The preparation method comprises the following steps of: firstly, mixing one or more than one of livestock manure and crop straws, aerobically fermenting at high temperature, drying and granulating to prepare the organic fertilizer for standby use; and respectively pulverizing the standby organic fertilizer, the urea, the potassium dihydrogen phosphate and the filling material, adding a plant growth regulator, mechanically and uniformly mixing, and packaging to obtain the rice seedling fertilizer.
