Embodiment 1

[0028] Embodiment 1, the making of watermelon gray mold specimen

[0029] (1) Collection of watermelon gray mold

[0030] Firstly, the fruits of watermelon botrytis with typical symptoms were collected from the field. The characteristic symptoms of watermelon gray mold samples are: "V"-shaped lesions are formed, and there is a gray-black mold layer on the lesions. The watermelon should only be medium ripe and relatively hard. The collected watermelon botrytis cinerea fruits were placed in collection boxes to avoid damage and brought back to the experiment.

[0031] (2) Configuration of color retention solution

[0032] The formula of the green color-preserving solution: 90 mL of 50% ethanol, 5 mL of formalin, 2.5 mL of glycerin, 2.5 mL of glacial acetic acid, and 10 g of copper chloride to make a mixed solution.

[0033] The configuration of 50% ethanol: Use a 100 mL graduated cylinder to measure 90 mL of absolute ethanol, pour it into a 200 mL beaker, and at the same time,...

Abstract

The method relates to a method for making plant disease specimens. The method is suitable for making disease specimens of fruits, hard in pericarp, such as citrus fruits including citrus, lemon and kumquat and melon fruits including pumpkin, white gourd and Hami melon. The method includes keeping color of the fruits hard in pericarp and with typical disease symptoms; scraping out flesh and pulp inside the fruits; using wood bits to replace the pulp to fill the fruits to completely support the fruits open; using glue to adhere soil on pericarp which is cut off; disposing the fruits in a drying oven at 45-55 DEG C until the fruits are dried. The fruit-type disease dried specimens made by the method maintain original color and luster of the fruits and are typical in symptom, free of obvious change in disease symptom and visual and convenient to observe. In addition, consumption of several volatile agents is reduced, so that environment pollution and physical damage are reduced; development of experimental teaching is further facilitated, and teaching effect is improved. Consequently, the method is suitable for popularization and application.

Description

technical field [0001] The invention relates to a preparation method of plant disease specimens, in particular to a preparation method of fruit disease specimens including citrus and melons. Background technique [0002] Plant disease specimens play a pivotal role in plant disease identification and experimental teaching. Plant disease specimens are not only an essential tool for practical teaching of plant protection-related majors in agricultural colleges and universities, but also important reference materials for scientific research institutes to provide social services and agricultural technology promotion centers to guide farmers in agricultural production. At present, the common types of plant disease specimens mainly include wax leaf specimens, dipped specimens and glass slide specimens. [0003] Most of the roots, tubers, fruits and other tissues of the diseased plants are preserved in the dipping solution to make dipping specimens. The dipping specimens are bright...
