Problems solved by technology

Although these modeling methods can simulate the disease phenotype of epilepsy, the time of administration is after birth rather than during pregnancy, so it can only reflect some emotional abnormalities caused by the environment or drugs after birth, which is related to the pathogenesis of fetal-derived adult epilepsy. There are big differences in

Embodiment 1

[0030] [Example 1] Construction of an animal model of fetal-derived adult epilepsy

[0031] 1. Experimental animals

[0032]SPF grade normal healthy Wistar rats were purchased from the Hubei Provincial Center for Disease Control and Prevention, animal license number: SCXK (Hubei) 2012-2014. This study was approved by the Ethics Committee of Wuhan University Medical Center, and was carried out in strict accordance with the relevant treatment guidelines of international laboratory animal protection certification and evaluation institutions.

[0033] The experimental animals were kept in a barrier environment with a temperature of 22-25° C. and a humidity of 50%, with 12 hours of day and night alternation.

[0034] 2. Experimental method

[0035] 20 male Wistar rats (body weight 260-300g), and 40 female Wistar rats (body weight 200-240g). Free to drink and eat, after 7 days of adaptive feeding, according to male: female = 1:2 cage, vaginal smear in the next morning, to determi...

[0076] [Example 2] Application of the animal model of fetal-derived adult epilepsy of the present invention in the study of the occurrence mechanism, early warning and intervention target of fetal-derived adult epilepsy

[0077] 1. Establish a fetal-derived adult epilepsy model according to the method of Example 1 of the present invention;

[0078] 2. Detect the changes of relevant indicators of hippocampus in offspring in utero and after birth to determine the mechanism of fetal-derived adult epilepsy.

[0079] 3. Using the fetal-derived adult epilepsy model established by the present invention to determine the clinical early warning and intervention targets of epilepsy.

[0080] To sum up, in the present invention, through the treatment of dexamethasone during pregnancy, the F1 offspring were routinely fed to 12 weeks of age after birth, and epileptic seizures were induced by subcutaneous injection of lithium chloride-pilocarpine. In order to detect EEG and take video of ep...

Abstract

The present invention provides a method for constructing an animal model of fetal-derived adult epilepsy and its use. The animal model of fetal-derived adult epilepsy is that normally pregnant rodents are given 0.2 mg / kg of dioxin per day on the 9th to 20th day of pregnancy. Subcutaneous injection of dexamethasone, F1 offspring were routinely fed until 12 weeks after birth, and epileptic seizures were induced by intraperitoneal injection of lithium chloride-pilocarpine. Epilepsy-related indicators were detected to indicate epilepsy susceptibility, and behavior was further detected at 18 weeks of age Psychologically relevant indications suggested anxiety, depression and cognitive impairment, and the animal model of fetal-derived adult epilepsy was determined to be obtained. The model established by the invention is novel, reliable and convenient, and has great significance for studying the mechanism of fetal-derived adult epilepsy, determining clinical early warning and drug prevention and treatment targets.

Description

technical field [0001] The invention relates to the technical field of animal model construction, in particular to a method for constructing an animal model of fetal-derived adult epilepsy and its application. Background technique [0002] Epilepsy is a paroxysmal brain dysfunction caused by excessive discharge of neuronal cell groups in the brain, and is a chronic disease of the nervous system characterized by recurrent epileptiform seizures. Epidemiological surveys found that the incidence of epilepsy worldwide is about 0.5-1.5%, and the number of people with the disease is about 50 million. Infancy, adolescence and early adulthood are the peak periods of epilepsy. The incidence of epilepsy in my country is about 0.6%, especially in males aged 10-19. Olli Nevalainen found brain cancer (17%), other cancers (15%), ischemic heart disease (11%), cerebrovascular disease (10%) and suicide (1.6%) through an analysis of the causes of death in 3,558 patients with confirmed epileps...
