Abstract

A method for regulating the release of exosomes and its medical application for regulating the release of exosomes are provided, and the release of exosomes is promoted by inhibiting the activity of the rapamycin target protein mTORC1. Inhibition of mTORC1 activation by rapamycin increases exosome release. Inhibition of mTORC1 activity by nutrient deprivation, growth factors and dietary restriction promotes exosome release. mTORC1 regulates exosome release through a Rab27A-dependent mechanism. mTOR is associated with Rab27A, and the interaction between Rab27A and mTOR is enhanced by rapamycin treatment. The release of exosomes, concomitant with autophagy, is influenced by changes in nutritional and growth factor conditions. The release of exosomes releases the cellular contents outside the cell by fusing with the plasma membrane, resulting in the loss of the cellular contents. The invention can regulate the release of exosomes, and provides a feasible solution for the medical use of regulating the release of exosomes.
