ID No.2. A pair of primers for identifying mutants and wild types of the upland cotton dwarfing material AS98 are found out. 90% or more of phenotypic variations can be explained in an F2 population constructed by the upland cotton AS98 and a wild type LHF10. The molecular marker can be used for molecular marker-assisted selection and map cloning ofupland cotton AS98 dwarfing varieties, and a certain theoretical basis is provided for revealing cotton dwarfing mechanism and cultivating new dwarfing varieties.

Description

Technical field [0001] The invention belongs to the field of molecular biology, and specifically relates to a molecular marker closely linked to the AS98 dwarf character of upland cotton. Background technique [0002] Plant dwarfing is an important agronomic trait. Dwarf varieties have the characteristics of strong lodging resistance, suitable for close planting and mechanized management, which are of great significance for reducing labor intensity, improving fertilizer utilization and increasing yield. Dwarfing mutants have important use value in dwarf breeding, and they are also Research the important genetic resources of plant stem growth and hormone regulation. [0003] Cotton has unlimited growth habit and the plant is tall. At present, most cotton-producing countries in the world widely use the chemical inhibitor methyramine to control the growth of cotton, so that the plant height is generally controlled at around 90-100cm. Especially since the success of the “dense, shor...
