The method comprises the following stepsof designing the nucleic acid amplification-chip electrophoresis titration sensing, carrying out nucleic acid amplification to form double-stranded DNA, carrying out chip electrophoresis titration sensing on the double-stranded DNA, and enabling the double-stranded DNA to enter an electrophoresis channel for a reaction to form the moving reaction boundary; firstly determining interface migration distances of the double-stranded DNA after amplification of a series of nucleic acids with known concentrations, and establishing a standard curve of the interface migration distances and the nucleic acid concentrations; and then carrying out amplification-electrophoresis titration sensing on nucleic acid to be detected to obtain the interface migration distance of the nucleic acid to be detected,and obtaining the concentration of the nucleic acid to be detected according to the established standard curve.
